INFLUENZA ANTIVIRALS NOVEL TARGET FOR THE TREATMENT OF INFLUENZA VIRUS BENEFITS PROTECTION AGAINST INFLUENZA VIRUS NO RESISTANCE DEVELOPMENT BROAD SPECTRUM ACTIVITY EFFECTIVE EVEN BEYOND 48 HOURS AFTER THE INFECTION KEYWORDS INFLUENZA FLU INFECTION ANTIVIRALS FORMYL PEPTIDE RECEPTOR 2 VIRAL REPLICATION LUNG INFLAMMATION BACKGROUND Despite vaccination, 5 million people are affected each year by the influenza virus in the world, causing more than 500,000 deaths annually. Current antivirals (neuraminidase, hemagglutinin and M2 protein inhibitors) are no longer effective 48 hours after infection and lead to the emergence of multi-drug resistance strains. Need for new antivirals with broad spectrum activity remains high to prevent epidemics and pandemics. HOW IT WORKS FPR2 (Formyl Peptide Receptor 2) plays a key role through the ERK / MAPK pathway in the inflammatory resolution following an acute inflammation but also in the replicative capacity of the influenza virus. Researchers show that an antagonist of FPR2 protects mice against influenza infection by blocking viral replication, as well as significantly reducing pulmonary inflammation. LIMITE L'ÉMERGENCE DE RÉSISTANCES Les antiviraux actuels (inhibiteurs de neuraminidase, hémagglutinine et protéine M2) ne sont plus efficaces 48 heures après l'infection et induisent l'apparition de nouvelles souches multi-résistantes. La demande de nouveaux antiviraux à large spectre d'activité reste très élevée pour prévenir les épidémies et pandémies. LARGE SPECTRE EFFICACE PLUS DE 48H APRÈS L'INFECTION MOTS-CLÉS INFLUENZA INFECTIONS GRIPPALES ANTIVIRAUX FORMYL PEPTIDE RÉCEPTEUR 2 RÉPLICATION VIRALE INFLAMMATION PULMONAIRE PROPRIÉTÉ INTELLECTUELLE BREVET TECHNOLOGIE Le récepteur FPR2 (Formyl Peptide Récepteur 2) joue un rôle central via la voie ERK/MAPK dans la résolution inflammatoire suite à une inflammation aigue mais aussi dans la capacité réplicative du virus influenza. Un antagoniste de FPR2 protège les souris contre les infections grippales en bloquant la réplication virale ainsi qu'en réduisant fortement l'inflammation pulmonaire. AVANTAGES • FPR2 est une nouvelle cible impliquée dans l'infection par le virus influenza • Inhiber FPR2 est une nouvelle stratégie thérapeutique contre la grippe.
