Les images correspondantes manquent de résolution spatiale en raison de la résolution temporelle d’acquisition au détriment de la résolution spatiale. Il est intéressant de pouvoir améliorer la résolution et la sensibilité de ces images dynamiques et fonctionnelles. L’application de méthodes numériques de traitement d’image pour améliorer ces images IRM est d’actualité en diagnostic clinique et préclinique. Nous proposons d’évaluer 2 pathologies pour appliquer et développer des méthodes de traitement d’images par un nouvel algorithme dit de ‘super résolution’. a) L'induction de la vascularisation tumorale est une étape clé de la progression tumorale et a été intensivement étudiée. La quantification des diverses caractéristiques des vaisseaux sanguins angiogéniques permet de fournir des informations de diagnostic et de pronostic, ainsi que d’évaluer l'efficacité d’un traitement anti tumoral. b) Par ailleurs, nous nous intéressons à des lésions rénales chez la souris où des images par IRM de diffusion permettent d’étudier l’état fibrotique. De premiers résultats obtenus par le partenaire 1 sur des images de perfusion IRM de tumeurs de souris ont montré l’amélioration conséquente fournie grâce à cette méthode de super résolution. Objectifs: Les objectifs de ce stage sont - d’utiliser des méthodes dynamiques d'imagerie et de diffusion par IRM pour les études de l'angiogenèse tumorale (perfusion) et de lésions rénales sur des modèles précliniques pour évaluer les thérapies. - de mettre au point ensuite sur ces images un traitement d’images basé sur la méthode dite de ‘super résolution’ permettant d’améliorer la sensibilité et la résolution de ces images IRM fonctionnelles. - d’évaluer le bénéfice de ce traitement sur le diagnostic de la pathologie et de thérapies. L’IRM classiquement utilisée révèle la distribution hétérogène des cellules tumorales et des capillaires, et teste leur fonctionnalité in vivo. L'utilisation d'agents de contraste permet de mesurer les paramètres de perfusion mesurés par méthodes d’IRM de contraste dynamique amélioré (DCE, Tofts et Kermode). Les analyses à partir de modèles pharmacocinétiques viseront à obtenir des cartes paramétriques reflétant les propriétés de perfusion du réseau vasculaire. Les images dynamiques nécessitant une acquisition pour enregistrer la cinétique de réhaussement de signal avec une bonne résolution 2/6 temporelle, la qualité de résolution des images sera diminuée. Les études d'imagerie seront développées sur le modèle murin CT26 de colon disponible dans notre laboratoire. Des Images de diffusion de reins pathologiques seront aussi testées. Il sera particulièrement intéressant de pouvoir améliorer la qualité des images ‘basse résolution’ par un traitement d’image développé par l’équipe partenaire dite ‘super résolution’. (Trinh et al ; Tran et al.) Ces méthodes sont basées sur l’apprentissage à partir de l’utilisation d’images similaires de même modalité d’imagerie et de bonne qualité en matière de sensibilité et de résolution, de contraste dans le but d’augmenter la résolution , le contraste et la sensibilité d’une image expérimentale dégradée. Durant le stage, l'étudiant sera formé sur différentes techniques d’acquisition IRM utilisées en oncologie et en pathologies rénales , des méthodes de traitement d’imagerie dynamique (DCE MRI et de diffusion), et le traitement d’image mathématique et informatique par une méthode dite de ‘super résolution’. Description des structures d'accueil: - Equipe UTCBS, Imagerie : Le laboratoire d’imagerie IRM et optique de l’UTCBS fait partie des plateformes d’imageries de Paris Descartes, doté d’une micro IRM 7T , ainsi que d’une plateforme d’imagerie optique. Les thèmes de recherche s’articulent autour de la méthodologie d’imagerie IRM et optique pour le suivi de pathologies et l’effet de traitement, du développement de nouveaux agents d’imagerie dédiés au diagnostic chez la souris. Equipe L2TI : Le thème de recherche du laboratoire concerne le traitement d’image incluant les images médicales IRM : l’extraction de l’information dans les images, en particulier à partir d’images basse résolution, l’édification et validation de modèles mathématiques, l’analyse quantitative d’image incluant la segmentation , la détermination de volumes. UTCBS, Imaging team : The UTCBS MRI and Optical Imaging laboratory is part of imaging platforms of Paris Descartes (PIV), equipped with a micro MRI 7T, and an optical imaging platform with cameras of 'luminescence and fluorescence imaging for preclinical studies. Research topics center around the MRI and optics imaging methodology for the tracking of diseases and the treatment effect, and development and exploitation of new imaging agents dedicated to diagnosis in mice. L2TI Team: The laboratory's research topic concerns the image processing of medical images including MRI: the extraction of information in the images, especially from low-resolution images, the construction and validation of mathematical models, quantitative image analysis including segmentation, determining volumes. Key words : Dynamic MRI, diffusion MRI, tumors, kidney disease, image processing by super resolution Duration : 6 months 4/6 Project description : The characterization of the disease state of tissues in small animals, is evaluated quantitatively by MRI with microvascular perfusion and diffusion imaging. The corresponding images lack of spatial resolution due to temporal resolution acquisition at the expense of spatial resolution. It is interesting to improve the resolution and sensitivity of these dynamic and functional images. The application of digital image post processing methods to improve these MRI images is topical in preclinical and clinical diagnosis. We propose to develop methods of image processing by a new algorithm called 'superresolution' applied to evaluate two disorders a) Cancer : Induction of the tumor vasculature is a key step in tumor progression and has been extensively studied. Quantification of various characteristics of angiogenic blood vessels allows providing information for diagnosis and prognosis, as well as to evaluate the effectiveness of an anti tumor treatment. Currently, several MRI imaging techniques provide information on the vasculature of the tumor. Perfusion MRI is recorded from the injection of tracer of commercial Gd complex type. The corresponding DCE (Dynamic Contrast Enhanced) images obtained with high temporal resolution could be improved by super resolution algorithm. First results by the partner 1 of MRI perfusion images of mouse tumors have shown significant improvement provided by this method of super-resolution. b) Furthermore, we are also interested in kidney damage in mice where images by diffusion MRI are used to study the fibrotic condition. Enhanced resolution images could be profitable to the quality of the fitting and the multiparametric diffusion derived data. Objectives: The objectives of this internship are - recording of dynamic imaging methods and diffusion MRI for studies of tumor angiogenesis (infusion) and kidney damage in preclinical models to evaluate therapies. - Then use of these images for processing based on the method called 'super-resolution' to improve the sensitivity and resolution of these functional MRI images. - Assess the benefit of this processing on the diagnosis and therapies of the pathology. a) The characteristics of the structure (volume of blood vessels), and tumor function (blood flow, vascular permeability, interstitial fluid pressure) can be measured by methods of dynamic contrast enhanced MRI (Tofts and Kermode). The analyses from pharmacokinetic models aim to obtain parametric maps reflecting the perfusion properties of the vascular network. Dynamic images require an acquisition to record the signal enhancement kinetics with good time resolution, the image resolution quality will be reduced. The assessment of tumor status and effect of antivascular therapies will be tested. b) Pathological kidney images obtained from diffusion MRI will also be tested for diagnosis of fibrotic type diseases and after the application of a therapy. From theses 2 imaging applications, it will be particularly interesting to improve the quality of 'low resolution' images by an image processing developed by the partner team called 'super- 5/6 resolution'. (Trinh et al. Tran et al) These methods are based on learning from the use of similar images of the same imaging modality and of good quality in sensitivity and resolution, in contrast to increase the resolution, contrast and sensitivity of a degraded experimental image. During the internship, the student will be trained on different MRI acquisition techniques used in oncology and renal pathologies, dynamic imaging processing methods (DCE MRI and diffusion), and in particular processing by computer and mathematical algorithm by a 'superresolution' method. Profile of the candidate The candidate has a biophysicist or physician profile in Master 2 , with expertise and interest in image and mathematical processing, or a computer science profile candidate interested in biological applications in diagnosis and therapy.
