Packing Device In addition to the broad range KNAUER´s pre-packed analytical and preparative columns, we offer also the most flexible and economical column packing dvice. Especially if you need to purify large quantities of material routinely, self packing your preparative columns is a more economical alternative. To meet this need, KNAUER offers column packing units in combination with a broad range of bulk material. The column filling devices are available in scale from analytical (2 mm ID) columns up to preparative columns with diameters up to 100 mm ID.. Introduction For achieving the highest quality of HPLC columns special packing procedures have to be pursued. In the laboratory glass columns are routinely packed without additional pressure just by gravitation. A slurry of the adsorbent with a suitable organic liquid is prepared (a corresponding amount of adsorbent is weighed and a selected slurry liquid mixture is added) and homogenized in an ultrasonic bath for 3 min, poured into the glass tube. Then the bed is adjusted by slightly knocking at the outer wall of the column while the liquid passes through. However, this method is only suitable for particle sizes larger than 20 µm and glass columns. Analytical and preparative stainless steel columns with particle sizes smaller than 20 µm need more sophisticated packing procedures. Generally spoken, the higher the packing pressure the better the packing result will be. Two different methods are common using either a constant flow or a constant pressure. Constant Flow Method As well the KNAUER HPLC Smartline pump 1000 as the preparative Pump 1800 can be used for constant flow column packing. Smooth flow rates are typically applied since this method is especially suitable for pressure sensitive adsorbents such as some polymers. The flow rate is constant at all times. The pressure increases as the column bed is more and more compressed. After a defined time interval (after a defined amount of liquid passed through the column) the packing process is stopped and the pump shut off. The column will be disassembled and equipped with sieves, filters, sealing rings end fittings and screw caps. Constant Pressure Method The KNAUER pneumatic pump 1900 is the pump of choice for constant pressure column packing. It is powered by pressurized air and has been developed for constant pressure applications. The advantage of using this pump and the constant pressure method for column packing consist in the about two times higher pressure (up to 1000 bar) which is applicable. The pneumatic pump converts inlet air pressure to hydraulic fluid pressure against the slurry that is needed for properly packing of the chromatographic bed inside the column. This is the method of choice for pressure stable materials such as silica. The pressure is applied and packing liquid is pumped through the slurry tank-column unit. 4 Knauer High Pressure Column Packing Device The pressure is constant at all times. The flow rate decreases as the column bed becomes more and more compressed. A pressure of nearly 600 bar is applied for columns of 3 through 4.6 mm ID. The 250 ml pump head is recommended for these column dimensions. For packing of preparative columns (column ID > 16 mm) with particle size ≥ 10 µm larger pump head size (500 or 1000 ml) will be recommended. After a certain period the pump is shut off, the column will be disassembled from the filling unit and equipped with sieves, filters, sealing rings end fittings and screw caps. The pump 1900 may be placed in any position. Be sure the pump, the slurry reservoir, and the column can be shielded by a safety shield. With the 100 ml pump head the external air pressure must under no circumstances never exceed 4.2 bar. Exceeding an external air pressure of 4.2 bar will lead to damage of pump head and pump mechanics. This damage is not covered by guarantee. Pump 1900 Compressed Air Connections Make sure that your compressed air system to be used with the pneumatic Pump 1900 is able to maintain pressures up to 7 bar. Without back pressure the air consumption would be 80 l/min. If the pneumatic Pump 1900 works against pressure the consumption of air is much lower depending on the liquid pressure. The connector to the compressed air is located on the lower rear panel of the pump. Connect the coupler for the pressure tube to the coupler socket. Push the pressure tube (not delivered) from the compressed air supply over the coupler and secure it with an appropriate clamp. Connect the coupler socket to the compressed air connector of the pump (see Fig. 3). To disconnect from the compressed air supply: Hold the brass part of the coupler with one hand and pull the blue part of the coupler socket away from the pump with the other hand. Fig. 3 Air pressure connection of the pneumatic Pump 1900 The pump should be rinsed with a methanol-water mixture (1:9 v/v) before first usage. Make sure that all hydraulic connections are suitable for the system pressure and flow rate of your liquid transportation system. Air Pressure Influences The resulting eluent pressure increases or decreases as a function of compressed air inlet pressure. Eluent pressure will nearly be constant, if the compressed air inlet 6 Knauer High Pressure Column Packing Device pressure remains constant. The Regulator for compressed air can be locked by pressing it to wards the instrument. The delivery volume depends on the required air pressure and the loss of pressure along the column or any device that generates back pressure. You can reach the maximum delivery volume at minimum eluent pressure below 1 bar and the maximum eluent pressure at the minimum flow rate. Fig. 4 shows the delivery volume as a function of the eluent pressure as an example for the 250 ml pump head. In this figure you can see the limit values for the liquid pressure and the flow rate corresponding to Table “Technical data of individual pump heads” and the maximum allowed air pressure for the pump head. By the constant required air pressure the flow rate decreases if the liquid pressure increases. Corresponding figures for the other pump heads are shown in the pump’s manual. This will remove any particles of metal left inside the column as a result of cutting and deburring. When all the solvent has evaporated, attach the appropriate fittings and frits to one end of the column, leaving the other end free for connection to the slurry reservoir. Liquid Slurry Components Choose a solvent that has a densitivity as close as possible to that of the packing particles, or of a relatively high viscosity, or best of all a combination of both Knauer High Pressure Column Packing Device 7 properties. In addition, polar solvents should be used with polar packings; non-polar solvents should be used for non-polar packings. This will help to minimize aggregation of the packing. Some solvent properties are listed below. The inlet is to be connected to the slurry container, either directly or via an empty precolumn. After the packing material has been thoroughly dispersed in the slurry solvent pour the slurry into the reservoir (slurry container) rapidly. Carefully fill the remaining volume of the reservoir with solvent, being sure not to disturb the slurry. The top of the slurry container is closed and the small opening on top is connected to the KNAUER pump 1900 with a suitable capillary. Pressurize to a significantly higher pressure than the column will be subjected to during usage. The pump will deliver a packing liquid into the slurry tank-column arrangement. Pumping is continued until pressurizing solvent elutes from the end of the column (this can be determined by collecting effluent in a graduated cylinder and comparing the reservoir volume). Wait for the pump to cycle and turn off the inlet flow immediately. Allow the column to depressurize totally on the pump. Disconnect the column from the reservoir and carefully scrape away the excess packing from the column with a sharp razor knife. Attach the sieves, filter, sealing ring, end fitting and screw cap. The column is now ready for conditioning or use. Quality Testing the Column After finishing the packing process every column should be individually tested with a suitable test mixture. With these test mixture the packing quality and column performance can be controlled by calculation of different parameters (e.g. asymmetry, theoretical plates, capacity and selectivity). We recommend a simple test mixture (e.g. 2 or 3 neutral compounds like benzene, naphthalene or toluene) under isocratic conditions. KNAUER test solution Y1017 for reversed phase columns contains a mixture of thiourea, benzene and naphthalene and the KNAUER test solution Y1015 for normal phase columns contains a mixture of benzene and nitrobenzene. Insbesondere für den Einsatz großer Materialmengen zur routinemäßigen Reinigung stellen selbstgepackte Säulen eine ökonomische Alternative dar. Zu diesem Zweck bietet KNAUER den Säulenfüllstand zusammen mit einer Vielzahl von Füllmaterialien an. Säulenfüllstände können für Säuleninnendurchmesser von 2 mm bis 100 mm bezogen werden. Einführung Für die Herstellung von HPLC Säulen höchster Qualität müssen spezielle Packungstechniken angewendet werden. Im Labor werden Glasssäulen routinemäßig ohne zusätzlichen Druck einfach unter Ausnutzung der Schwerkraft gefüllt. Eine Aufschlämmung des Adsorbents in einem geeigneten organischen Lösungsmittel wird im Ultraschallbad drei Minuten homogenisiert und in die Säule gefüllt. Die Packungsdichte kann durch zusätzliches leichtes Klopfen an der Säulenwand verbessert werden. Dieses Verfahren ist jedoch nur für Glassäulen und Partikeldurchmesser über 20 µm anwendbar. Analytische und präparative Edelstahlsäulen und Partikeldurchmesser < 20 µm erfordern erheblich anspruchsvollere Packungstechniken. Allgemein gilt, dass mit steigendem Druck während des Füllvorganges ein besseres Packungsresultat erzielt wird. Zwei unterschiedliche Füllmethoden sind üblich, dabei wird entweder mit konstantem Fluss oder mit konstantem Druck gearbeitet. Methode mit konstantem Fluss Sowohl die KNAUER HPLC Smartline Pumpe 1000 als auch die präparative Pumpe 1800 können zum Säulenfüllen mit konstantem Fluss verwendet werden. Es werden üblicherweise moderate Flussraten gewählt, da diese Methode in erster Linie für druckempfindliche Adsorbentien zum Einsatz kommt. Bei einer während des gesamten Füllvorganges konstanten Flussrate steigt der Druck in dem Maße wie die Packung zunehmend komprimiert wird. Nachdem eine festgelegte Menge Lösungsmittel durch die Säule gefördert wurde, ist der Füllprozeß beendet. Die Säule wird von der Füllanlage demontiert und mit Sieben, Filtern, Dichtringen und Endverschraubungen versehen. Methode mit konstantem Druck Die pneumatische Pumpe 1900 von KNAUER ist zum Säulenpacken mit konstantem Druck die Pumpe der Wahl. Sie ist speziell für Arbeiten mit konstantem Druck entwickelt worden. Die Methode offenbart den Vorteil, dass die Säulen mit wesentlich höheren Drücken (bis zu 1000 bar) gefüllt werden können. Die pneumatische Pumpe wandelt den Antriebsluftdruck in den zur optimalen Packung innerhalb der Säule erforderlichen hydraulischen Druck um. Die Methode ist insbesondere für druckstabile Knauer Hochdruck Säulenfüllstand 11 Adsorbentien, wie Kieselgele geeignet. Der Druck fördert das aufgeschlämte Säulenfüllmaterial aus dem Vorratstank in die Säule. Während des gesamten Füllvorganges unter konstantem Druck nimmt die Flussrate in dem Maße ab, wie die Packung zunehmend komprimiert wird. Bei Säulendurchmessern von 3 bis 4,6 mm wird in der Regel mit einem Druck von ca. 600 bar gearbeitet. Für diese Säulendimension empfehlen wir den 250 ml Pumpenkopf. Zum Packen von präparativen Säulen (Säulendurchmesser > 16 mm) und Partikelgröße > 10 µm sind die größeren Pumpenköpfe (500 oder 1000 ml) zu empfehlen. Nach einer festgelegten Zeit wird der Füllvorgang beendet. Die Säule wird vom Füllstand demontiert und mit Sieben, Filtern, Dichtringen und Endverschraubungen versehen. Die Pumpe 1900 kann überall aufgestellt werden. Sie muss aber zusammen mit dem Vorratstank und der Säule durch ein Schutzschild gesichert werden. Ein höherer Antriebsluftdruck kann zu Schäden an der Pumpe führen. Diese fallen nicht unter die Garantie. Druckluftanschluss für die Pumpe 1900 Stellen Sie sicher, dass die für den Betrieb der Pneumatischen Pumpe 1900 verwendete Pressluftquelle einen Druck von bis zu 7 bar zur Verfügung stellt. Ohne Gegendruck liegt der Luftverbrauch bei etwa 80 l/min. Sobald die Pneumatische Pumpe 1900 gegen einen Flüssigkeitsdruck arbeitet, ist in Abhängigkeit von diesem der Verbrauch deutlich geringer. Der Druckluftanschluss der Pumpe befindet sich im unteren Bereich der Geräterückseite. Verschrauben Sie die Schlaucholive mit der Druckkupplung und schieben Sie den Druckschlauch (nicht im Lieferumfang) auf die Olive. Sichern Sie den Schlauch mit einer passenden Schelle. Schieben Sie die Druckkupplung auf den Druckluft-anschluss der Pumpe, siehe Abb. 3. Zum Trennen von der Druckluftquelle halten Sie die Schlaucholive der Druckkupplung mit einer Hand fest, während Sie den blauen Ring mit der anderen Hand von der Pumpe weg ziehen. Vergewissern Sie sich, dass alle Schlauchverbindungen für den Systemdruck und die Flussrate geeignet sind. Knauer Hochdruck Säulenfüllstand 13 Pressluftdruckeinflüsse Der resultierende Flüssigkeitsdruck erhöht sich oder sinkt in Abhängigkeit vom Eingangsdruck der Pressluft. Wird der Pressluftdruck konstant gehalten, verbleibt der Flüssigkeitsdruck ebenfalls praktisch konstant. Das geförderte Volumen hängt von dem vorliegenden Pressluftdruck, wie auch von dem durch eine Säule oder einen sonstigen Widerstand erzeugten Gegendruck ab. Das maximale Fördervolumen wird bei minimalem Flüssigkeitsdruck, < 1 bar, und der maximale Flüssigkeitsdruck bei minimaler Flussrate erreicht. Die Abb. 4 zeigt das Fördervolumen als Funktion des Flüssigkeitsdruckes für den 250 ml Pumpenkopf. In der Abbildung ist entsprechend auch der Tabelle „Technische Daten der einzelnen Pumpenköpfe“ der Grenzwert für den Flüssigkeitsdruck und den maximal zulässigen Pressluftdruck ersichtlich. Bei konstant anliegendem Pressluftdruck steigt die Flussrate, wenn der Flüssigkeitsgegendruck sinkt. Entsprechende Abbildungen für die anderen Pumpenköpfe sind dem Handbuch der Pumpe zu entnehmen. Das andere Ende bleibt zur Verbindung mit dem Slurrytank zunächst frei. 14 Knauer Hochdruck Säulenfüllstand Transportlösungsmittel Wählen Sie ein Lösungsmittel dessen Dichte der des Adsorbents möglichst nahe kommt, eine relativ hohe Viscosität aufweist oder am besten beide Bedingungen erfüllt. Beachten Sie auch das polare Lösungsmittel für polare Adsorbentien und unpolare Lösungsmittel für unpolare Packungsmaterialien verwendet werden. Einige Lösungsmitteleigenschaften sind in der Tabelle zusammengestellt. Die Aufschlämmung des Adsorbents im Lösungsmittel wird im Ultraschallbad homogenisiert und danach zügig in den Tank gefüllt. Die Slurry wird mit dem reinen Lösungsmittel vorsichtig bis zur Füllung des Tanks überschichtet. Die obere Öffnung des Tanks wird verschlossen und mit einer geeigneten Kapillare an die Pumpe 1900 angeschlossen. Setzen Sie die Füllapparatur mit der Pumpe unter einen Druck, der deutlich über dem voraussichtlichen Betriebsdruck der Säule liegt. Das Packungsmaterial wird mit dem Pumpeneluenten in die Säule transportiert. Dabei wird der Packungsvorgang durch kontinuierliches pumpen so lange durchgeführt, bis das Pumpenlösungsmittel am Säulenende ankommt (Bestimmung kann über Volumenabmessung mit einem Mesßzylinder erfolgen).Danach kann der Fluss abgeschaltet werden und die Einrichtung wird langsam und vollständig entspannt. Die Säule wird von der Füllvorrichtung abgenommen und der Überstand des Säulenkuchens vorsichtig mit einem scharfen Messer entfernt. Nach der Installation der Siebe, Filter und Dichtung ist die Säule zum Konditionieren bereit. Säulenqualitätskontrolle Nach Beendigung des Füllprozeßes sollte jede Säule mit einer geeigneten Standardprobe getestet werden, damit mehrere Parameter (z.B. Asymetrie, theoretetische Bodenzahl, Kapazität und Selektivität) die Packungsqualität und Säulenleistungsfähigkeit unter Beweis stellen. Wir empfehlen eine einfache Mischung von 2-3 neutralen Verbindungen für eine Trennung unter isokratischen Bedingungen zu verwenden. Die KNAUER Testlösung Knauer Hochdruck Säulenfüllstand 15 für Umkehrphasen ist eine Mischung aus Benzen, Naphthalen und Thioharnstoff und die für Normalphasen aus Benzen und Nitrobenzen.
