Currently, the only quantitative method available to assess HIV-1 nuclear import relies on quantitative PCR to amplify exclusively nuclear localised forms of the viral DNA. This method is important when assessing residual infection in clinical trials, determining the efficiency of integrase inhibitors, and for fundamental research seeking to understand mechanisms of HIV nuclear import. However, this technique is subject to experimental bias since the 2-LTR circles that are quantified represent a minute proportion of total viral DNA (~0.02 copies per cell), primer design does not discriminate auto-integration events, and 2-LTR circles may be used as a reserve supply for proviral integration. We wish to develop a new approach for quantifying the translocation of the HIV-1 PIC without PCR amplification or reporter construct, but rigorously quantitative and adaptable to high throughput screening. The aim of the project is to establish the proof of concept for this approach, which is currently the object of intellectual property protection. Once validated, this approach will be used for high-throughput screening of RNA interference or chemical-based libraries for conditions that hamper or enhance HIV-1 nuclear translocation.
