Allain (Angers) Charlotte Lamirault Noms et Prénoms des allocataires (MESR, Région, Paris-Sud…) de l’équipe durant les deux dernières années / PhD students of the team with a scholarship during the past two years Charlotte Lamirault Un candidat est-il déjà pressenti : OUI Is a candidate already foreseen : YES/NO 2 Projet de thèse en français et en anglais / Thesis project in French and English Title of the project: Silencing mutant huntingtin for reverting emotional, cognitive and synaptic anomalies in the BACHD rat model for Huntington Disease Summary of the project (15 lines maximum): The project will aim at characterizing the neuronal alterations of the prefronto-amygdalo-striatal network of BACHD rats, a transgenic rodent model of Huntington disease (HD), underlying emotional/executive phenotype and at determining whether normalization of this network function by mhtt reverting in early life can ameliorate adult emotional and cognitive deficits. First, we will study in young (presymptomatic, 1 month) and adult rats (early symptomatic, 6 months) different executive functions and their modulation by induced positive or negative emotional states. Second, at these same ages (1 and 6 months), neuronal functioning in targeted structures of the network will be tested using cellular imaging (early genes expression, mhtt aggregates) and in vivo electrophysiology (plasticity of prefronto-striatal pathway and its modulation by amygdala and plasticity of prefronto-amygdala pathway). Third, the importance of amygdala function will further be addressed by testing whether the emotional/motivational deficits observed in HD adult rats can be restored by acting on the amygdala or the prefrontal cortex at the presymptomatic age of 1 month, i.e. by normalizing plasticity through mutant huntingtin (mhtt) silencing (by lentivirus injections) in our newly developed rat model of HD (BACHD).
