Le développement de ces nouveaux outils pharmacochimiques radicalaires est articulé autour de trois méthodologies principales : les réactions SRN1 et apparentées, la stratégie TDAE, les réactions avec l’acétate de manganèse. Celles-ci sont associées à différentes réactions (cyclisation, couplage métallo-catalysé…) assistées ou non par la technologie micro-ondes et en privilégiant des protocoles respectueux de l’environnement. A l’heure actuelle, ces 3 méthodologies définissant de nouveaux outils synthétiques pour l’obtention de nouvelles molécules à potentialités anti-infectieuses sont déclinées selon 5 grands axes. I. Projet « Quinazopôle » (quinazolines à visée antiplasmodiale) II. Projet « imidazole et thiazole» (antiparasitaire : Trichomonas…) III. Projet « Quinone » (anti-Alzheimer et maladies à prions) IV. Projet « Benzodioxole et apparentés » (antiviral) V. Projet « Furane» (antiparasitaire : Leishmania…) Ces travaux de recherche s’appuient d’une part sur l’utilisation d’un plateau technique « microondes » original et performant dédié à la « Chimie Verte » (plateforme comprenant 4 réacteurs micro-ondes dont 1 avec passeur et permettant de réaliser des réactions, en parallèle, avec contrôle de la température et de la pression, dans des réacteurs de 0,5-2 mL jusqu’à 1 L) et d’autre part sur l’adhésion de la chimiothèque locale de l’équipe PCR (plus de 1500 composés) à la chimiothèque nationale permettant de favoriser de nouvelles collaborations avec des biologistes universitaires et industriels et mettre en évidence de nouvelles propriétés biologiques par l’intermédiaire de criblages. ! #$ % & ' ( ) * " !&' ( ) * +++%, - % . - %/ " 4 / PUBLICATIONS 2012-2013 PUBLICATIONS NATIONALES PUBLICATIONS INTERNATIONALES New methodology for the synthesis of thiobarbiturates mediated by manganese(III) acetate Bouhlel A., Curti C., Vanelle P.
