CE Marked Material HIV-1 RNA Working Reagent 1 for NAT Assays NIBSC code: 99/634-xxx Instructions for use (Version 5.0, Dated 20/12/2013) This material is an „Annex II List A‟ IVD and complies with the requirements of the “EU in vitro diagnostic medical device directive 98/79/EC”. 1. INTENDED USE This product is CE marked for use as an IVD in Europe. In all other territories it is the sole responsibility of the Recipient to ascertain whether it can be used as an IVD. The HIV-1 RNA Working Reagent 1 manufactured by NIBSC has been on the market since 1996. It is intended for use in conjunction with diagnostic test systems / assays that detect HIV-1 RNA to monitor the performance of these systems. It can be used to monitor the consistency of test performance using statistical process control on a daily basis and, over a period of time, as a retrospective monitor of batch performance. The use of these products will indicate whether the sensitivity of an assay system in routine use is varying with time or with batch. It is for the user to determine limits from the use of our products and whether our products should be detected in every series of tests and the appropriate steps to take if our products are not detected or less well detected than previously. The products are supplied internationally to „professional‟ users, typically laboratories in hospitals, public health organisations, research organisations and commercial companies. They are not designed to provide any direct clinical data on any given specimen. These reagents should be used in addition to the batch specific positive and negative controls supplied with each batch of kit by its manufacturer. Our products are supplied for use in their final form and users are instructed not to further dilute the material other than as required in individual test procedures. 2. CAUTION This preparation is not for administration to humans. As with all materials of biological origin, this preparation should be regarded as potentially hazardous to health. It should be used and discarded according to your own laboratory's safety procedures. Such safety procedures should include the wearing of protective gloves and avoiding the generation of aerosols. Care should be exercised in opening ampoules or vials, to avoid cuts. The HIV-1 RNA Working Reagent 1 contains infectious HIV-1 diluted in human plasma and must only be handled in appropriate containment facilities by fully trained and competent staff and in accordance with the local national safety guidelines (such as the UK "Protection against blood-borne infections in the workplace: HIV and hepatitis", Advisory Committee on Dangerous Pathogens, HMSO, London) 3. The HIV-1 RNA Working Reagent 1 contains 1ml of liquid and consists of an HIV-1 genotype B virus diluted in pooled human plasma. The human plasma is supplied by the National Blood Service (NBS) and all packs have been pre screened by the NBS, prior to dispatch to NIBSC, and tested negative for the following markers: HBsAg, anti-HCV, anti-HIV, HCV RNA and anti-HTLV. 5. STORAGE The HIV-1 RNA Working reagent 1 should be stored at colder than -65oC prior to testing and each vial is intended to be used only once. The undiluted reagent should react positively in most NAT assays. 6. DIRECTIONS FOR OPENING Vials have a screw cap; an internal stopper may also be present. The cap should be removed by turning anti-clockwise. Care should be taken to prevent loss of the contents. Please note: If a stopper is present on removal of the cap, the stopper should remain in the vial or be removed with the cap. 7. USE OF MATERIAL No attempt should be made to weigh out any portion of the material The user should follow the assay kit manufacturer‟s instructions. The HIV1 RNA Working reagent 1 should be used in addition to the positive and negative controls supplied with each lot of kit and treated as if it were a routine test sample. Only the controls supplied with each lot of kit by the manufacturer are used to determine the validity of assays and to calculate the cut-off value that forms the basis for donor screening or diagnosis. The undiluted reagent should react positively in most NAT assays. THIS MATERIAL IS SUPPLIED FOR USE IN ITS FINAL FORM AND SHOULD NOT BE FURTHER DILUTED OTHER THAN AS REQUIRED IN INDIVIDUAL TEST PROCEDURES. Users should be aware that different batches of the same assay kit and/or different assay conditions may give different values with this reagent. 8. STABILITY Reference materials are held at NIBSC within assured, temperaturecontrolled storage facilities. Upon receipt, vials of the HIV-1 RNA Working reagent 1 should be stored at colder than -65oC. The stability of this material is being monitored by NIBSC. The stability of previous batches of the HIV-1 RNA Working reagent 1 have not lost potency when stored below -65oC for periods of up to 7 years. The shelf life assigned to working standard will be 5 years when stored at colder than -65oC. The expiry date is stated on the vial label. Each vial is for single use and it should be used on the day of thawing and then discarded. UNITAGE The HIV-1 RNA Working Reagent 1 code 99/634-*** is assigned a value of 3.56 IU (log10)/ml. The manufacturing process is identical to previous batches of 99/634-***. The working reagent has been assayed in triplicate in three independent assays in conjunction with the International Standard for HIV-1 RNA. The working reagent is considered acceptable if the overall geometric mean falls within the range: 2.99 – 3.80 Log10 IU/ml. The code 99/634 is followed by a 3 digit batch identification number. Users who have data supporting any deterioration in the characteristics of any Working Standard/Reagent are encouraged to contact NIBSC. A freeze-thaw indicator is included in the package along with interpretation instructions. If the freeze-thaw indicator shows that the reagent has been completely or partially thawed, it should not be used. UNITÉS Une valeur de 3.56 UI (log10)/ml à été attribuée au réactif de travail 1 pour la détection de l‟ARN du VIH-1, code 99/634-***. Le procédé de fabrication est identique à celui des lots précédents de 99/634-***. Le réactif de travail a été testé en triple exemplaire dans trois dosages indépendants conjointement à l'étalon international pour la détection de l‟ARN du VIH-1. Il est considéré comme acceptable si la moyenne géométrique globale se situe dans une plage de 2,99 – 3,80 log10 UI/ml. Le code 99/634 est suivi d‟un numéro d‟identification de lot à 3 chiffres. 4. CONTENU Pays d‟origine du matériel biologique : Royaume-Uni. Le réactif de travail 1 pour la détection de à l‟ARN du VIH-1 contient 1 ml de liquide composé d‟un virus VIH-1 génotype B dilué dans un plasma humain en pool. Le plasma humain est fourni par le National Blood Service (NBS) et tous les lots ont subi un dépistage préalable au NBS avant leur envoi au NIBSC et ont été trouvés négatifs vis-à-vis de marqueurs suivants : AgHBs, anti-VHC, anti-VIH, ARN du VHC et antiHTLV. 5. CONSERVATION Le réactif de travail 1 pour la détection de l‟ARN du VIH-1 doit être stocké à une température inférieure à -65 °C avant les tests et chaque flacon ne doit être utilisé qu‟une seule fois. Le réactif non dilué doit réagir positivement dans la plupart des tests NAT. 6. CONSIGNES D‟OUVERTURE Les flacons sont dotés d'un bouchon à vis et éventuellement, d'un bouchon étanche interne. Retirez le bouchon en le faisant tourner dans le sens inverse des aiguilles d'une montre. Veillez à ne pas renverser le contenu. Veuillez noter : si un bouchon interne est présent, il doit rester dans le flacon ou être retiré avec le bouchon à vis. 7. EMPLOI DU MATÉRIEL Ne pas tenter de peser une partie du matériel quelle qu'elle soit. L'utilisateur doit suivre les instructions fournies par le fabricant de la trousse de dosage. Le réactif de travail 1 pour la détection de l'ARN du VIH-1 doit être utilisé en plus des contrôles positif et négatif fournis avec chaque lot de trousses et traité comme un échantillon de test de routine. Seuls les contrôles fournis avec chaque trousse par le fabricant sont utilisés pour déterminer la validité des dosages et calculer la valeur limite qui constitue la base de dépistage des donneurs ou de diagnostic. Le réactif non dilué doit réagir positivement dans la plupart des tests NAT. STABILITÉ Les matériels de référence sont conservés au NIBSC dans des installations de stockage assurées, à température contrôlée. Les flacons de réactif de travail 1 pour la détection de l'ARN du VIH-1 doivent être conservés à une température inférieure à - 65 °C dès réception. La stabilité des lots précédents de réactif de travail 1 pour la détection de l'ARN du VIH-1 n‟a pas diminué au cours d'une période de conservation pouvant aller jusqu'à 7 ans à une température inférieure à -65 °C. La durée de conservation attribuée au réactif de travail sera de 5 ans quand il est conservé à une température inférieure à -65 °C. La date de péremption figure sur l‟étiquette du flacon. Chaque flacon ne doit être utilisé qu'une seule fois et ceci le jour même de sa décongélation, puis éliminé. Les utilisateurs disposant de données indiquant une détérioration des caractéristiques d'un standard de travail sont priés de contacter le NIBSC. Un indicateur de congélation/décongélation est inclus dans le lot avec les instructions d‟interprétation. Si cet indicateur indique que le réactif a été complètement ou partiellement décongelé, il ne doit pas être utilisé. Dans ce cas, veuillez contacter le NIBSC et fournir l'heure/la date de réception ainsi que l‟état du lot. Le NIBSC suit la politique de l‟OMS en ce qui concerne ses matériels de référence. 9. Das HIV-1-RNS-Arbeitsreagens 1 enthält 1 ml Flüssigkeit und besteht aus einem HIV-1-Genotyp B-Virus, der in gepooltem menschlichem Plasma verdünnt ist. Das menschliche Plasma wird vom Nationalen Blutdienst (National Blood Service, NBS) geliefert und alle Packungen sind vom NBS vor dem Versand an das NIBSC gescreent und für folgende Marker als negativ befunden worden: HBsAg, Anti-HCV, AntiHIV, HCV RNS und Anti-HTLV. 5. LAGERUNG Das HIV-1-RNS-Arbeitsreagens 1 muss vor dem Testen bei unter – 65°C gelagert werden, und jedes Fläschchen darf nur einmal benutzt werden. Außerdem kann ein Innenstopfen vorhanden sein. Der Schraubdeckel sollte durch Drehen gegen den Uhrzeigersinn abgenommen werden. Es muss darauf geachtet werden, dass der Inhalt nicht verschüttet wird. Bitte beachten: Ist bei Abnehmen des Schraubdeckels ein Stopfen vorhanden, muss der Stopfen entweder im Fläschchen verbleiben oder mit dem Deckel entfernt werden. 7. VERWENDUNG DES MATERIALS Es darf nicht versucht werden, einen Teil des Materials abzuwiegen. Der Benutzer muss die Anweisungen des Assay-Kit-Herstellers befolgen. Das HIV-1-RNS-Arbeitsreagens 1 muss zusätzlich zu den positiven und negativen Kontrollen verwendet werden, die mit jeder KitCharge geliefert werden, und wie eine Routinetestprobe behandelt werden. Nur die mit jeder Kit-Charge vom Hersteller gelieferten Kontrollen werden zur Feststellung der Gültigkeit von Assays und zur Berechnung des Grenzwerts benutzt, der die Basis für SpenderScreening oder Diagnose darstellt. DIESES MATERIAL IST ZUR VERWENDUNG IN SEINER ENDGÜLTIGEN FORM BESTIMMT UND DARF NICHT WEITER ALS FÜR INDIVIDUELLE TESTVERFAHREN ERFORDERLICH VERDÜNNT WERDEN. Die Benutzer müssen beachten, dass unterschiedliche Chargen des gleichen Assay-Kits und/oder unterschiedlicher Assay-Zustände mit diesem Reagens unterschiedliche Werte ergeben können. 8. STABILITÄT Referenzmaterialien werden beim NIBSC in gesicherten, temperaturgeregelten Lagerräumen aufbewahrt. Die Fläschchen mit dem HIV-1-RNS-Arbeitsreagens 1 müssen nach Erhalt bei unter –65°C gelagert werden. Die Stabilität dieses Materials wird vom NIBSC überwacht. Die Stabilität früherer Chargen des HIV-1-RNSArbeitsreagens 1 hat bei Lagerung unter –65°C über Zeiträume von bis zu 7 Jahren nicht an Wirksamkeit verloren. Die dem Arbeitsstandard zugewiesene Haltbarkeit beträgt bei Lagerung unter –65°C 5 Jahre. Das Verfalldatum ist auf dem Fläschchenetikett angegeben. Jedes Fläschchen ist für den einmaligen Gebrauch bestimmt und darf nur am Tag des Auftauens benutzt werden. Es muss anschließend entsorgt werden. Benutzer, die Daten vorzuweisen haben, die auf eine Verschlechterung der Eigenschaften eines beliebigen Arbeitsstandards/Reagens deuten, werden aufgefordert, sich an das NIBSC zu wenden. In dem Paket ist ein Frost-Tau-Indikator zusammen mit Anweisungen zur Interpretation enthalten. Wenn der Frost-TauIndikator anzeigt, dass das Reagens vollständig oder teilweise aufgetaut ist, darf es nicht benutzt werden. Wenn dies der Fall ist, wenden Sie sich bitte an das NIBSC und teilen den Zeitpunkt und das Datum des Erhalts des Pakets und seinen Zustand mit.
