Carreira, examiner Prof. Dr. Karl-Heinz Altmann, co-examiner Zürich, 2015 Abstract ix Abstract Schiglautone A I is a triterpenoid which has been extracted in 2011 from the stems of Schisandra glaucescens (Figure I). Its 6/7/9 tricyclic framework is unique among natural products. Noteworthy structural features include also 7 stereogenic centers, 3 of which are quaternary, and an olefin at the bridgehead position of the bridged 7/9 rings. This intriguing and challenging skeleton motivated our synthetic studies aiming at its total synthesis. The first part of this thesis describes the synthetic efforts towards the target and the successful construction of a key intermediate bearing all the carbon stereogenic centers. Figure I: Stucture of schiglautone A (I). After preliminary synthetic investigations, a convergent retrosynthetic approach was designed (Scheme I). Schiglautone A was expected to arise from tetracyclic enone II by retro-aldol. The 5membered ring was disconnected first, leading to tricyclic diene III. The central 7-membered ring of diene III was expected to be the result of a HECK cyclization of enol triflate derivative of ketone IV. Ketone IV was identified as a pivotal intermediate as it bears all the carbon stereocenters and contains an alkyne and a ketone moiety, which allow for a variety of cyclization strategies to build the 7-membered ring. Ketone IV was disconnected into two fragments of comparable sizes and complexity, aldehyde V and ketone VI. Scheme I: Retrosynthesis of schiglautone A (I); R = CH2CH2CH=C(CH3)2 and R’ = TBS. In order to probe the feasibility of the key step and gain insight on the reactivity of the molecule, the construction of a model system was investigated (Scheme II). Electrophilic alkynylation with Abstract x dichloroacetylene at C(10) and functionalization of the hindered ketone moiety delivered aldehyde VII in seven steps from commercially available material. The quaternary stereocenter at C(14) arose from a cyclopropane opening. A HECK cyclization was performed on the enol triflate derivative of VIII, which resulted in a mixture of products, among which desired diene IX and isomeric diene X were obtained. Scheme II: Synthetic studies on a model system. As the HECK cyclization showed promising results on the model system, the enantioselective synthesis of the two fragments V and VI was accomplished. Derivatization of WIELAND-MIESCHER ketone XI led to diol XII (Scheme IIIA). Oxidative cleavage allowed for the formation of a keto aldehyde, which was converted to aldehyde V in 5 steps. The construction of ketone VI was accomplished in 12 steps from acetophenone XIII (Scheme IIIB). Secondary alcohol XIV was obtained by NOYORI’s reduction of ketone XIII. The chiral information was transferred to XV by application of a lithiation-borylation strategy. BIRCH reduction of the arene and LUCHE reduction of the resulting enone provided allylic alcohol XVI. The enzymatic resolution of alcohol XVI was followed by a directed cyclopropanation to afford alcohol XVII. Reductive cleavage of the cyclopropropane delivered ketone VI with a d.r. of 17:1 and in enantiomerically pure form. Scheme III: Enantioselective synthesis of aldehyde V (A) and ketone VI (B). The two fragments, aldehyde V and ketone VI, were coupled by an aldol addition to give enone XVIII (Scheme IV). The quaternary stereocenter at C(14) was installed by opening of cyclopropane XIX, which was elaborated form enone XVIII. Ketone IV was thus obtained as a single diastereoisomer. The formation of this intermediate represents an important achievement towards the synthesis of schiglautone A, and application of the HECK cyclization should allow for the completion Abstract xi of the synthesis. The versatility of ketone IV renders it a valuable starting point for the exploration of alternative cyclization strategies. Scheme IV: Synthesis of ketone IV. In the second part of this thesis, the influence of a Cl-OH exchange on the biological activity of leukotoxin diol XX-threo was investigated (Scheme V). Diol XX-threo is a known toxin that has been surmised to be a causative agent of the acute respiratory distress syndrome and to be involved in multiple pathologies. Chlorine atoms and hydroxyl groups are classical isosteres, and their exchange can lead to modulation of biological activity, as illustrated by the example of the artificial sweetener sucralose. Scheme V: Synthesis of leukotoxin diol, its erythro analog, and the chlorinated analogs. R = H or Me. The diols XX and their chlorinated analogs XXI were synthesized in 6 steps from alkyne XXII and aldehyde XXIII. The induction of apoptosis by these different compounds was evaluated on a variety of cells by measuring the activity of caspase 3. Dichloride XXI-threo displayed a comparable toxicity to the diols towards HepG2 cells, and a reduced toxicity towards Hepa 1-6 cells. Notably, both the diols and the corresponding chlorinated compounds displayed variable activities depending on the type of cells. As the mechanism of action of leukotoxin diol is unknown, it was speculated that the difference of solubilities of the compounds as well as possible variations in their metabolism may account for the disparities in their ability to induce apoptosis. Abstract xii Résumé Schiglautone A I est un triterpénoïde extrait des tiges de Schisandra glaucescens en 2011 (Figure I). Sa structure tricyclique 6/7/9 est unique parmi les produits naturels. Parmi ses nombreuses remarquables caractéristiques structurales, on compte également 7 centres asymmétriques, dont 3 quaternaires et une oléfine en tête de pont. Sa fascinante architecture a déclenché notre intérêt à entreprendre des études en vue d’accomplir sa synthèse totale. La première partie de cette thèse décrit les efforts de synthèse engagés pour atteindre la molécule cible et la construction réussie d’un intermédiaire clé qui comprend tous les carbones asymétriques. Figure I: Stucture of schiglautone A (I). Après une première approche infructueuse, une stratégie de rétrosynthèse convergente a été conçue (Schéma I). Schiglautone A devrait provenir de l’énone tétracyclique II via une réaction de rétroaldolisation. Le cycle à 5 a été déconnecté en premier, menant au diène tricyclique III. Le cycle central à 7 devrait être le résultat d’une cyclisation de HECK sur le triflate d’énol dérivé de la cétone IV. Cette dernière a été identifiée comme un intermédiaire charnière, puisqu’elle comprend tous les carbones asymétriques. Elle porte également un alcyne et une cétone, ce qui permet l’utilisation d’une variété de stratégies de cyclization pour construire le cycle à 7. La cétone IV a été déconnectée en deux fragments de taille et de complexité comparable, l’aldéhyde V et la cétone VI. Afin de tester la faisabilité de l’étape clé et d’acquérir des connaissances quant à la réactivité de la molécule, l’élaboration d’un système modèle a été étudiée (Schéma II). L’addition électrophile Abstract xiii de l’alcyne à C(10) et la fonctionalisation de la cétone ont permis d’obtenir l’aldéhyde VII en 7 étapes. Le centre asymétrique quaternaire à C(14) a été introduit par ouverture d’un cyclopropane. Le triflate d’énol dérivé de VIII a été soumis à une cyclisation de HECK et a fourni un mélange de produits, parmi lesquels le diène IX et son isomère X ont été formés. Schéma II: Etude d’un système modèle . Les résultats prometteurs obtenus lors de la cyclisation de HECK sur le systéme modèle nous ont incités à entreprendre la synthèse asymétrique des deux fragments V et VI, avec succès. Le diol XII a été élaboré à partir de la cétone de WIELAND-MIESCHER XI (Schéma IIIA). Un clivage oxydatif a permis de former un céto-aldéhyde qui a été ensuite converti en aldéhyde V en 5 étapes. La cétone VI a été construite en 12 étapes à partir de l’acétophénone XIII (Schéma IIIB). Une réduction de NOYORI a été appliquée à la cétone XIII, donnant l’alcool secondaire XIV. Sa chiralité a été transférée à XV par une stratégie de lithiation-borylation. L’alcool allylique XVI a été obtenu par une rédution de BIRCH du noyau aromatique suivie d’une réduction de LUCHE sur l’énone ainsi synthétisée. La résolution enzymatique de l’alcool XVI suivie d’une cyclopropanation dirigée a permis la synthèse de l’alcool XVII. Un clivage réducteur du cyclopropane a fourni la cétone VI avec un d.r. de 17 :1, et sous forme énantiomériquement pure. Les deux fragments, l’aldéhyde V et la cétone VI, ont été couplés par une réaction d’aldolisation, donnant l’énone XVIII (Schéma IV). Le cyclopropane XIX, élaboré à partir de l’énone XVIII, a été ouvert pour donner le centre asymétrique quaternaire C(14), permettant ainsi l’obtention de la cétone IV comme un seul diastéréoisomère. La formation de cet intermédiaire représente une réalisation Abstract xiv importante en vue de la synthèse de schiglautone A, et l’application de la réaction de HECK sur cette cétone devrait permettre l’achèvement de la synthèse. La polyvalence de la cétoen IV en fait un point de départ précieux pour l’exploration de stratégies alternatives de cyclisation. Schéma IV: Synthèse de la cétone IV. Dans la deuxième partie de cette thèse a été étudiée l’influence d’un échange Cl-OH sur l’activité biologique de leukotoxin diol XX-threo (Schéma V). Ce dernier est une toxine reconnue que l’on a supposé être à l’origine du syndrome de détresse respiratoire aiguë et d’être impliqué dans de nombreuses pathologies. Les atomes de chlore et les groupes hydroxyl sont des isostères classiques, et leur inter-échange permet une modulation de l’activité biologique, comme démontré par l’exemple de l’échange réussi Cl-OH dans le cas de l’édulcorant sucralose. Les diols XX et leurs analogues chlorés ont été synthétisés en 6 étapes à partir de l’alcyne XXII et de l’aldéhyde XXIII. L’induction de l’apoptose par ces composés a été évaluée sur différentes cellules par mesure de l’activité de caspase 3. Le dichloride XXI-threo a montré une toxicité comparable à celle des diols sur les cellules HepG2, et une toxicité réduite envers les cellules Hepa 1-6. Cependant, une variabilité importante de l’activité selon le type de cellules a été observée, même dans le cas des diols. Le mécanisme d’action de leukotoxin diol étant à ce jour non-élucidé, l’hypothèse a été proposée que la différence de solubilité de ces composés ainsi que des variations possibles dans leur métabolisme pouvait être à l’origine des disparités observées dans les résultats biologiques.
