Allain, Angers) Un candidat est-il déjà pressenti : OUI/NON Is a candidate already foreseen : YES/NO 2 Projet de thèse en français et en anglais / Thesis project in French and English Titre du projet : Impact d’une hyper-activation développementale de l’amygdale sur le comportement de peur chez le rat adulte et ses corrélats neurophysiologiques Résumé du projet (15 lignes maximum) : La phase de développement périnatal est une phase particulièrement sensible au stress dont l’impact est visible jusqu’à l’âge adulte. L’amygdale, structure du système limbique, est impliquée dans l’apprentissage émotionnel et les réponses comportementales de peur et d’anxiété. Un dysfonctionnement de l’amygdale au cours du développement pourrait être à l’origine d’un développement modifié des connectivités neuronales intra-amygdaliennes et/ou entre l’amygdale et d’autres structures cérébrales, et d’une modification du comportement émotionnel chez l’adulte. Certains symptômes associés à l’autisme pourraient être la conséquence d’une hyper-activation de l’amygdale montrant ainsi une plasticité singulière au cours du développement. Nous chercherons donc à tester l’impact de l’induction d’une hyperactivation amygdalienne chez le raton (par injection de lentivirus qui bloque la voie de régulation de mTOR) sur les comportements d’anxiété et de peur conditionnée chez le rat jeune et adulte, ainsi que sur les connectivités fonctionnelles entre l’amygdale et d’autres structures cérébrales (par exemple, le cortex préfrontal) pendant le conditionnement de peur. Cela nous permettra de déterminer de manière causale le rôle de l’amygdale au sein de son réseau cérébral et l’impact de de son fonctionnement à différents stades de développement (juvénile, adolescent et adulte). Title of the project: Impact of a developmental hyper-activation of the amygdala on fear behavior in adult rats and its neurophysiological correlates Summary of the project (15 lines maximum): The perinatal development is a phase particularly sensitive to stress whose impact is visible until the adulthood. The amygdala, structure being part of the limbic system, is involved in fear memory and anxiety. A dysfunction of the amygdala during development can result in a modification of intra-amygdala neuronal connectivity and/or between the amygdala and other brain structures, and a modification of the emotional behavior in the adult. The current hypotheses suggest that some symptoms seen in Autism could be the consequence of an overactivation of the amygdala resulting in a modified plasticity during development. In the present project, we will test the impact of the induction of an over-activation of amygdala in a juvenile rat (by injection of a lentivirus which blocks the tract of regulation of mTOR) on anxiety behaviors and fear conditioning in adolescents and young adults as well as the functional connectivity between the amygdala and other brain structures (for example, the prefrontal cortex) during fear conditioning. This will allow us to decipher, in a causal relationship, the role of the amygdala within its cerebral network and the impact of its functioning at different stages of development (juvenile, adolescent and adult).
