Dieter Schlüter, co-examiner Zürich 2008 ABSTRACT Carbohydrates play a pivotal role in a range of biological processes. This thesis will present the exploration of various new tools to investigate carbohydrate-protein interactions. The first chapter of this thesis will deal with attempts to synthesize an oligosaccharide directly onto a functionalized, gold chip surface. The aim was to use the carbohydrate functionalized surface as a microarray. For this purpose, a suitable set of building blocks were designed, synthesized and the glycosylations optimized in solution. The glycosylations were then attempted on a gold surface, functionalized with a self-assembled monolayer (SAM). Matrix assisted laser desorption time of flight (MALDI-ToF) mass spectrometry was used to analyze compounds present on the surface. Further analysis of samples were carried out using variable angle spectroscopic ellipsometry (VASE), contact angle and polarization modulation-infrared reflection-adsorption spectroscopy (PM-IRRAS). The second part of this thesis will focus on the development of bio-sensors. A set of fluorescent glycodendrimers were assembled by a one-pot approach and then tested with Concavalin A (ConA) for binding. The effect of ConA binding on the glycodendrimer fluorescence and turbidity was investigated, as well as the effect of increasing the density of carbohydrates on the exterior of the glycodendrimer. Further investigations were made into the development of a surface based approach for the detection of carbohydrates. Using ConA immobilized onto a surface and a Ru(II) complex adorned with monosacharides, a method of detecting several small sugars was developed. Following on from this, a protein binding detection system was developed, involving gold islands and thiol functionalized carbohydrates, which did not require the use of a Ru(II) complex. VII In the last chapter of this thesis, the detection of fluorous tagged glycans by MALDI-ToF mass spectrometry was investigated. N-Glycans obtained from glycoprotein asialo fetuin were labeled with a fluorous tag, purified by fluorous silica and high-pressure liquid chromatography (HPLC). The tagged glycans were analyzed by MALDI-ToF mass spectrometry and both the tri and biantennary glycans were detected. VIII RÉSUMÉ Les carbohydrates jouent un rôle capital dans un grand nombre important de processus biologique. Cette thèse a pour but d’explorer de nouvelles méthodes pour étudier les intéractions carbohydrates-protéines. Le premier chapitre de cette thèse traitera des tentatives de synthèse d’un oligosaccharide sur une surface d’or, fonctionalisée. Deux bloques de glycoside pour la glycosylation ont été synthétisés et les glycosylations optimisées en solution. Les glycosylations ont été tentées sur des surfaces d’or fonctionnalisées avec un SAM. L’utilisation des systèmes SAM binaires pour faciliter la glycosylation sur la surface a aussi été étudiée. L’analyse des produits présents sur la surface d’or a été réalisée principalement à l’aide de Matrix assisted laser desorption time of flight (MALDI-ToF), ainsi que l’ellipsométrie et PM-IRRAS. La deuxième partie de cette thèse se concentrera sur la conception de bio-capteurs: une série de glycodendrimères fluorescents ont été construits par auto-assemblage puis l’affinité de ces derniers glycodendrimères pour la Concavaline A (ConA) a ensuite été testée. L’effet de la reconnaissance de la protéine sur la fluorescence et la turbidité des glycodendrimères a été examiné, ainsi que l’effet de l’augmentation de la densité de carbohydrate sur la partie extérieure du glycodendrimère. Des études plus approfondies ont été menées sur le développement d’une approche qui nécessite une surface pour la détection de carbohydrates. Une méthode pour détecter plusieurs mono et disaccharides a été accomplie, en utilisant la ConA (immobilisée sur une surface) et un complexe à base de Ru(II) décoré de monosaccharide. Suite à ce projet, un système de détection de protéines sur une surface, basé sur des îles d’or a été conçu et réalisé. Cette méthode repose sur la fonctionnalisation des îles d’or par des carbohydrates portant un thiol et ne nécessite pas de complexes à base de Ru(II). IX Le dernier chapitre la détection par spectrométrie de masse MALDI-ToF de glycans marqués au fluor a été étudiée. Les N-glycans obtenus de la glycoprotéine asialo fetuin ont été étiquetés avec un marqueur fluoré, purifiés par silice fluorée et high-pressure liquid chromatography (HPLC). Ces derniers ont été analysés par spectrométrie de masse MALDIToF et aussi bien les tri- et biantennaires ont été détectés. X
