Ces infections sont à la fois un défi pour la compréhension des relations hôte hôte-pathogène pathogène et une priorité pour le développement de nouvelles thérapeutiques. Le programme de recherche actuel vise à explorer la dynamique de réplication virale, au cours de la primo-infection, infection, dans les phases de transition latence latence-réactivation réactivation et les situations d’intégration dans le génome cellulaire, au cours des infections herpétiques. Cela concerne en particulier les infections à herpèsvirus humain 6 (HHV (HHV-6) 6) et à virus herpes simplex (HSV). L’identification des facteurs de persistance concerne à la fois l’hôte (réponse immune et susceptibilité généti que en collaboration avec d’autres équipes du CIMI) et le virus (variabilité génétique, intégration chromosomique, expression différentielle de certains gènes critiques). La susceptibilité et la résistance de la réplication virale à des inhibiteurs spécifi spécifiques ques sont caractérisées dans des modèles cellulaires in vitro et chez les sujets traités in vivo, à la fois sur le plan phénotypique et sur le plan moléculaire, avec la combinaison d’études génétiques et de quantification de l’expression protéique. Les objectifs finaux sont le développement de la chimiothérapie antivirale, l’étude de ses interactions avec les effecteurs immunitaires, la caractérisation des mécanismes de résistance aux antiviraux et l’identification de marqueurs d’évolution de l’infection. Herpesvirus infections are relevant models of persistent viral infections, particularly in the context of coinfections with HIV and in immunocompromised patients. The infections are a challenge in the understanding of interactions between host and pathogens, and a priority for the development of novel therapies. Our current research program aims at exploring the dynamics of viral replication, during primary infection, in transition phases from latency to reactivation and in situations of chromosomal integration of viral genome. Our main targets are human herpesvirus 6 (HHV-6) and herpes simplex virus (HSV). The identification of persistence factors concerns both host (analysis of immune response and genetic susceptibility to infection in collaboration with other CIMI teams) and virus (genetic variability, chromosomal integration, specific gene expression). The susceptibility and resistance of viral replication to specific inhibitors are investigated both in vitro in cell culture models and in vitro in treated patients, in terms of phenotypic characterization and molecular analyses combining genetic and gene expression studies. The final goals are the development of antiviral chemotherapy, study of its interactions with immune effectors, characterization of resistance mechanisms, and identification of evolution markers during infection.
