ZUSAMM ENFASSUNG V Zusammenfassung Pflanzen können als vielversprechende Expressionssysteme zur Produktion von rekombinanten Proteinen für Human- und Veterinärmedizin verwendet werden. Eine Vielzahl von Antikörpern und Antikörperderivaten wurde seit den ersten Berichten von pflanzlich hergestellten Immunglobulinen in verschiedenen Pflanzenarten produziert. Die Antikörperketten von zwei HIV neutralisierenden Immunglobulinen (2F5 und 2G12) wurden mit elastinähnlichen Polypeptiden (ELP) fusioniert, um eine Akkumulationssteigerung der rekombinanten Proteine in transgenen Pflanzen zu erzielen. Ausgewählte Pflanzen mit Expression der Immunglobulingene wurden anschließend gekreuzt. Daraus resultierten vier verschiedene Antikörpervarianten: O hne ELP, mit ELP an leichten oder schweren Ketten bzw. mit ELP an allen vier Antikörperketten (ubiquitäre Expression). Bei Pflanzen mit samenspezifischer Akkumulation der rekombinanten Proteine wurden Kreuzungen zur Erzeugung der Antikörper ohne und mit ELP an allen vier Ketten durchgeführt. Die ELP-basierte Expressionssteigerung konnte für die einzelnen Antikörperketten und auch die vollständigen Immunglobuline bestätigt werden. Stabile, homozygote Tabaklinien mit Akkumulation der kompletten Antikörper wurden durch die Nutzung der Doppelhaploiden-Technik erzeugt. Ausgehend von einer embryogenen Pollenkultur wurden haploide Pflanzen regeneriert, deren Genomaufdopplung zu Doppelhaploiden (DH) führte. Doppelhaploide Pflanzen mit Immunglobulingenexpression wurden identifiziert und deren Reinerbigkeit durch die Analyse der Nachkommen bestätigt. Ein Gendosiseffekt konnte ebenfalls nachgewiesen werden. Affinitätsgereinigte Antikörper wurden hinsichtlich ihres Bindeverhaltens zum jeweiligen Antigen mit Oberflächen-Plasmon-Resonanz näher charakterisiert. Die kinetischen Parameter der Antigenbindung waren nahezu identisch zu den Werten der aus CHO-Zellen stammenden Standardantikörper. Am Fc-Teil aller rekombinanten Immunglobuline wurden überwiegend N-Glycane vom oligomannosidischen Typ identifiziert. Ein negativer Einfluss von ELP auf diese posttranslationale Modifizierung und Proteinqualität konnte nicht gefunden werden. Die Faltung, Zusammenlagerung und der Transport aller Antikörpervarianten wurden durch ELP nicht behindert. In Pflanzen produzierte 2G12 Antikörper und Antikörper-ELP-Fusionen neutralisierten HI-Viren in vitro. Experimente zur ELP-basierten Reinigung der rekombinanten Proteine führten zu einer starken Anreicherung der Antikörperfusionen aus Pflanzenextrakten. 114 ZUSAMM ENFASSUNG Summary Plants are promising vehicles for the expression of recombinant proteins with regard to medical and veterinary purposes. Since the first reports of recombinant antibodies derived from transgenic plants, various antibody derivatives have been produced in different crop species using several expression systems. In order to enhance the accumulation of two HIV-1 neutralizing antibodies (2F5 and 2G12) in tobacco leaves and seeds, synthetic repeats of elastin- like polypeptides (ELPs) were C-terminally fused to both antibody chains. The individual chains were expressed both with and without C-terminal ELP fusion. Crossing these plants in all combinations resulted in transgenic lines producing the full antibod ies in four formats in tobacco leaves, with ELP on either the light or heavy chains, on both or on neither. For seed-specific expression, plants without and with ELP on all four antibody chains were produced. An increase of the expression based on ELP fusion was confirmed for the individuell antibody chains and for the full- length antibodies. Homozygous lines offer a great potential for the large scale production of antibodies. Such lines can be created by self-pollination of F1 plants and analysis of the next two generations. Alternatively, haploid plants can be produced by embryogenic pollen culture and the genome of these haploids can be doubled. The genome was doubled spontaneously or after colchicine treatment leading to doubled haploids (DH). Homozygosis of the DH plants was confirmed by the examination of the gene dosage effect and by testing the sexual progeny. Characterization of the affinity-purified antibodies from tobacco leaves by surface plasmon resonance spectroscopy showed that the kinetic binding parameters were identical to those of the CHO cell counterparts lacking ELP. N-Glycan analysis of these preparations revealed that all four derivatives of 2F5 and 2G12 contained predominantly oligo-mannose type N-glycans. ELP fusion has no significant effect on N-glycan structure and no adverse effect on protein quality. ELP fusions do not interfere with folding, assembly, trafficking in the secretory pathway, or post-translational modification. In vitro neutralization assays demonstrated that the HIV-1 inactivation capacity of the tobacco produced 2G12 was equivalent or even better compared to the CHO standard. ELP fusion to the heavy chains of 2G12 resulted in HIV neutralization with minor differences relative to the standard. First experiments on the enrichment of recombinant antibody ELP fusions in tobacco leaf extracts were performed and indicated a simplified recovery of the recombinant proteins. 115
