In large vertebrate genomes about 50 000 starting points (replication origins) are activated during S phase. A spatiotemporal program controls both origin positions and their timing of firing but their regulation remains largely unknown. The PhD project relies on both genetic and genome-wide studies to understand molecular events involved in this regulation crucial for genomic integrity. These motifs have the capacity to be structured into G-quadruplex (G4). Deletions of the G-rich motif in two model origins showed their important function in origin activation in the DT40 avian cell line. Point mutations inside the G-rich motif of one model origin have demonstrated the importance of forming this structured G4 for replication initiation. We also showed that a G-rich motif is not sufficient to induce an origin of replication and identified a 200 bp cooperating cis-module on one model origin. The aim of this PhD project is to refine our vision of molecular mechanisms involved in origin function. Firstly, we plan to delineate more precisely on our two model origins the minimal size and sequence motif of the cooperating cis-module. We will define how these cis-elements might influence locally chromatin organization during the cell cycle. Based on this new information, we will search for enrichment of the new identified signal(s) inside replication origins genome-wide. We will also develop a new origin mapping method that will delineate very early firing origins in the avian DT40 cells and in human cells. We will determine common genetic features of these specific origins in order to decipher new potential regulatory pathways involved in the control of early firing. Proteomic analyses will be performed to identify specific and new trans-factors associated with these early replicated origins. Altogether this work will provide new key information on the regulation of the spatiotemporal program of DNA replication.
