Financement PARI Conseil Régional de Bourgogne Job description Descriptif de recherche Molecular imaging is used to study diseases in cancerology, cardiology and neurology, to develop new treatments, to perform clinical diagnostics and monitor treatments. The capacity to image and monitor biological events in vivo and in real time is of great interest to disclose a detailed picture of fundamental biochemical and physiological processes in living organisms. Suitable radioisotopes such as 18F have a very short half-life time: 109.8 minutes. The development of molecular probes should integrate strong expertise in chemistry. One of the main challenges of chemists is thus the development of rapid synthetic methods for introducing short-lived positron-emitting isotopes into molecules of interest. By an organometallic strategy our team recently completed the selective fluorination in ortho-position of aryltetrazine, difficult to achieve by conventional organic synthesis, in only few minutes paving an innovative path to the application to radioactive fluorination 18[F] (see Angew. Chem. Int. Ed. 2016, DOI: 10.1002/anie.201601082” and US patent 2015 US 62/260, 741). To extend this work two paths will be explored here: (1) the development of new ways of organometallic functionalization of bioconjugable aryltetrazine and (2) application of these methods for manufacturing bimodal probes. Thus, the implementation of its innovative synthesis tools makes conceptually possible transfer to the "hot" chemistry (radioisotope 18F) and may provide new tools and compounds for orthogonal bioconjugaison reactions in vivo. The profile of an expert in organometallic catalysis, organic chemistry and purification is needed for this task. The bioconjugaison and imaging probes implementation will be dealt in cooperation with Dr. R. Decréau as biochemist. Experience PhD or post-doctorate associate in molecular chemistry with experience in synthetic organic and/or organometallic chemistry and homogeneous catalysis. Experience in fluorine chemistry would be also appreciated. Organizing and communication skills, teamwork, and initiative would be important in order to interact with partners. French or foreign candidates are welcome but excellent level either in English or French is required. The date of PhD defense must be after 01/12/2012. Le rôle du chimiste est décisif pour incorporer aux molécules bioactives des isotopes radioactifs, contrôlables par imagerie, comme le fluor [18] à courte durée de vie (110 min). La fluoration par fonctionnalisation directe de liaison C–H permet à la fois de limiter les étapes de synthèse et la production de déchets afférents. Par une stratégie organométallique notre groupe a récemment accompli la fluoration sélective en position ortho d’aryles tétrazine très difficiles à obtenir par synthèse organique classique en seulement quelques minutes ouvrant ainsi une voie innovante vers l’application à la fluoration radioactive 18[F] en imagerie médicale (voir Angew. Chem. Int. Ed. 2016, DOI: 10.1002/anie.201601082”). Ce résultat sur une molécule bioconjugable a retenu l’attention de la SATT grand EST et la prise de brevet a été réalisée fin 2015 (Demande 2015 US 62/260, 741). L’objectif du projet qui prolonge ce travail se décline en deux axes : (1) le développement de nouvelles voies de fonctionnalisation organométallique d’aryltétrazines bioconjugables, et (2) l’application de ces méthodes à la fabrication de sondes bimodales. Ainsi, la mise en place de ses outils de synthèse innovants rend conceptuellement possible le transfert vers la chimie « chaude » (radioélément 18F) et la valorisation à terme vise ici à fournir de nouveaux outils et composés permettant des réactions de bioconjugaison orthogonale invivo. Le travail d’un chercheur expérimenté est nécessaire pour cette tâche, et son profil est celui d’un expert en catalyse organométallique, chimie organique et purification. L’approche de bioconjugaison et d’introduction de sondes d’imagerie sera traitée coopération avec le Dr. Richard Decréau biochimiste de formation. Profil du candidat Titulaire d’un doctorat en chimie moléculaire de synthèse. Autonomie, sens de l’organisation et de la communication, goût du travail en équipe, bonne maîtrise de l’anglais et/ou du français sont indispensables.
