Lors de ma première mobilité de Juillet à Octobre 2012 à l’IIT (Italian Institute of Technology à Gênes-Italie) au département de Neuroscience and Brain Technology, j’ai réalisé des études sur les maladies neurodégénératives en étudiant l’impact de nanovecteurs sous forme de nanoliposomes riches en acides gras polyinsaturés que j’ai élaborés en France. Cette mobilité a donné lieu à quatre conférences internationales, un brevet en cours de dépôt et une publication soumise à Scientific Reports-Nature. En 2013 (Mars-Septembre) : j’ai obtenu une CRCT (Congé pour Recherches ou Conversions Thématiques) de 6 mois de l’Université de Lorraine qui m’a permis de réaliser une deuxième mobilité au sein d’un laboratoire, dépendant de Harvard (département Harvard Medical School) (rang mondial dans le classement de Shanghai : 1) et du MIT (Massachusetts Institute of Technology) (rang mondial dans le classement de Shanghai : 4), qui est l’un des meilleurs laboratoires de recherche sur le développement de nouveaux matériaux utilisés dans les secteurs de la cosmétique et du biomédical. De plus, ce laboratoire est également attaché à Brigham Women’s Hospital (BWH) qui est l’un des meilleurs hôpitaux au niveau mondial. Au cours de cette expérience, j’ai fonctionnalisé des hydrogels à l’aide de nanoliposomes à base de lécithine de saumon afin d’améliorer leurs propriétés fonctionnelles et mécaniques ainsi que leur activité biologique. Cette mobilité m’a permis de combiner mes connaissances en nanotechnologie avec leurs activités en développement de biomatériaux pour une application cosmétique. L’objectif de cette mobilité a été de rapporter de nouvelles compétences au sein du Laboratoire d’Ingénierie des Biomolécules tels que la biologie, le développement de systèmes 3D par microfluidique et la fabrication de nouveaux matériaux biodégradables. De plus, plusieurs collaborations ont été établies pendant mon séjour avec entre-autre l’Université de Sydney (Australie) (une publication acceptée n°3); l’Université de McGill (Canada), Université de Gênes (Italie) (une publication acceptée n°6), l’Université de Wuhan (Chine), l’Université de Caroline du Sud (USA) (une publication acceptée n°3 et une publication soumise), 3 l’Université de Tufts (USA), et l’Université de Stanford (USA). Ces collaborations ont abouti au dépôt et à l’obtention de projets de recherche. Cette mobilité a donné lieu à 1 conférence internationale, 4 posters, une publication acceptée à Advanced Healthcare Materials et une publication soumise à Acta Biomaterialia. Brevet en cours. 15 Projet de recherche Le thème de recherche, dans lequel je me suis investie, fait intervenir des connaissances relevant d’une part, de la physico-chimie des interactions moléculaires dans les systèmes lipidiques et, d’autre part, des transferts de molécules bioactives à travers les bicouches lipidiques d’une membrane. Ces recherches trouvent leur application à l’étude de l’encapsulation de principes actifs (hydrophiles et hydrophobes) dans des liposomes. Je m’intéresse à la cinétique de ces transferts à l’intérieur des liposomes mais également à l’extérieur en vue d’une utilisation en agro-alimentaire, nutraceutique, pharmacopée et/ou dans le domaine biomédical. Le schéma ci-dessous présente de façon synthétique le résumé de mon activité de recherche en cinq parties : 1. Structuration et formulation des nanovecteurs lipidiques (nanoliposome, nanoémulsion) ; 2. Encapsulation de la molécule active (avec différentes propriétés physico-chimiques) ; 3. Caractérisation multi-échelle afin de comprendre le comportement des nanovecteurs avant et après l’encapsulation ainsi que les interactions entre le vecteur et son principe actif ; 4. Etude du transfert moléculaire, à différentes échelles, de la molécule active au sein du vecteur et du vecteur vers les systèmes simulants ou biologiques ; 5.
