(key words) Candidat pressenti : OUI NON Contenu scientifique du programme de la thèse (en anglais) EPEC is a major cause of infantile diarrhea in developing countries. To induce the disease, EPEC uses a type III secretion system to inject bacterial effectors into host cells. As for all type III secretion system, injection requires the prior insertion of a pore-forming complex, the translocon, into target cell membranes. In previous works, we have shown that the pore-forming activity of the EPEC translocon is regulated by the serine protease activity of the EspC autotransporter on the EspD and EspA components. While EspC prevents cytotoxicity through the control of the translocon-mediated pore-forming activity during the early stages of bacterial infection, EspC enhances cytotoxicity at later stages of infection, an activity that has been linked to its T3SS-dependent intracellular delivery. The PhD work will build on preliminary works in the laboratory to provide with a structural characterization of the EPEC translocon, and potential conformers that are targeted by EspC. Mutagenic analysis will be performed in complementation to the structural characterization, to define the mechanism of the recognition of translocon components by EspC. The results will have general implications on the mechanism of type III secretion, and potentially provide insights into the mechanism of EspC delivery into host cells following its interaction with the T3SS translocon. Further extension of the work includes the validation of these results in cellular infection models, as well as in vivo, using the related mice enteropathogen Citrobacter Rhodentium. The structural characterization will be performed using high-resolution cryo-electron microscopy.
