Nous avons récemment démontré que la testostérone et son récepteur AR (androgen receptor) sont très importants pour la régénération de la myéline dans un modèle de démyélinisation chez la souris (Hussain et al., 2013). Le but de ce projet est d’analyser et de comparer les expressions des gènes cibles, en utilisant le modèle de démyélinisation focale de la moelle épinière par la lysolecithine (LPC), entre les souris intactes, les souris démyélinisés par la LPC traitées ou non par la testostérone. Les moelles épinières de ces souris seront coupées au cryostat et des sections de tissus contenant les lésions seront isolées et collectées par la technique de microdissection par laser. L'analyse transcriptomique par puce Affymetrix se fera à différents temps après l'injection de la LPC. Une fois les gènes importants sont identifiés, nous étudierons leur gain ou perte de fonction, par rapport à la prolifération et la différenciation des oligodendrocytes, par des expériences in vitro (culture organotypique et culture en monocouche) et des techniques de biologie cellulaire et moléculaire. Ainsi, la compréhension du mécanisme d’action résultant de l’interaction entre AR et ces gènes cibles permettrait d’améliorer la réparation de la myéline dans les maladies démyélinisantes comme la SEP. Title of the project: Promyelinating effect of testosterone and its receptor AR: transcriptomic study Summary of the project (15 lines maximum): The destruction of myelin observed in demyelinating diseases such as multiple sclerosis (MS) leads to inflammatory responses and neuronal degeneration, forming a major cause of handicap in young adult. Therefore, there is an urgent need for innovative therapies promoting neuroregeneration and myelin repair. We have recently demonstrated that testosterone and the androgen receptor (AR) obviously known for their reproductive functions, seem surprisingly very important for myelin regeneration in a rodent model of chronic demyelination (Hussain et al., 2013). An essential question will be addressed within the frame of the present project concerns the transcriptomic analysis between spinal cords of intact, lysolecithin (LPC) demyelinated and LPC plus testosterone treated mice. Briefly, after stereotaxic spinal cord lesions and treatment with or without testosterone, serial cryostat sections of the spinal cord will be collected and processed for laser capture microdissection. The transcriptomic analysis will be done at different time point after LPC injection, using an Affymetrix chip. Once important genes are identified, we will study through gain- and loss of function experiments, using in vitro (organotypic slice culture and monolayer cultures) and cell/molecular biology technics, their potentials to regulate oligodendrocyte proliferation and differentiation to myelinating oligodendrocytes.
