We will study whether the transferrin receptor (TfR1) is rerouted to the bacteria, how the bacteria can reprogram the characteristic cellular cycle of transferrin/TfR1, whether the expression and/or cellular localization of other proteins involved in iron metabolism within the eukaryotic epithelial cell are modulated during Chlamydia- infection. Chlamydia trachomatis (Ct) is not only responsible for trachoma, a major cause of acquired adult blindness, but also the most common bacterial sexually transmitted infection worldwide, including developed countries. Chlamydia pneumoniae (Cp) is responsible for frequent respiratory diseases among children, the elderly, and immunodepressed patients. As strict intracellular bacteria, Chlamydiaceae develop within epithelial cells of the conjunctiva or genital mucosa and divert most of the host cell metabolism for its own benefit. It has long been known that the development of this bacterium is highly dependent on the availability of host-cell iron, although the Chlamydiaceae genomes do not encode known siderophores or siderophore receptors. We have recently shown that this bacterium diverts the cellular transferrin to the bacterial inclusion and to the bacteria (1). In this Ph.D. thesis, the candidate will use non-tumoral models of mucosal human epithelial cells to examine: - whether part of the pool of the cellular transferrin receptor (TfR1) is rerouted together with transferrin to the bacterial inclusion and/ or to the bacteria. - how the bacteria can reprogram the characteristic cycle of transferrin/TfR1 (expression of particular TSNARE to the surface of the bacterial inclusion ...) - whether the expression and/or localization of other proteins involved in iron metabolism of the eukaryotic epithelial cell are modulated in during Chlamydial- infection. Depending on the results, the candidate will develop strategies to prevent the uptake of iron by the bacteria, which should significantly impact its development and open the path to new antibiotic strategies against this bacterium. Alternatively, if we identify new iron-acquisition proteins addressed to the bacterial inclusion, we will use these cargo proteins as trojan horses to address grafted- or nanoparticule-grafted-bactericidal molecules to Chlamydia within this inclusion.
