Résumé du projet (15 lignes maximum) : Les cellules de Schwann (CSs) sont les cellules gliales myélinisantes du système nerveux périphérique qui, en synthétisant la myéline, entourent les axones et assurent une conduction rapide de l’influx nerveux. Il existe une communication étroite et indispensable entre les CSs et les axones dont dépend la migration et la division concomitante des CSs, contrôlée principalement par le facteur axonal : neuréguline dont les récepteurs erbB Schwanniens relaient la transduction du signal. Ce phénomène, d’une importance primordiale pour la myélinisation périphérique, est bien fascinant mais très peu connu. Le but de ce projet est 1) de pouvoir disséquer d’une façon précise chez le poisson zèbre, in vivo, le comportement des CSs (i.e migration, division, nature et polarité de migration..), depuis leur formation jusqu’à ce qu’ils commencent à myéliniser dans un animal intact ; 2) quelles voies sous-jacentes à la voie neuréguline/erbB sont impliquées dans ce processus et quel est leur(s) rôles?. Le projet fait recours à des approches d’imagerie confocale à haute résolution, des expériences d’ablation cellulaire au laser, à la microscopie électronique et à des approches cellulaires et moléculaires. Cette étude va contribuer à une meilleure compréhension du développement fondamental des CSs et potentiellement aux mécanismes pathogéniques impliqués dans des neuropathies périphériques. Title of the project: In Vivo monitoring of Schwann cell migration and division in zebrafish, cellular and molecular approach. Summary of the project (15 lines maximum): Schwann cells (SCs) are the myelinating glial cells of the Peripheral Nervous System (PNS) that ensure axonal ensheathment and rapid nerve impulse propagation along myelinated axons. There is a close and essential cross talk between SCs and the corresponding axons that is required for early SCs migration and division, mainly mediated by the axonal factor: neuregulin and the erbB receptors that relay the signal in SCs. This phenomenon is of great importance for SC development and myelination but remains poorly understood. The aim of this project is 1) to monitor and dissect the behaviour of SCs (i.e migration, division, type and polarity of migration..), since their birth till they start myelinating in an intact zebrafish embryo; 2) what are the signaling pathways downstream of the neuregulin/erbB one that are involved in this process and what is their role(s)?. The project will heavily rely on confocal microscopy at high resolution, SCs laser ablation, Electron Microscopy and cellular and molecular approaches.
