Ceci signifie que la taille et le contenu en gènes de la région pseudoautosomale, les gènes ancestraux ou acquis du chromosome Y, et les gènes du chromosome X échappant à l’inactivation varient fortement d’un phylum à l’autre. Ces variations ne sont connues en détail que chez deux espèces, l’homme et la souris, qui présentent des situations très différentes. De par sa position phylogénétique intermédiaire entre ces deux espèces chez les Euarchontoglires, le lapin est une espèce intéressante pour éclairer la dynamique d’évolution des chromosomes sexuels dans ce phylum. La caractérisation des chromosomes sexuels chez le lapin, espèce où ils sont très mal décrits, sera donc entreprise en établissant les points suivants: (i) la structure de la région pseudo-autosomale (PAR), en particulier l'emplacement de la frontière, le contenu en gènes et la taille; (ii) le contenu en gènes ancestraux du chromosome Y de lapin; et (iii) la liste des gènes du chromosome X échappant à l'inactivation. Cette caractérisation est une étape indispensable pour l'équipe d'accueil qui utilise l'espèce cunicole, depuis 2010, pour ses recherches sur la différenciation du sexe, notamment pour les approches de génomique fonctionnelle (transgenèse additive, mutagenèse ciblée par nucléase CrispR/Cas9). Title of the project: Characterization of sex chromosomes in rabbit Summary of the project (15 lines maximum): In mammals, the evolution of sex chromosomes occurred independently in each phylum. This means that the length and gene content of the pseudoautosomal region, the ancestral or acquired genes of the Y chromosome, and the X-chromosome genes escaping inactivation greatly vary from one phylum to another one. These variations are only known in detail in two species, human and mouse, which exhibit very different situations. Given its intermediate phylogenetic situation between these two species, the rabbit is an interesting species to shed light to the evolution dynamics of sex chromosomes. So the characterization of sex chromosomes in rabbits, a species where they are not well described, will be carried on in order to establish the following points: (i) the structure of the pseudoautosomal region (PAR), especially the location of the pseudoautosomal boundary (PAB), and the length and genes content; (ii) the ancestral gene content in the Y chromosome of rabbits; and (iii) the list of X chromosome genes escaping X inactivation. This PhD project represents a crucial step for the research team studying sex differentiation since many years, which made the choice to investigate the rabbit species since 2010. This species has been chosen in order to develop functional genomic studies such as additive transgenesis, but also gene editing by using nucleases like the CrispR/Cas9 system. Connaissances et compétences requises pour la thèse / Knowledge and skills required for the thesis Le candidat, titulaire d’un master 2 ou titre équivalent, doit posséder de bonnes connaissances de base en génétique, évolution, génomique, biologie moléculaire et biologie cellulaire. L’acquisition des techniques de base de biologie moléculaire (clonage, PCR, qPCR,…), de culture cellulaire et de FISH au cours des stages précédents est hautement souhaitable.
