Le tubercule olfactif, partie du cortex olfactif primaire et du striatum ventral, pourrait être le lieu où ces odeurs prennent une signification positive ou négative. La signification pourrait naître de la reconnaissance d'un nombre limité de molécules odorantes présentes dans l'odeur naturelle. Les objectifs de la thèse sont (i) de comparer l’effet des odeurs naturelles à celui de molécules odorantes qui en sont extraites sur le comportement de rats et l'activité des neurones du tubercule olfactif, (ii) de rechercher un rôle modulateur des systèmes dopaminergiques et cholinergiques sur les réponses des rats et des neurones, enfin (iii) de construire et comparer des cartes statistiques de densité des neurones activés par les odeurs naturelles et les molécules odorantes candidates. Les méthodes mises en œuvre sont: l'électrophysiologie extracellulaire chez le rat anesthésié, l'immunodétection de la protéine Fos prise comme marqueur d'activation neuronale, la pharmacologie in vivo, les observations comportementales, la reconstruction 3D de modèles anatomiques. Title of the project: Odor coding by the olfactory tubercle in rats Summary of the project (15 lines maximum): Natural odors released by food, sexual partners or predators elicit specific behaviors in rats. The olfactory tubercle, part of the olfactory cortex and ventral striatum, may be a brain area where these odors acquire a positive or negative valence. Valence could originate from the analysis of a limited number of odorant molecules present in the natural odors. Aims of the thesis are: (i) to compare the effect of natural odors to that of odorant molecules on the behavior and olfactory tubercle neurons activity in rats, (ii) to investigate the modulatory role of dopaminergic and cholinergic pathways on neurons responses, and (iii) to build and to compare 3D statistical density maps of neuronal populations activated by natural odors and odorant molecules. Methods include behavioral observations, extracellular electrophysiology in anaesthetized rats, in vivo pharmacology, immunohistochemistry of Fos protein, a marker of neuronal activation, and 3D reconstruction of anatomical models.
