Lincomycin and clindamycin are antibiotics used for the treatment of infections caused by Gram-positive bacteria. The cavity around C(1) of clindamycin seen in the crystal structure of its complex with the 50S ribosomal subunit of Deinococcus radiodurans suggests that substitutions at C(1) and the introduction of groups larger than the C(1)–SMe group may add favourable interactions with the nucleobases of the bacterial ribosome. To obtain such analogues, I intended to develop the chemistry of nitroethers, to elaborate a method to prepare 1-nitropiperidinoses, and to apply the results to the synthesis of C(1) mono- or di-C-substituted pyranose and piperidinose analogues of lincomycin. S-Aryl and S-arylalkyl analogues of lincomycin were obtained by S-glycosylation of the lincosamine-derived sulfoxides with arylthiols, or by S-alkylation of the glycosyl thiol with alkyl bromides. A protected 1-C-nitro-1-deoxy lincosamine was synthesized, and C(1)– alkylated lincomycin derivatives (ulosides) were obtained by Henry reaction or Michael addition of these 1-deoxy-1-nitropyranoses, followed by solvolytic substitution of the tertiary nitro group by SMe. These lincosamine derivatives were coupled to propylhygric acid, and deprotected to afford C(1)-substituted lincomycins. These lincomycin analogues (as well as the other analogues described below) were tested against both lincomycin-sensitive and lincomycin-resistant Mycobacterium smegmatis strains, relying on the genetic model developed by Böttger et al. which allows to assess the activity and the specificity (and thus the toxicity) of any new antibiotics. The S-alkyl derivatives were found to be as active and selective as lincomycin. The lipophilic uloside analogues of lincomycin were 2 to 4 times less active than lincomycin, whilst the hydrophilic analogues turned out to be inactive. To establish a method to obtain 1-deoxy-1-nitro-piperidinoses and to use them for the synthesis of C(1)-substituted, chain-elongated piperidinose analogues of lincomycin, I first developed a route to glucose-derived 1-deoxy-1-nitropiperidinoses and then applied the method to the synthesis of a 1-nitropiperidinose derived from lincosamine. To disfavour elimination of the anomeric nitro group in 2-nitropiperidine, both N-(tertbutyloxy)carbonylated1 and N-tosylated piperidinose-derivatives were synthesized. After several attempts to prepare the desired compounds via the lactam-oxime 217, or the N– bocylated amino-oximes 223, or by oxidation of anomeric azides 228 with HOF (hypofluorus acid) or of anomeric phosphimines 230 by ozone, I obtained the 1-nitropiperidinoses 220, 237 and 245 by ozonolysis of anomeric piperidinose-derived nitrones, as evidenced by IR and NMR spectroscopy. The nitrones were obtained by oxidising the cyclic N- benzylhydroxylamines that were formed upon treatment of protected N,O-hemiacetals with Nbenzylhydroxylamine in 58 to 78% yield from the corresponding N,O-hemiacetals. However, cleavage of the anomeric C,N-bond of the N-bocylated 1-nitropiperidinose 237 occurred readily upon warming to ambient temperature and/or upon removing the solvent. The similarly synthesized N-tosylated 1-nitropiperidinose 245 was stable up to temperatures of ca. –10°; above this temperature, the anomeric nitro group was replaced by a hydroxy group. All attempts to study the reactivity of the highly reactive, protected nitropiperidinoses towards aldehydes and Michael acceptors failed to lead to the expected products. Finally, the amide moiety of lincomycin was replaced by a 1,2,3-triazole ring, since 1,2,3triazoles are known peptide analogues resistant to hydrolytic cleavage and oxidative transformations. The 1,2,3-triazoles were readily prepared by [3+2] cycloaddition of the C(6)–azide derived from lincosamine and the C(2)–alkyne derived from propylhygric acid. A small library of 1,2,3-triazole analogues of lincomycin was synthesized, substituting the proline ring by acyclic substituents, cycloalkyl groups, or by an aromatic ring. These analogues were inactive towards wild type ribosomes and mutants. 1 For simplification, N-(tert-butyloxy)carbonyl will be replaced by N-bocyl Résumé Cette thèse a pour but le développement de nouvelles méthodes associées aux dérivés anomères C-nitro et de les appliquer à la synthèse d’analogues innovants de la lincomycin. La lincomycin et la clindamycin sont des antibiotiques utilisés pour le traitement des infections causées par les bactéries ‘Gram-positif’. La structure cristallographique de la clindamycin complexée à la sous-unité 50S du ribosome du Deinococcus radiodurans a permis de mettre en évidence une cavité autour de C(1). Cet espace permettrait d’introduire des substituants en position anomère de la lincomycin ou de remplacer le groupe C(1)–SMe par un groupe plus large, pouvant ainsi former de nouvelles interactions avec l’ARN ribosomal. Les analogues envisagés ont été obtenus par l’utilisation de C(1)–nitroéthers, par l’élaboration d’une méthode de synthèse de C(1)-nitropipéridinoses et par l’application de ces méthodes à la synthèse de dérivés pyranoses et pipéridinoses de la lincomycin, mono- ou di-C-substitués en position C(1). Les dérivés S-alkylés et S-arylalkylés ont été obtenus par S-glycosylation de sulfoxides dérivés de la lincomycin avec des arylthiols ou par S-alkylation du glycosyl thiol avec des bromures d’alkyles. Les dérivés de la lincomycin substitués en position C(1) (ulosides) ont été préparés à partir de 1-deoxy-1-nitropyranoses dérivés de la lincomycin, par réaction de Henry ou par addition de Michael. Ces analogues de la lincomycin (et les analogues décrits ci-dessous) ont été testés sur des souches de Mycobacterium smegmatis sensibles et résistantes à la lincomycin, ces tests s’appuyant sur le modèle génétique développé par Böttger et al., qui permet d’établir l’activité et la spécificité (et donc la toxicité) des nouveaux antibiotiques. Les dérivés S-alkylés de la lincomycin ont montré une activité et une sélectivité semblables à celle de la lincomycin. Les ulosides lipophiliques étaient 2 à 4 fois moins actifs que la lincomycin, alors que les ulosides hydrophiliques étaient inactifs. Pour établir une méthode de synthèse des 1-déoxy-1-nitropipéridinoses, utilisés pour la synthèse de pipéridinoses à chaîne allongée, analogues de la lincomycin, substitués en C(1), j’ai tout d’abord établi une voie permettant la synthèse de 1-nitropipéridinoses sur les derivés du glucose et adapté celle-ci à la synthèse de 1-nitropipéridinoses, derivées de la lincosamine. Pour défavoriser l’élimination du group nitro en position anomère, les N-bocylés et N-tosylés 1-nitropipéridinoses ont été préparées. Après plusieurs essais de préparation de ces intermédiaires via la lactame-oxime 217, ou les amino-oximes N-bocylées 223, ou par oxydation d’azides anomères 228 avec HOF (acide hypofluoreux), ou de phosphimines anomères 230 par l’ozone, les 1-nitropipéridinoses 220, 237 and 245 ont été obtenues par ozonolyse de pipéridinosyl-nitrones, caractérisées par spectroscopie IR et RMN. Ces nitrones ont été synthétisées par oxydation de N-benzylhydroxylamines cycliques, préparées par traitement des N,O hémiacétales correspondantes avec N-benzylhydroxylamine, avec un rendement de 58 à 78% à partir des N,O hémiacétales respectives. Cependant, la coupure de la liaison anomère C,N de la nitropipéridinose N-bocylées 237 se produit lorsque le milieu réactionnel est ramené à température ambiante et/ou lorsque le solvant est évaporé. La 1nitropipéridinose N-tosylée 245 était stable jusqu’à –10°C ; au-dessus de cette température, le groupe nitro anomère a été remplacé par un groupe hydroxy. Tous les essais effectués afin d’étudier la réactivité de ces 1-nitropipéridinoses sur des aldéhydes ou des accepteurs de Michael ont échoué. Enfin, la fonction amide de la lincomycin a été remplacée par un cycle 1,2,3-triazole. Les 1,2,3-triazoles sont connus pour être des analogues de peptides résistants aux coupures hydrolytiques et aux transformations oxydantes. Les analogues 1,2,3-triazoles ont été préparés par des cycloadditions [3+2] du C(6)-azide dérivé de la lincosamine et du C(2)-alcyne dérivé de l’acide propylhygrique. Une librairie d’analogues 1,2,3-triazoles de la lincomycin a été préparée, la proline ayant été remplacée par des substituants acycliques, par des groupes cycloalkylés ou par des cycles aromatiques. Ces nouveaux analogues de la lincomycin se sont avérés inactifs.
