For each step of the cardiac action potential, flow of ions takes place in the myocytes. And for each movement of a specific ion, ion channels are implicated. When the movement of ions across the channels is disturbed, a prolongation of the QT interval may be observed on ECG. Then, ventricular arrhythmias may develop and they can be lethal if untreated. Among cardiac ion channel, hERG encodes the pore-forming subunit that conducts the rapid component of the delayed rectifier potassium current (IKr) in the heart. Drug-induced blockade of hERG may result in increase susceptibility to QT-interval prolongation, leading to ventricular tachycardia and sudden cardiac death. Such findings have led to a recommendation from the ICH (International Conference on Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use) that all new drugs be tested in assays to assess hERG block involvement before they are administered to humans. Assays include the patch-clamp analysis of "ion channel cell lines", heterologous expression systems where human ion channel proteins are expressed in noncardiac cell lines after stable gene transfer. Efficient ion channel cell lines expressing the hERG channel are a very sensitive way to assay hERG channel activity and predict cardiac risk early and accurately in the drug development process. And besides hERG, key ion channels implicated in the cardiac action potential can be tested. Bien qu'il existe de nombreuses molécules anticancéreuses, il demeure un besoin constant de nouvelles thérapeutiques plus efficaces et moins toxiques. Au début des années 70, émerge l’idée de cibler le système vasculaire tumoral. D’abord avec des stratégies d’inhibition de l’angiogénèse tumorale (Folkman J 1971), puis en 1982, le professeur Denekamp propose une autre approche : la destruction des néovaisseaux tumoraux (Denekamp J 1982). Cette idée a donné naissance à une nouvelle classe de molécules anticancéreuses : les « VDAs » pour Vascular Disrupting Agents. Ces molécules agissent sur les cellules endothéliales constituant les néovaisseaux tumoraux et provoquent un collapsus du système vasculaire qui produit un arrêt du flux sanguin conduisant à une nécrose de la masse tumorale. En recherchant des molécules antiprolifératives par un criblage phénotypique, nous avons identifié un nouvel agent VDA, la molécule D5. Par rapport aux autres VDA existants, D5 présente une structure chimique originale, une nouvelle cible thérapeutique et est administrable per os. Dans un premier temps, l’effet anti-prolifératif de D5 a été évalué in vitro sur un panel d’une dizaine de lignées cancéreuses humaines, montrant une efficacité particulière sur les lignées de cancers du poumon et du rein. Puis, les tests in vivo sur des modèles de xénogreffes avec une lignée de carcinome pulmonaire humain (HTB 177) et un modèle de cancer du rein (786-O), ont montré que la molécule est efficace per os chez la souris, provoquant une diminution de la masse tumorale par un phénomène de nécrose. Ensuite, les études en microscopie biphotonique ont permis de montrer que la nécrose massive était due à l’arrêt du flux sanguin dans les tumeurs, immédiatement après l’administration de D5, et ont permis de classer D5 parmi les VDA. De plus, les études de toxicité chronique sur des souris nude ont montré que D5 était bien tolérée in vivo. L'ensemble de ces résultats met donc en évidence le potentiel de D5 en tant que candidat médicament. Le brevet d’application de D5 pour le traitement du cancer, fournit la base pour son entrée dans des essais pré-cliniques et cliniques dans le cadre d’une start-up récemment créée, Ecrins Therapeutics. 1ère Journée Scientifique du Médicament – 23 Juin 2010 3 DIAGNOSIS, TREATMENT AND FOLLOW-UP OF DIABETES USING ZNT8-DIRECTED TECHNOLOGIES Yann de Boysson1*, Fabrice Chimienti1 1 MELLITECH SAS * Contact : [email protected] Mellitech est une société de R&D bio-pharmaceutique dans le domaine du diabète, créée à Grenoble en Octobre 2005 pour valoriser une technologie CEA/Université Joseph Fourier. La société compte actuellement 7 personnes et a obtenu des fonds publics (Oséo, ANR, ..) et privés pour un montant > 2MEuros depuis sa création. Son activité s’articule autour du développement de technologies ciblant un bio-marqueur majeur du diabète, la protéine ZnT8. ZnT8 est un transporteur du zinc très majoritairement exprimé dans la cellule beta pancréatique -sécrétrice d’insuline-, et a un rôle critique dans le processus de maturation et de sécrétion de l’insuline, qui est co-cristallisée avec le zinc intracellulaire. ZnT8 est une protéine encodée par slc30a8, un gène majeur du diabète de type II (Nature, Science, Nature Genetics 2007), et, en tant qu’auto-antigène majeur, est très fortement impliquée dans le processus de destruction auto-immun des cellules beta dans le diabète de type I (PNAS 2007). C’est à ce jour, avec l’insuline, la seule molécule impliquée dans les 2 types de diabète. Le diabète, en particulier le type II lié au manque d’activité physique et à l’obésité/la mauvaise alimentation, est la principale cause de cécité, de maladie rénales, d’amputations des membres chez l’adulte, et accroit considérablement le risque des complications cardiovasculaires. Prévalence mondiale 2010 : 285 millions de patients / 2030 : 440 millions. Les 3 projets de R&D menés au sein de Mellitech ou en collaboration sont : Le screening, la découverte et le développement de candidats-médicaments agonistes de ZnT8 pour le traitement du diabète de type II : l’activation de ZnT8 permet l’augmentation du stock d’insuline disponible via un accroissement modéré des niveaux de zinc intra-vésiculaire. Cible industrielle : industrie (bio-) pharmaceutique. Le développement de candidats-médicaments analogues de ZnT8 pour le traitement précoce du diabète de type I : l’injection de la protéine ZnT8 ou de peptides issus de ZnT8 va servir de leurre pour que le système auto-immun n’attaque pas les cellules beta en ciblant la protéine endogène. Cible industrielle : industrie (bio-) pharmaceutique. Le développement d’anticorps dirigés contre ZnT8 pour l’imagerie in vivo de la cellule sécrétrice d’insuline, visant à optimiser le diagnostic et le suivi du diabète : ces agents, en s’accrochant spécifiquement à la protéine ZnT8, permettront de visualiser in vivo les cellules beta fonctionnelles. Les études initiales se sont concentrées sur l’utilisation de transporteurs viraux. Mais, bien qu’efficaces, les problèmes associés à l’utilisation de ce type de vecteurs (immunogénicité, toxicité …) ont orienté les recherches vers le développement de vecteurs non-viraux tels que lipides cationiques, polymères, dendrimères et peptides.1 Les aminoglycosides naturels (antibiotiques polycationiques) sont connus pour interagir avec l’ARN et dans une moindre mesure avec l’ADN. Des conjugués de néomycine et de kanamycine avec des groupements hydrophobes (cholestérol, chaines grasses) ont été développés comme vecteur de gène et se sont révélés très efficaces.2 Afin de développer de nouveaux vecteurs non-viraux, de petite taille, moins toxiques et bio-inspirés, une série de lipides cationiques comportant comme tête polaire cationique la néamine, a été synthétisée et leur capacité de transfection évaluée. Prostacyclin analogues are the most effective drugs to treat sclerodermic digital ulcers, but their systemic use is limited by their frequent side effects. Topical administration of these drugs may be a way of getting around the toxicity of systemic treatments. Iontophoresis, a non invasive controlled transdermal delivery system using a low-intensity electric current, could be an interesting alternative to ointments. Some authors suggest vasodilator iontophoresis as potentially interesting in the treatment of digital ulcerations in scleroderma. However, a commonly used vasodilator such as sodium nitroprusside is not a good candidate given its short half-life and transient effect. We recently showed that cathodal iontophoresis of iloprost and treprostinil increased cutaneous vascular conductance (CVC) on the hindquarter skin of rats. We tested whether the prostacyclin analogues treprostinil and iloprost, delivered by cutaneous iontophoresis, induce sustained vasodilatation in healthy volunteers. Methods. Twenty healthy volunteers received treprostinil by cathodal iontophoresis on the forearm. Skin blood flux was quantified using laser Doppler imaging. Results. Treprostinil at 250 µM induced an increase in cutaneous vascular conductance AUC80min (31897 +/- 24390 %BL.min) compared to NaCl 0.9% (P<0.005), treprostinil 2.5 µM (P<0.005) and treprostinil 25 µM (P<0.005). This was confirmed when flux was recorded for up to 10 h. Systemic and skin tolerance of treprostinil were good. Iloprost was stopped early because of local toxicity. Conclusions. Cutaneous cathodal iontophoresis of 250 µM treprostinil induced a large and sustained increase in cutaneous blood flow in healthy volunteers, without systemic or topical side-effects. The kinetics of the increased CVC following iontophoresis of treprostinil would be compatible with intermittent iontophoresis of the drug. Therefore, topical delivery of treprostinil via iontophoresis could be considered as an alternative method of administration to increase the drug concentration locally avoiding its major systemic adverse effects. Treprostinil cathodal iontophoresis should be further investigated as a new local therapy for digital ulcers in patients with scleroderma. Disrupting interaction between PDZ-containing protein PSD-95, and its natural ligand 5-HT2A receptor, was found to reduce hyperalgesia in a rodent model of neuropathic pain. Thus, inhibiting this interaction could lead to the development of a novel class of analgesic agents. Interaction of natural ligands with PDZ domains can be divided into a conserved generic site (S0) and an adjacent specificity-determining site (S1). Our strategy is to design bidentate ligands constituted by: one generic ligand interacting with S0, linked by a spacer to a specific ligand interacting with S1. These bidentate ligands are designed to inhibit PDZ interaction with specificity. A molecular docking study allowed us to design a generic S0 ligand, to which was attached a variable size diamine linker, itself linked to an amino acid aimed to interact with the S1 site of PDZ domains. A series of 15 novel bidentate ligands was prepared in 6 to 11 steps in good overall yields (24-43%). Some of these ligands showed an inhibitory activity against 5HT2A serotonin receptor/PSD-95 interaction, as assessed by pull down assay using a synthetic peptide corresponding to the 10 C-terminal residues of the receptor as a bait. However, these time lags are not compatible for instance, with the analysis of some infectious disease. There is a strong need to develop new tools to identify pathogenic bacteria in a shorter time. To achieve this goal, Surface Plasmon Resonance imaging (SPRi) technology has been successfully used for the specific detection of growing bacterial populations using proteins and/or antibodies targeting these bacteria. This strategy of simultaneous bacterial growth monitoring and specific detection, using the SPR imaging, enabled the detection of less than 50 cfu inoculation within few hours. First Streptococcus pneumoniae R6 was used as a model, and then Escherichia coli K12 provided the proof that this method can be applied to other strains. Our quantitative results are consistent with the expected doubling time of bacteria reported in the literature. The first step of this process consists in lesion recognition by proteins such as DDB2. Using breast cancer cells genetically modified to present different levels of DDB2 expression, we found that this protein sensitizes cells to UV-B induced stress and platinating agents. Using HPLC-MS-MS, we have followed DNA repair of 6 different DNA lesions after cells exposure to UV-B and of 1,2-d(GpG) crosslinks after cells exposure to cisplatin. We have shown that DDB2 enhances repair activity for UV-B induced cyclobutane pyrimidine dimers. On the contrary, DDB2 does not seem to play a role in repair of cisplatin lesions. Experiments using DNA probes damaged by UV or cisplatin allowed us to trap proteins associated with DNA lesions. Using this approach, we have demonstrated that the affinity of DDB2 was stronger for UV induced alterations than for cisplatin adducts. The weak affinity of DDB2 for cisplatin crosslinks was confirmed by Surface Plasmon Resonance. As a conclusion, these preliminary results confirmed the role of DDB2 in DNA repair of UV-induced damage but not for those produced by cisplatin. Relationships between DDB2 and platinating agents sensitization may involve mechanisms not requiring direct DNA damage recognition by DDB2. 1ère Journée Scientifique du Médicament – 23 Juin 2010 10 LIPID NANOPARTICLES: A MULTIFUNCTIONAL PLATFORM FOR THE CONTROLLED DELIVERY OF SIRNA J. In this way, this mechanism is an useful approach in research for the understanding of dysregulation occurring in pathogenesis, especially in cancerogenesis. Moreover, siRNA shows a specific and effective gene silencing activity through a sequence specific down-regulation of the complementary messenger RNA as well as a tremendous potential as therapeutic agent for treatment of numerous diseases. However, the siRNA-mediated gene knock down requires that siRNA can reach the protein complex RISC in the cytoplasm, where siRNA allows the specific degradation of corresponding mRNA. Unfortunately, the siRNA is relatively vulnerable in the extracellular environment due to the presence of degradation enzymes and its high molecular weight associated to its anionic charge limit considerably its cell incorporation across the plasma membrane. To overcome these obstacles, multifunctional nanoparticles comprising an imaging contrast agent are emerging as an original and promising approach in the monitored delivery of siRNA. We have designed lipid nanocarriers entrapping lipophilic cyanine derivatives for fluorescence imaging purposes, as a biocompatible alternative of the well-known quantum dots. Recently, we have adapted this formulation of lipid nanoparticles by incorporating some cationic compounds in order to design a novel nanocarrier dedicated to siRNA delivery. These modifications do not influence the colloidal stability of the dispersion and enable a complexation of positively charged nanoparticles with TAMRA-stained siRNA. The transfection efficiency using these complexes is under investigation in a cell model overexpressing the green fluorescent protein (GFP). Malgré leur fréquence, elles sont largement méconnues. On distingue deux formes principales de pathologies : la rectocolite hémorragique et la maladie de Crohn, qui partagent de nombreuses similitudes épidémiologiques, thérapeutiques et cliniques. La principale différence réside dans la localisation des lésions. Le récepteur CB2 est le deuxième sous-type de récepteurs aux cannabinoïdes connu à ce jour. Sa localisation au niveau des cellules immunitaires met en évidence l’implication de ce récepteur dans les effets immunomodulateurs induits par les cannabinoïdes. De récentes publications s’accordent à donner un rôle de plus en plus important au récepteur CB2 dans l’inflammation et le contrôle de la réponse immunitaire. Actuellement, on compte uniquement 4 agonistes CB2 en développement pour des indications thérapeutiques relatives à la douleur inflammatoire, la dermatite atopique, l’allergie (prurits) et les accidents vasculaires cérébraux. Aucun ligand CB2 n’est actuellement en développement dans le traitement des MICI. Notre projet a pour objectif de faire émerger une nouvelle classe d’agents thérapeutiques ciblant les récepteurs CB2, et utilisable dans le traitement des MICI. Nous avons choisi de nous focaliser sur cette niche thérapeutique particulière des MICI suite aux premiers résultats très prometteurs obtenus in vivo. A l’heure actuelle, la preuve du concept a déjà été validée et ce projet a donné lieu à 7 publications, deux brevets et deux ANR. Ainsi, l'objectif général du projet est de sélectionner parmi les différents agonistes CB2 obtenus, un candidat médicament possédant des propriétés pré-ADME-Tox et des résultats in vivo suffisants pour pouvoir être proposés à un partenaire industriel. L’étude des propriétés ADME-Tox sera un facteur important dans le choix des composés à évaluer in vivo. In a first step, the efficiency of a MALDI-TOF procedure to characterize the average molecular weight of the derivative bulk mixture was demonstrated by comparing the results with those obtained from complementary NMR and HPLC techniques. The use of recombinant proteoliposomes containing therapeutic membrane proteins is a recently developed technology in our laboratory, allowing biologically active proteins to penetrate across the plasma membrane of eukaryotic cells. Here, we report the therapeutic effect of proteoliposomes containing pro-apoptotic human Bak integrated into long-circulating liposomes produced with an innovative cell-free expression system. In vitro experiments on different glioma cell lines demonstrate that Bak proteoliposomes are able to be internalized and to exert an apoptotic effect via activation of the mitochondrial pathway (cleavage of caspase 9, release of cytochrome c). Preliminary results on mice subcutaneous glioblastoma model show a manifest delay in the tumor growth and a total regression of the tumor and survival of 60% of the treated mice. Nanoprecipitation of the new facial amphiphiles, or binary mixtures of them, resulted in nanoparticles with average hydrodynamic diameters ranging from 100 to 240 nm that were stable in suspension for several months (see Fig., A). The size, surface potential and topology of the nanoparticles are intimately dependent on the functionalization pattern at the CD scaffold. Interestingly, CD-decorated CD-based nanoparticles of 70 nm diameter could be prepared from amphiphilic heterodimers (see Fig., B). The possibility of inserting biorecognizable ligands in the architecture by taking advantage of the inclusion capabilities of the external CD units offers new and versatile possibilities for such systems. The efficient encapsulation ability of a poorly water soluble drug (diazepam) was demonstrated for some derivatives, the drug release profile being dependent on the type of formulation (nanomatrix or nanoreservoirs). La conception de molécules avec propriétés biologiques est d’intérêt majeur et ouvre un champ de recherche très important en biologie moléculaire et en recherche pharmacologique. Due à leur large specifité de l’activité antimicrobienne contre les bactéries Gram positifs et Gram négatifs, aussi que contre unes des espèces pathogéniques des fongus et protozoaires, les sels quaternaires d’ammonium représentés unes des plus utilises antiseptiques et désinfectants. Nous avons mis au point une synthèse de nouveaux sels symétriques et non-symétriques de bispyridinium et on a investigue leur activité antimicrobienne. Il est également étroitement associé à l’apparition de lymphomes B chez l’immunodéprimé et du carcinome indifférencié du nasopharynx. La protéase virale, codée par le gène BVRF2, est essentielle pour la formation de nouveaux virions lors du cycle viral réplicatif. Des siRNA ciblant l’ARN messager de cette protéine ont été évalués en culture cellulaire et ont montré un fort potentiel d’inhibition de l’ARNm ainsi que de la production de particules virales infectieuses (S Larrat, Antivral Therapy, 2009). Afin de poursuivre l’évaluation de cette stratégie thérapeutique anti-EBV, l’efficacité du meilleur siRNA antiprotéase a été comparée en culture cellulaire à celle des antiviraux conventionnels ciblant la polymérase virale. Matériel et méthode : Les cultures cellulaires sont réalisées à partir de cellules épithéliales 293-B95.8/F contenant un bacmide codant pour l’EBV (souche B95-8) fusionné à la GFP (green fluorescent protein) développées par l’équipe d’Hammerschmidt (Hammerschmidt, PNAS, 1998). L’expérience d’inhibition de la multiplication virale est réalisée sur des cellules dans lesquelles le cycle réplicatif d’EBV a été induit par la transfection de plasmides codant pour des gènes viraux activateurs (BZLF1 et Balf4). L’activité du siRNA anti-protéase PR1 est évaluée en parallèle de celle du ganciclovir (10 µg/ml), du foscarnet (150 µg/ml), et du cidofovir (6 µg/ml) par la diminution de l’ARNm protéase en comparaison de l’ARNm d’un gène cellulaire (GAPDH) et par l’inhibition du pouvoir infectieux des surnageants de culture mesurée en sur-infectant des cellules Raji. L’association du siRNA PR1 et du ganciclovir a également été évaluée. Résultats : Pour l’ensemble des antiviraux testés, l’inhibition de l’ARNm protéase, par rapport au contrôle sans antiviral, est significative (p<0.05) alors que l’ARNm GAPDH ne change pas (p>0.1). Cette inhibition est identique pour le siRNA PR1, pour le ganciclovir, le cidofovir et le foscarnet mais statistiquement plus importante pour l’association siRNA PR1 + ganciclovir (p<0.001). En revanche, seul le siRNA anti-protéase, en association ou non, permet d’obtenir une diminution significative (p<0.01) du titre viral infectieux. Conclusion : Ces résultats confirment l’intérêt des siRNAs ciblant la protéase d’EBV comme alternative thérapeutique pour inhiber la réplication du virus. En effet, si les médicaments anti-herpesviridae ciblant la polymérase montrent une activité intéressante sur la baisse des ARNm viraux, seuls les siRNAs anti-protéase permettent de diminuer le titre viral infectieux. The World Health Organisation (WHO) estimates that 120-140 million people are infected worldwide (2-3% of the world population), while 3-4 million are newly infected each year. Chronic HCV infection is associated with chronic liver inflammation and fibrosis and can lead to cirrhosis and hepatocellular carcinoma in absence of treatment. The HCV RNA-dependent RNA polymerase (RdRp NS5B) is a particularly attractive target because of its key role in viral replication and the fact that this enzyme has no functional equivalent in mammalian cells. Although a large number of inhibitors of the four allosteric sites of the enzyme was identified, none of them reached the market yet due to various concerns, including safety. We have thus identified naturally occurring 2-benzylidenebenzofuran-3-ones (aurones), flavonoid derivatives with promising pharmaceutical potential, as new templates for non-nucleoside HCV RdRp inhibitors. We firstly discovered natural aureusidin as a potent inhibitor of HCV RdRp. The aurone target site, identified by site-directed mutagenesis, is located in Thumb Pocket I of HCV RdRp. The RdRp inhibitory activity of 42 aurones was rationally explored in an enzyme assay. Molecular docking studies were used to determine how aurones bind to HCV RdRp and to predict their range of inhibitory activity. Seven aurone derivatives were found to have potent inhibitory effects on HCV RdRp, with IC50s below 5 µM and excellent selectivity. The most active aurone analogue was (Z)-2-((1-butyl-1H-indol-3-yl)methylene)-4,6-dihydroxybenzofuran-3(2H)-one, with an IC50 of 2.2 µM. La recherche et développement en instrumentations optiques est particulièrement active et de nouveaux systèmes sont proposés et améliorés régulièrement. OPTIMAL est une plateforme technologique académique âgée de 6 ans équipée d’instrumentations optiques de pointe pour la bioluminescence, la fluorescence 2D en temps réel à différentes échelles et l’imagerie 3D bimodale couplant la fluorescence et les rayons X. Certaines des instrumentations du parc sont disponibles dans le commerce mais la plupart a été développé localement en collaboration avec des partenaires industriels. A partir de sa gamme technologique OPTIMAL a également développé des modèles biologiques et des procédures pour accélérer la recherche et développement en oncologie préclinique. Ces procédures ainsi que des protocoles personnalisés comme par exemple (1) le suivi non invasif de la progression tumorale chez la souris en bioluminescence, (2) l’utilisation de la fluorescence en temps réel in vivo pour l’évaluation de la biodistribution et des pharmacocinétiques de nouveaux composés et (3) l’évaluation de l’angiogénèse in vivo en fluorescence 3D, sont proposés aux équipes académiques et aux industriels. Several anti-cancer drugs like paclitaxel (PTX) used for breast cancer treatment, have hydrophobic backbone chain and poor aqueous solubility, leading to reduced therapeutic efficiency of the drug. Optimization of present cancer treatment is important as it induces several side effects to other organs associated with drug and drug carrier. There is a crucial need to develop drug carriers that could decrease the toxic side effects and increase the water solubility. In present work, polyelectrolyte films were constructed using the layer-bylayer assembly of chemically modified hyaluronic acid (HA) with induced hydrophobic nanocavities as anionic polyelectrolyte and quaternised chitosan (qCHI) or poly(L-lysine) (PLL) as cationic polyelectrolyte. The films were characterized using fluorescence and absorbance measurements. Paclitaxel was loaded inside the film in tunable amount depending on the number of deposited layers, the initial paclitaxel concentration as well as on the type of polycation. The films were further enzymatically hydrolyzed indicating biodegradability. Influence of chemical cross-linking of the film on release kinetics of paclitaxel was studied. Furthermore, metabolic activity and viability of two types of breast cancer cells (HBL-100 and MCF-7) cultured on the Paclitaxel-loaded films were investigated over several days of culture. Ces clusters de lésions peuvent conduire à des cassures doubles brins et sont beaucoup plus toxiques et mutagènes que les mono lésions. Ils peuvent être à l'origine de processus de cancérisation. Notre objectif est d'étudier à la fois d'un point de vue biochimique et structural ces groupes de lésions de l'ADN et leur réparation chez une bactérie radiorésistante DEIRA (Deinococcus Radiodurans). Cette bactérie est capable de survivre à l'exposition à des doses de plusieurs milliers de grays, responsables de la formation de centaines de cassures double brin par génome. De telles doses sont des centaines de fois supérieures aux doses létales pour la plupart des autres bactéries. Nous nous intéressons particulièrement à une protéine de réparation, la Fpg qui reconnaît spécifiquement une base oxydée, la 8oxoguanine. Nous présenterons le protocole d'expression et de purification de cette protéine que nous avons développé au laboratoire. Parallèlement, nous avons débuté une étude structurale par RMN et Modélisation moléculaire d'oligonucléotides contenant 2 lésions : selon la position relative des lésions dans l'ADN nous avons montré que la protéine Fpg d'E. Coli est active ou inhibée. The bottleneck of this strategy is the hydrophobic composition of cellular membranes that restrains the uptake of these molecules. A new and efficient protein delivery vector deriving from the Epstein-Barr virus transcriptional activator ZEBRA has been recently exploited in our laboratory. Analysis of different truncated forms of ZEBRA revealed that the minimal domain (MD) required for cellular uptake is a 48 amino acids region spanning residues 170-220. Fusion of ZEBRA-MD to different reporter proteins, such as EGFP or β-galactosidase, is sufficient to induce the internalization into living cells. ZEBRA-MD reporter proteins transduction through the cell membrane requires binding to cell surface-associated heparan sulfate proteoglycans, and occurs mostly via direct translocation. MD is able to transduce a wide range of mammalian cells with 100% efficiency and no toxic side effects have been shown yet. MD-delivered proteins remain functional when transduced into cells, as demonstrated by the enzymatic activity of internalized MD- βgalactosidase. Treatment of severe haemophilic patients is based on the administration of factor VIII or IX concentrates. However, some patients develop antibodies inhibiting the activity of the therapeutic clotting factor. In these cases, haemostatic therapies which bypass the tenase complex must be used : recombinant factor VIIa or activated prothrombin complex concentrates. Unfortunately, some patients do not respond (1) and rare cases of thromboembolism are reported. Alternative therapies for a better efficiency and safety need therefore to be developed. Gla domain less Xa (GDXa) is a truncated factor Xa. As it is unable to bind phospholipid membranes, it is almost devoid of prothrombin into thrombin activation capacity. In this work, we test GDXa to restore thrombin generation in severe haemophilic patients plasmas. We demonstrate that GDXa is able to restore in vitro thrombin generation of severe A haemophilic plasmas in an exclusive tissue factor dependent way in contrast to factor Xa. We assume that GDXa may compete with Xa for binding to TFPI. The formation of a GDXa/TFPI complex may limit the formation of the factor Xa/TFPI complex which is essential to shutdown the tissue factor/VIIa coagulation initiation complex. By deshinibiting the initiation of coagulation, GDXa may restore thrombin generation. At first, we have initially compared the enzymatic inhibition profiles of GDXa by TFPI and we have shown a lower TFPI inhibition capacity against to GDXa as against to Xa. We have also shown that despite its very short half life in amidolytic assays after incubation in plasma, GDXa retains the capacity to restore thrombin generation in haemophilic plasmas at least 40 min after incubation. Actuellement, parmi les patients, environ 70% présentent des tumeurs superficielles, 25% des tumeurs infiltrantes et 5% des tumeurs de type métastatiques. Cependant, 50 à 80% des tumeurs superficielles récidivent, et 16% évoluent vers une infiltration des tissus. Ces tumeurs sont peu chimiosensible vis-à-vis des médicaments disponibles actuellement et il est donc nécessaire de trouver de nouvelles molécules plus efficaces. Les flavonoïdes sont des métabolites secondaires ubiquitaires chez les végétaux et appartenant à la famille des polyphénols. Ils interviennent dans plusieurs processus biochimiques et physiologiques des plantes et présentent une activité inhibitrice de nombreux systèmes enzymatiques in vitro tels que le récepteur à l'EGF ou certaines Cdks. De plus, les flavonoïdes ne semblent pas être toxiques pour l’organisme, puisqu’ils sont consommés en grandes quantités dans l’alimentation courante (agrumes, thé vert, soja). Nous avons récemment développé un nouveau dérivé de chalcone, le 1-(2,4dimethoxyphenyl)-3-(1methylinodolyl) propenone (ou IPP51) qui, après 24 h de traitement à une concentration de 5µM, provoque une accumulation au stade pro-métaphase de la mitose de cellules dérivées de tumeurs de vessie avant d'induire l'apoptose au bout de 48h. Ce blocage en mitose s'accompagne de fuseaux mitotiques anormaux et nos études préliminaires suggèrent que l'IPP51 agit comme un inhibiteur de la polymérisation des microtubules. Enfin, des études menées sur des souris porteuses de tumeurs obtenues par injection sous-cutanée de cellules humaines dérivées de tumeurs de vessie montrent que l'injection d'IPP51 réduit significativement la croissance tumorale. L'urothélium est un tissu qui se renouvelle très lentement. En conditions normales, la plupart des cellules urothéliales sont quiescentes. Ciblant la mitose, l'IPP51 devrait donc toucher préférentiellement les cellules tumorales qui prolifèrent plus rapidement. Actuellement, seules des thérapeutiques curatives sont disponibles, traitant cependant de manière imparfaite ou symptomatique les différents symptômes radio-induits. Différentes situations permettent d’anticiper une exposition à des doses significatives de radiations, et disposer d’une méthode prophylactique visant à protéger le corps humain des dommages radio-induits serait une avancée majeure, et ce quel que soit le contexte, civil, militaire, médical : incident radiologique à bord d’un bâtiment de la Marine Nationale à chaufferie nucléaire (12 réacteurs embarqués en 2011) ; troupes militaires en opération exposées à un risque probable ou imminent d’irradiation externe ; exposition de la sécurité civile après un accident radiologique ou nucléaire ou un attentat de type « source radioactive cachée » ; personnel responsable de la maintenance et du démantèlement des centrales nucléaires ; exposition du personnel responsable du fonctionnement des irradiateurs industriels (stérilisation matériels ou aliments) ; exposition de la population et des intervenants lors d’accidents de centrales nucléaires (exemple récent de Fukushima I) ; protection des tissus sains lors des séances de radiothérapie ou même lors d’examens radiologiques répétés. En dépit des nombreux projets de recherches dans ce domaine, seul le N-(3-aminopropyl)-2aminoéthylphosphorothioate (WR-2721 ou Amifostine), découvert à la fin des années 1960 aux Etats-Unis et commercialisé sous le nom d’Ethyol (Yuhas and Storer 1969), présente à ce jour une activité radioprotectrice satisfaisante. Toutefois, son utilisation reste uniquement limitée à certaines applications thérapeutiques en raison de ses effets secondaires importants (hypotension, nausées) aux doses thérapeutiques. De récents travaux conduits par l’équipe du Dr G. Rima de l’université Paul Sabatier de Toulouse en collaboration avec le service de santé des armées (B. Célariès et al, 2005 et B. Célariès et al 2006) ont néanmoins mené à la découverte d’une structure chimique de type phosphorothioate présentant une activité radioprotectrice équivalente à celle de l’éthyol, et ce, à une posologie trois fois moindre sur modèle murin. L’évaluation des capacités radioprotectrices, financée par la DGA et réalisée in vivo par l’IRBA, a également permis de révéler des effets secondaires moindres rendant particulièrement pertinente une éventuelle mise sur le marché de cette molécule. Trois phases de développement sont initialement prévues les deux prochaines années : 1) Elucidation des problèmes de reproductibilité ; amélioration de la solubilité. 2) Introduction de variations structurales : établissement d’une relation structure-activité. 3) Développement d’un procédé robuste de production de l’analogue le plus efficace. Aujourd’hui, la thrombolyse est le seul traitement efficace mais doit être administré dans les 4h30 suivant les symptômes et ne permet donc la prise en charge de seulement 10% des patients. Hormis la rééducation, il n’existe, à ce jour, aucun traitement efficace administrable dans les semaines suivant l’AVC (phase subaiguë). Dans ce contexte, la transplantation de cellules souches, encore appelée thérapie cellulaire, a émergé comme stratégie de réparation. Parmi les sources cellulaires, les cellules souches mésenchymateuses humaines (CSMh) issues de moelle osseuse ont montré une bonne tolérance et un bénéfice fonctionnel après injection à la phase aiguë de l’infarctus cérébral (IC) chez l’animal. Cependant, leur mécanisme d’action et leur effet après une administration à la phase subaiguë de l’IC restent mal connus. Parmi les hypothèses de mécanisme d’action des CSMh, celle d’un effet paracrine sur l’angiogenèse et la microvascularisation cérébrale semble être une piste prometteuse. En effet, après un IC, le remodelage microvasculaire joue un rôle prépondérant pour favoriser la neurogenèse endogène et la migration des neuroblastes propices à la récupération fonctionnelle. L’objectif de notre travail préclinique est donc d'évaluer si ces CSMh, injectées à la phase subaiguë de l'IC, modifient la microvascularisation cérébrale dans notre modèle transitoire d’ischémie chez le rat (oACM). L’identification de facteurs angiogéniques impliqués dans cet effet des CSMh permettra d’envisager une optimisation de la source thérapeutique. Nous avons réalisé un suivi IRM in vivo de la microvascularisation cérébrale (volume sanguin cérébral, diamètre des vaisseaux et densité microvasculaire) et une analyse des facteurs angiogéniques par biologie moléculaire (RT-PCR) durant 16 jours de suivi au sein de 2 groupes de rats ayant subi une ischémie: l’un traité par injection IV de CSMh à J8 (oACM CSMh, n=4), et l’autre par injection IV de PBS (oACM PBS, n=4). Une cinétique d’expression des facteurs angiogéniques a été réalisée à partir des cerveaux prélevés aux différents temps du suivi (H2, J1, J2, J3, J7, J9 et J16) par RT-PCR. Nos résultats préliminaires montrent que l’administration de CSMh, à la phase subaiguë de l’IC expérimental, favorise la formation de néo-vaisseaux et leur stabilisation précoce, augmentant ainsi la densité vasculaire. If not repaired, the abasic site may promote misincorporation of nucleotides in vitro and can be cytotoxic and mutagenic. The repair of abasic sites is thus essential to cell viability. We were interested in examining whether aminoglycosides (figure 1) used as antibiotic drugs could cleave DNA specifically at abasic sites. The cleavage of pBr322 plasmid DNA containing an average of 1.8 abasic sites per molecule was studied in the presence of various natural and synthetic aminoglycosides. The ratio of conversion from the supercoiled form to the nicked form was measured and the nicked plasmid fraction revealed the strong cleavage efficiency of most of them at abasic sites. The half maximal effective concentration (EC50) was determined for the most active aminoglycosides at pH 7.2 in the presence of 100 mM KCl (Figure 2). Among nine natural aminoglycosides studied and used for their antibiotic properties, the most efficient one was neomycin with a 1 M EC50. Clearly, natural and synthetic aminoglycosides appeared to be able to cleave strongly DNA at abasic sites. A rationalized method was developped for the design of a novel APCE mode dedicated to small molecules detection. The strategy was based on two properties of nucleic acid aptamers (Apt) used as MRE: (i) the conformational change triggered by the target complexation known as the structure-switching concept [3] and (ii) the ability of nucleic acids to form duplex structures with their complementary sequences (CS). The assay configuration, designed for multi-analyte sensing used an unmodified Apt and its fluorescein-labeled (LIF detection) and cholesteryl-tagged complementary strand (Chol-CS*). The separation of the cholesteryl-tagged species i.e. free Chol-CS* and the duplex formed with Apt (duplex*) was realized through the size-dependent shift of their electrophoretic mobility in the presence of a neutral micellar phase (MEKC) [4]. When the target was introduced in the pre-incubated sample, the hybridized form was destabilized resulting in the decrease in the duplex* peak area and the concomitant increase in the free Chol-CS* peak area. By introducing a poly(T) tail of an adequate length at one extremity of the Chol-CS* species, the electrophoretic separation of different Chol-CS* from each other and from their associated duplex* was successfully performed. Using a cell-based assay designed to probe for microtubule polymerization status, we identified azaindole derivatives as cell permeable microtubule depolymerizing agents. These compounds induce a G2/M cell cycle arrest and exert potent cytostatic effects on several cancer cell lines including a multidrug resistant cell line. In vitro experiments revealed that azaindole derivatives inhibit tubulin polymerization and bind to the colchicine binding site, strongly indicating that tubulin is the cellular target of azaindole derivatives. In vivo experiments, using a chicken chorioallantoid xenograft tumor assay, revealed that these compounds exhibit a potent anti-tumoral effect. This experiment, together with an assay that probes for the growth of vessels from spheroid of endothelial cells, shows that azaindoles derivatives also exhibit anti-angiogenic activities. Tumor cell motility is the hallmark of invasion and an essential step in metastasis. The identification of small molecule inhibitors that target key regulators of cell motility would help to understand this fundamental process and may lead to novel therapeutic opportunities. Using an original high throughput phenotypic screen, our team has discovered and characterized a specific inhibitor of LIM Kinase (LIMK), which was named LIMINIB (patent WO2010/000141). Phosphorylation of the actin binding protein cofilin by LIMK is the last step of the Rho-GTPase cascade that regulates actin dynamics. LIMK has been identified as an enzyme whose inhibition is supposed to stop the progression of invasive cancers. We have demonstrated that LIMINIB reversibly inhibits cofilin phosphorylation in vitro and in cells. LIMINIB blocks actin microfilament dynamics, inhibits cell motility and stabilizes microtubules. We have established that the microtubule stabilizing effect of the compound is the result of its inhibitory effect on LIMK and is independent of its effect on the actin cytoskeleton. The inhibitor is effective not only in cancerous cell lines but also in mice, where it delays tumors formation while showing a good tolerability. L'intégrité de la protéine encapsulée a été contrôlée en utilisant les deux méthodes complémentaires : la spectrométrie UV/Vis. (méthode classique de contrôle de la conformation 3D, reposant sur la localisation des bandes d’absorbance spécifiques + calculs des principaux rapports d'absorbance) et la conductimétrie via un biocapteur bi-enzymatique (méthode non conventionnelle basée sur la réaction de protéolyse combinée par 2 enzymes, protéinase K + pronase, en fonction de la conformation globale de la protéine). La conformation native de la Mb est maintenue dans des conditions standard, tandis que différents degrés de dénaturation de la protéine ont pu être observés en modifiant les conditions d'encapsulation. Il a été montré que l'élimination du solvant à pression réduite et, dans une moindre mesure, que l’emploi d'un PCL de masse moléculaire plus élevée ont conduit à l’altération de la Mb. Dans le 1er cas, l'intégrité perdue peut être attribuée au solvant résiduel rapidement piégé lors de la solidification accélérée des particules. Dans le 2ème cas, la dénaturation peut être expliquée par une augmentation du temps d'exposition de la Mb aux interfaces eau/solvant organique due notamment à une augmentation de la viscosité de la phase organique. Enfin les 2 méthodes de contrôle protéique ont donné des résultats comparables. Viral systems perform all these tasks and are far more efficient than current synthetic ones. Hence, mimicking the viral structure might help designing nanocarriers endowed with such capabilities. Herein, we report a two-step procedure for loading short interference ribonucleic acids (siRNA) within a polymer matrix in the nanometer size-range. At first, colloidal particles were prepared by complexing siRNA molecules with polyethylene imine in aqueous media. The pH, the ionic strength of the medium as well as several N/P ratios were studied among other parameters to find the right window where small, stable and positively charged complexes could be formed. In a second step, the electrostatic complexes were coated by an oppositely charged block copolymer containing both polysaccharide and polypeptide segments. This copolymer being amphiphilic by nature, the second step was achieved in presence of a water-miscible organic solvent. Hence, the final structure matched closely the morphology of viral particles with a core made of condensed nucleic acids and a shell of a copolymer based on saccharidic and peptidic units. Especially, it is believed that such a capsid of copolymer would prevent the dissociation of the PEI/siRNA complexes that are known to be less stable than PEI/DNA ones. The virus-like structure was evidenced by fluorescence microscopy, light scattering and microelectrophoresis. In vitro cell transfection studies were performed on HeLa cells using the luciferase assay. First results showed high transfection of the nanocarriers but low gene silencing. La haute résolution de la cristallographie des rayons X permet une analyse détaillée de l’interaction des drogues avec des différents oligonucleotides, aussi que de l’influence du solvant dans ces interactions [1]. Les rayons X ne sont pourtant presque pas sensibles aux atomes d’hydrogène et ne fournissent qu’une image statique d’une structure possible dans des conditions spécifiques. Des informations complémentaires sont essentielles pour un design raisonnable des nouvelles drogues de chimiothérapie dans le cadre d’un mécanisme d’activité qui dépend des nombreux facteurs comme l’acidité des protons [2], la cinétique des réactions, la stœchiométrie et la stabilité des complexes, etc. Ces informations sont pourtant à la portée d’autres techniques comme la Calorimétrie isotherme à titration (ITC), la Cristallographie de Neutrons ou la spectroscopie RMN. La détermination expérimentale de la position précise des atomes d’hydrogène ou deutérium dans des structures cristallines de macromolécules biologiques est indispensable pour la compréhension de la relation structure-activité. La complémentarité des techniques exploitées sera illustrée par notre expérience avec des études des ligands à possible activité anticancéreuse (des dérivés d’acridines et de phenazines produits à Auckland Cancer Research Center). Il s’agit des stratégies d’autant plus importantes pour les médicaments qui doivent cibler des structures très spécifiques dans des environnements complexes : on soulignera l’importance de l’utilisation intégrée des méthodes pour la validation des résultats et la maitrise des propriétés. Disrupting interaction between PDZ-containing protein PSD-95, and its natural ligand 5-HT2A receptor, was found to reduce hyperalgesia in a rodent model of neuropathic pain. Thus, inhibiting this interaction could lead to the development of a novel class of analgesic agents. Starting from a hit previously identified by our laboratory, we carried out a structure activity relationship (SAR) study. 7 indole-based compounds were synthesized in 3-5 steps with good overall yields (50-85%). These molecules were evaluated by NMR as ligands of the PDZ1 domain of PSD95. Three of these compounds were found able to interact with the targeted protein during NMR. One of the new compounds showed an improvement of the binding affinity compared to the hit compound and could be considered as a lead molecule for the development of new series of PDZ ligands. In a preclinical study we already compared the quality of tumor removal of peritoneal carcinomatosis (PC), performed under “conventional” light, versus near-infrared image guided (NIR) surgery using a tumor targeting probe (RAFT(c[-RGDfK-])4-Alexa Fluor® 700) and a portable clinical imaging device ("Fluobeam"). The results showed that NIR image guided surgery using RAFT-RGD and the "Fluobeam" greatly improved the quality of the surgery of PC by doubling the number of detected nodules and reducing significantly the duration of the surgery. The aim of the first project is the transfer of this technology, consisting of the RAFTRGD-probe and the "Fluobeam", into a clinical veterinary phase as an evaluation regarding clinical surgery in human, that is to say the clinical surgery of spontaneous tumors in domestic animals. The second project is the chemical design of an innovative fluorescent molecule for the diagnosis and the therapy of cancer. This molecule is based on the RAFT-c(RGD)4, targeting the Integrin αvβ3. By combining this with a cleavable cassette for tumor-overexpressed enzymes, the tumor specificity should increases. The principle is that the fluorescence of the molecular complex is quenched while remaining intact and it will be turn on as soon as the peptide bond of the cleavage cassette is cut by the enzyme. Les inhibiteurs d’histone déacétylase (iHDAC) tiennent une place importante dans les traitements ciblant les mécanismes épigénétiques, à savoir une action qui perturbe le niveau d’acétylation post-traductionnelle des résidus lysines localisés à l’extrémité des histones. Les premiers médicaments dont le mode d’action se réfère à l’inhibition de la déacétylation des histones sont apparus depuis quelques années seulement dans le domaine de la cancérologie principalement. Très récemment, des chercheurs du CNRS, de l’Université Joseph Fourier et du CHU de Grenoble ont montré qu’un produit naturel de type cyclotétrapeptide, le FR235222 (Figure 1), agit efficacement contre différent parasites apicomplexes, bloquant leur croissance et perturbant leur différenciation. Cette étude a révélé notamment que cette molécule induit la différentiation de T. gondii, de la forme réplicative (tachyzoïte) vers la forme kystogénique (bradyzoïte).[1] T. gondii HDAC3 (TgHDAC3), une enzyme déacétylase de classe I, a été identifiée comme étant la cible de FR235222. Des études ex vivo ont également montré que la différenciation du bradyzoïte au tachyzoïte, est abolie par FR235222, élargissant ainsi le champ d’application de cette molécule pour des traitements visant aussi bien des phases aigües que chroniques de la toxoplasmose.[2] Figure 1 A partir de la structure de FR235222, une série de nouveaux analogues cyclotétrapeptides a été synthétisée en peu d’étape à partir d’un intermédiaire commun et en faisant varier les caractéristiques structurales importantes (voir Figure 1) permettant de conserver l’efficacité en tant qu’iHDAC tout en améliorant l’index de sélectivité. Cependant son application clinique est limitée par sa faible solubilité en milieu aqueux (0,064 mg.mL-1 à 25°C) et sa faible biodisponibilité par voie orale [1]. La mise en œuvre de cyclodextrines amphiphiles bioestérifiées dont la synthèse est maîtrisée dans notre laboratoire [2,3] et d’esters de polyéthylène glycol, permet de viser deux objectifs : i) d’une part d’améliorer la mise en solution de l’ART à travers son association aux vecteurs colloïdaux de cyclodextrines (CDs) modifiées, ii) d’autre part d’améliorer la biodisponibilité de l’ART et son efficacité thérapeutique à travers l’hydrophilisation de la surface des vecteurs pour augmenter leur résidence vasculaire en évitant une capture trop rapide par les organes du système des phagocytes mononuclées [4], suite à leur administration intraveineuse. Dans ce travail, les CDs bioestérifiées ont été obtenues par transestérification de la γ-CD en présence de décanoate de vinyle. La réaction est catalysée par la thermolysine [5]. Les dérivés obtenus (γ-CDC10), principalement caractérisés par spectrométrie de masse MALDI/MS, conduisent par la méthode de nanoprécipitation, à des systèmes submicroniques de forme sphéroïdale de type nanosphères ou nanoréservoirs, de tailles monodisperses de l’ordre de 70 et 220 nm, respectivement. Ces nanostructures d’ester de γ-CDC10 prennent en charge des quantités appréciables d’ART avec un taux de recouvrement de l’ordre de 95 %, permettant ainsi d’envisager une administration par voie parentérale. A titre d’exemple, la concentration de l’ART dans les suspensions aqueuses avoisine 0,4 mg. mL-1, soit une augmentation de la dose d’ART mise en solution en milieu aqueux d’un facteur 6,25 pour le cas des nanosphères. Dans le cas des nanoréservoirs, les concentrations sont de l’ordre de 1,6 mg.mL-1, soit une augmentation d’un facteur 25. Les suspensions colloïdales d’ART ont été stabilisées avec du stéarate de PEG 4000 ou 1500. Cette couronne hydrophile, qui protège les particules de la floculation par répulsion stérique, devrait également permettre de les rendre furtives. Toutes les suspensions colloïdales chargées d’ART sont actives in vitro sur des souches chloroquino-résistantes (K1) et chloroquino-sensibles (3D7) de Plasmodium falciparum. Les CI50, de l’ordre de 3 à 6 ng.mL- 1 déterminées après 72h d’incubation des nanosystèmes dans le milieu de culture, sont comparables à celle d’une solution d’ART en solvant non aqueux déterminées dans les mêmes conditions. Ainsi, en accord avec les études in vitro de dissolution, l’ART est relarguée à partir de son support nanoparticulaire et peut alors exercer son activité antiparasitaire. Enfin, l’évaluation de la sécurité d’emploi et de la furtivité des nanovecteurs (tests in vitro et in vivo) est en cours d’évaluation. (Sterculiaceae) is tropical herbaceous used in traditional pharmacopeia at Burkina Faso for treatment of respiratory diseases such as cough and asthma. This study aims to highlight the effect of plant on enzymes activity implied in smooth muscle contraction (PDE4A1α) or inflammatory reaction (PLA2 and 5-lox I-B). Methods: Fractions were prepared from hydro-alcoholic extract of roots using dichloromethane, nhexane and ethyl acetate. Fluorimetric and photometric methods were used to measure enzymatic activity in presence of the hydro-alcoholic extract and its fractions. Results: Significant inhibitory effect of Waltheria indica L. on activity of three enzymes was compared to aminophylline or indomethacin. Ethyl acetate (F3) and residual (F4) fractions whose effect are similar to initial hydro-alcoholic extract (HA), are more effective than n- hexane (F1) and dichloromethane (F2) fractions. For PLA2 and 5-lox I-B, F3 and F4 fractions induced a 50% inhibition at the concentration of 0.05 mg/mL and a 100% inhibition at the concentration of 0.5 mg/mL. Their effect on PDE4A1α activity is less only of 40% and seems not to be dosedependent. Conclusion: the present study demonstrates that Waltheria indica L. inhibited the activity of three enzymes implied in smooth muscle contraction confirming its potential interest in respiratory diseases treatment. Since F3 and F4 fractions are more potent than F1 and F2 ones, additional studies will be performed using NMR and mass spectrometry for compounds identification. Isometric contraction effects will be studied on tracheal, extrapulmonary and intrapulmonary bronchial rings for better pharmacological characterization. In the family of the aminoglycosides, neomycin B has also appeared to be able to strongly inhibit the interaction between the HIV-1 TAR RNA and the transactivating protein Tat.2 However, aminoglycosides have no real antiviral activity. In the search for new antiviral agents, a small aminoglycoside, neamine, which corresponds in neomycin B to a crucial recognition element for TAR RNA interaction, has been modified selectively on different hydroxyl functions. Several derivatives able to strongly inhibit the TAR-Tat binding with submicromolar concentrations were identified using FRET experiments.3 Affinity constants for TAR of the inhibiting derivatives also were measured by SPR and microcalorimetry (ITC). Due to the rapid emergence of resistant bacterial strains and the toxicity of certain antibiotics, it is imperative to discover and develop novel antimicrobial drugs. In the search for new antibiotics agents, neamine derivatives were prepared through modification of the amino or the hydroxyl functions in order to increase the affinity for rRNA and prevent the activity of aminoglycoside-modifying enzymes. For this purpose, it is necessary to develop short and regioselective methodologies to introduce for example aromatic rings able to improve the affinity for the RNA target, through groove binding and/or stacking1. NH 2 O HO O 3' NH2 H2N O HO NH2 6 O neamine derivative The antibiotic activities of different neamine derivatives were measured on a wild type and resistant cells. Neamine carrying two or three naphthylmethylene groups in position 3’, 4’ and/or 6 were found to be interesting antibiotic agents against a broad range of Gram (+) and (-) strains surexpressing resistance to aminoglycosides2.
