Implication du système ocytocinergique dans la vulnérabilité aux addictions induite par un stress précoce : étude translationnelle L'addiction désigne l'asservissement d'un individu à une substance ou une activité dont l’usage est chronique et compulsif. L’activation de la voie dopaminergique du système mésocorticolimbique ou « circuit de la récompense » est au cœur du contrôle de ces comportements. L’activité de cette voie peut être modulée par l’ocytocine, hormone impliquée dans la composante hédonique des relations sociales (attachement mère-enfant) et dans la régulation de l’axe corticotrope, axe de la réponse au stress. Le stress constitue un facteur de prédisposition aux conduites addictives. Notre équipe a pu montrer que le modèle de stress périnatal chez le rat (PRS, i.e. stress de contention au cours de la gestation associé à la diminution des soins maternels) est un modèle de programmation précoce de troubles liés au stress (anxiété, dépression, addiction) impliquant l’hyper-réactivité de l’axe corticotrope. Le phénotype « anxieux » des rats PRS est causé par une diminution de l’activité des synapses glutamatergiques de l’hippocampe ventral, dépendant de l’hypotonus ocytocinergique des rats PRS. Ce phénotype est corrigé par l’administration chronique d’un agoniste des récepteurs à l’ocytocine. Le projet vise à définir le rôle de l’hypotonus ocytocinergique dans la vulnérabilité à l’addiction que développent les rats PRS. Pour cela nous évaluerons chez les rats PRS i) l’activité du système ocytocinergique à l’aide d’agonistes et antagonistes des récepteurs à l’ocytocine ; ii) les conséquences de ces traitements sur les comportements de type « addiction » (Test d’auto-administration, préférence de place conditionnée), et de type anxieux (test d’émergence, labyrinthe en croix surélevé) et dépressif (test de nage forcée, test de préférence au sucrose) ; iii) l’effet de ces traitements sur l’activité du système mésocorticolimbique (récepteurs et transporteurs de la dopamine et de l’ocytocine des neurones dopaminergiques) particulièrement dans le noyau accumbens, structure clé de cette voie. Cette étude préclinique pourrait conduire à cibler le système ocytocinergique dans le traitement pharmacologique des conduites addictive et s’inscrit dans le cadre d’une subvention de l’agence nationale de la recherche (ANR) – Santé mentale et addictions (SaMentA) obtenue par le laboratoire : «Modulation périnatale de la programmation des troubles psychiatriques : ocytocine et étude translationnelle ». Maîtrise des techniques biochimiques (Fractionnement subcellulaire, western-blot, ELISA) Role of the oxytocinergc system in the vulnerability to addiction induced by early-life stress: translational research Addiction is the enslavement of an individual to a substance or an activity whose use is chronic and compulsive. The activation of the dopaminergic pathway of the mesocorticolimbic system or "reward circuit" lie at the core of the control of these behaviors. The activity of this pathway can be modulated by oxytocin, a hormone involved in the hedonic component of social relationships (mother-infant bonding) and in the regulation of the HPA axis, axis of the stress response. Stress is a predisposing factor to addictive behavior. Our team was able to show that the model of perinatal stress in rats (PRS, i.e. restraint stress during gestation associated with lower maternal care) is a model of early programming of stress-related disorders (anxiety, depression, addiction) involving hyperactivity of the HPA axis. The anxious-like phenotype of PRS rats is caused by a reduction in the activity of glutamatergic synapses in the ventral hippocampus as a consequence of the oxytocinergic hypotonus observed in PRS rats. This phenotype is corrected by chronic administration of an agonist of the oxytocin receptors. The project aims to define the role of oxytocinergic hypotonus in the vulnerability to addiction developed by PRS rats. Hence, we will evaluate in PRS rats i) the activity of the oxytocinergic system using agonists and antagonists of the oxytocin receptors; ii) the effect of these treatments on "addiction" (self-administration, conditionned place preference), anxiety(light and dark box test, elevated plus maze), depression-like behaviors (forced swim test, splash test) ; iii) the effect of these treatments on the activity of the mesocorticolimbic system (dopamine and oxytocin receptors and transporters in dopaminergic neurons) especially in the nucleus accumbens, a key structure of this pathway. This preclinical study could pave the way for targeting the oxytocinergic system in the pharmacological treatment of addictive behaviors and is in the line with a grant obtained by our group from the French National Research Agency (ANR) mental health and addictions (SaMentA) : “Perinatal modulation of psychiatric disorders programming: oxytocin and translational study”.
