— 149 NOTE RESISTANCE OF CHINOOK SALMON (Oncorhynchus tschawytscha) FINGERLINGS EXPERIMENTALLY INFECTED WITH VIRAL HEMORRHAGIC SEPTICEMIA VIRUS Wilma O R D Institut National de Laboratoire 78850 — la Recherche Agronomique d'Ichtyopathologie THIVERVAL-GRIGNON Viral Hemorrhagic Septicemia (V.H.S.) is an infectious and contagious disease of rainbow trout (SaImo gairdneri) w h i c h produces muscular hemorrhagic lésions and necrosis of the hematopoetic S y s t e m in infected fish. A t least 40 % of French c o m m e r c i a l fish f a r m s are affected by V.H.S. giving rise to very considérable annual économie losses. Previous w o r k has shown that brown trout {SaImo trutta) and C o h o salmon (Oncorhynchus kisutch) are résistant to V.H.S. (de K I N K E L I N et al. 1974). To further our understanding of the susceptible host range of this disease the effect of e x p o s u r e t o V.H..S has been studied in c h i n o o k s a i m o n c o m p a r e d w i t h the susceptible species, the rainbow trout. Rainbow trout and chinook salmon fingerlings (mean length 6 cm) were divided into control and infected groups. The infected fish (10 rainbow trout, 7 chinook salmon) w e r e injected intraperitoneally w i t h 0.2 ml STOKER médium containing 10'' plaque f o r m i n g units ( p . f u . ) of t h e virus of V . H . S , strain 07/71, serotype I (VESTERGARD-JÛRGENSEN 1972). The controls (8 rainbow trout, 6 chinook salmon) w e r e injected with the same volume of virus free STOKER médium. Prior to injection, ail fish were anaesthetized in a solution of 100 ppm tricaine metanesulphate (MS-222). To evaluate the course of the disease, t w o cidence of mortality and a quantitative analysis of the présence of virus. One fish from each infected 2, 4 and 15 post injection. Thèse viral extracts were then inoculated into Fathead M i n n o w ( G R A V E L L and MALSBERGER 1965) cell cultures according t o previousiy described methods (de KINKELIN et SCHERRER 1970). A t the conclusion of the experiment, ail fish w e r e sacrificed and analysed for V . H . S . virus. The w a t e r température of the f i s h tanks was maintained at 13 ± loC. Results of viral analyses and mortalities in thèse t w o species following e x p o s u r e to V.H.S. are p r e s e n t e d in table 1. N o mortality o c c u r r e d and no virus was isolated in chinook salmon. In c o m p a r i s o n , rainbow trout displayed heavy mortalities and virus was detected on days 2, 4, 5 and 8 in both sacrificed and dead trout. Highest viral yields occurred 4 to 5 days post infection. Viral analyses on fish sacrificed at the conclusion of the experiment w e r e all négative. Thèse présent findings suggest that chinook salmon fingerlings are ref r a c t o r y to V.H.S. This may be a common feature in the Oncorhynchus genus as t h e C o h o salmon has aiso been s h o w n t o be résistant. H o w e v e r as susceptibility to virus could be âge dépendant, caution must be taken in extrapolating thèse results to c o v e r other âge groups of chinook salmon. Nethertheless p r i e r t o undertaking this experiment, t h e behaviour of chinook salmon of up to 15 cm length exposed t o a naturel infection of V.H.S. was observed in the s a m e fish farm w h i c h had been used to check C o h o salmon susceptibility (de K I N K E L I N et al. 1974). No viral associated losses w e r e recorded in ponds or cages placed d o w n s t r e a m from infected rainbow trout ponds. SUMMARY This w o r k compares the susceptibility of chinook salmon (Oncorhynchus tschawytscha) and rainbow trout (SaImo gairdneri) fingerlings injected with the virus of Viral Hemorrhagic Septicemia. In chinook salmon, no deaths occurred and no virus could be isolated. In contrast, high viral titres w e r e found in rainbow trout amongst w h i c h there was a heavy mortality. Le c o m p o r t e m e n t du saumon chinook {Oncorhynchus tschawytscha) a été examiné et cet animal s'est révélé réfractaire à l'infection expérimentale réalisée par l'injection intrapéritonéale de 10* unités formant plage (u.f.p.) par animal d'un poids moyen de 2 g. A u c u n e mortalité n'est survenue chez les saumons et le virus n'a pas été isolé chez les animaux sacrifiés en fin d'expérience (t. I). Inversement les truites arc-en-ciel servant de témoins sensibles ont présenté une mortalité associée à l'isolement v i r a l . Le petit nombre des animaux d'expérience s'explique par le fait qu'un faible effectif v e n a i t d'être introduit en France, mais l'observation de ces animaux placés dans des conditions naturelles d ' i n f e c t i o n , c o m m e au cours des essais antérieurs pratiqués avec le C o h o (de K I N K E L I N , LE BERRE et MEURILL O N 1974), a c o n d u i t au même résultat que l'inoculation d u v i r u s . Le saumon C h i n o o k peut d o n c être considéré comme réfractaire à la S.H.V.
