2. Aims of research
The main aims of the team are the development of (i) separation methods, namely capillary
zone electrophoresis (CZE), frontal analysis capillary electrophoresis (FACE), capillary
isoelectrofocusing (CIEF), isotachophoresis (CITP), and electrochromatography (CEC), and
(ii) the application of hyphenation of electrokinetic separation methods to mass spectrometry
(CE-MS), as a way to improve analysis selectivity and sensitivity. The work is also focused
on the characterization of non-covalent interactions in solution and at liquid-solid interfaces
and the determination of physicochemical properties of some solutes and media.
3. Research topics
- electrokinetic separations in hydro-organic or totally organic media and in the
presence of ionic liquids. Organic media enable to separate positional isomers such as fucose
monosulfates by modulating ion pairing and solvation phenomena, or amphetamines by
modulating ion pairing effects and ionization degree. Applications to the analysis of
hydrophobic proteins by isoelectrofocusing in the presence of glycerol were also developed.
- electrokinetic preconcentration methods, for the analysis of compounds at trace level.
Involving field-amplified based-stacking effect and isotachophoretic stacking effect (alone or
coupling both mechanisms), we succeeded in analyzing trace cationic impurities in ionic
liquids, chemical warfare agents in soils and metal cations in heat exchanger fluids of nuclear
power plants.
- methodology of the coupling of mass spectrometry (MS) to capillary electrophoresis
(CE). Several approaches were developed to maintain the electrical contact for electrokinetic
separation, in order to prevent from electrolyte depletion at the outlet end of a bare fusedsilica capillary for the analysis in negative polarity mode of polyanionic compounds having an
absolute-value of the electrophoretic mobility superior to the electroosmotic one. These
methodological developments allow to exploit fully MS advantages, such as its ability to
specifically fragment isomers which co-migrate in electrophoresis, which illustrates the high
complementarity of separation selectivity provided by CE and of detection selectivity provide
by MS. CE-MS thus associates high separation power, high sensitivity, and important mass
and structural information and is nowadays a key method to resolve complex mixtures such as
the ones encountered in metabolomic.
- interaction characterization. Experimental approaches and data treatment were developed
to characterize non-covalent interactions. As examples, active pharmaceutical ingredient
incorporation in liposomes (interaction constant and stœchiometry), protein-polysaccharide
complexes, surfactant aggregation threshold and inclusion complexation, such as that of ionic
liquid cations in cyclodextrins, were studied.
- characterization of physico-chemical properties. As an example, an on-line and
automated protocol which requires very low sample volumes was developed to determine
viscosity, conductivity and UV absorbance of ionic liquids. The determination of zeta
potential and charge density of nanoparticules or the measurement of size distribution of
synthetic and natural polyelectrolytes or nanoparticules constitute other examples.
Cuurent application fields:
- chiral molecules (enantioseparation)
- positively and negatively charged
- amphiphilic compounds, surfactants
- ionic liquids
nanoparticules (10-500 nm)
- synthetic polyelectrolytes
- trace inorganic and organic cations
- natural polyelectrolytes: proteins,
- cell metabolites
polysaccharides, DNA, humic acids
- warfare agents
