En utilisant Phelipanche ramosa comme modèle, nous avons démontré que les graines nécessitent une période de conditionnement d’au moins quatre jours avant de pouvoir germer en réponse à un stimulant artificiel, le GR24. Par une approche transcriptomique sans à priori (cDNA-AFLP), 58 fragments dérivés de transcrits (TDF), présentant une modification de leur profil d’expression suite à un traitement au GR24, ont été isolés. Parmi ces TDFs, deux transcrits surexprimés correspondent à un gène, PrCYP707A1, codant pour une ABA 8-hydroxylase, enzyme de catabolisme de l’acide abscissique (ABA). Grâce à des expériences de RACE PCR, deux ADNc pleine longueur, PrCYP707A1 et PrCYP707A2, ont été obtenus. Alors que ces deux gènes présentent un niveau d’expression relativement bas durant la période de conditionnement, une première baisse de la teneur en ABA est observée, celle-ci étant essentiellement attribuée à une diffusion hors de la graine. Après le conditionnement, l’application de GR24 provoque une forte surexpression de PrCYP707A1 durant 18 heures, suivie d’une seconde chute de la teneur en ABA, détectable après 3 jours de traitement. Or, un traitement par un inhibiteur spécifique des CYP707A, l’Abz-E2B, conjointement au traitement GR24, entraine une inhibition de la germination des graines. Ces différents résultats montrent que la germination s’effectue après une levée de dormance des graines de P. ramosavia le catabolisme de l’ABA médiée par une activation du gène PrCYP707A1 par le stimulant de germination. Des expériences d’hybridation in situ, sur des graines de P. ramosa ont par ailleurs montré que le GR24 induit une accumulation d’ARNm PrCYP707A1 au niveau des cellules du périsperme situées sous le micropyle. Ces résultats, ainsi que ceux d’une autre étude, suggèrent que les récepteurs aux stimulants de germination seraient localisés dans ces cellules du périsperme des graines. Le pois chiche et la fève occupent 86% des superficies des légumineuses. Au cours de ces dernières décennies, les aires occupées par ces légumineuses diminuent de 30% et l’orobanche est parmi les principaux facteurs qui causent ces pertes. Les moyens employés pour la lutte contre ce fléau sont relativement diversifiés mais restent inefficaces ou à effets indésirables. Le criblage de génotypes résistant/tolérants reste la méthode la plus adaptée sauf que la résistance est rare chez les variétés disponibles aux agriculteurs. La production artificielle des cultivars résistants/tolérants utilisés dans un programme de lutte intégrée pourrait être efficace contre ce fléau qui limite les cultures des légumineuses. Dans ce cadre, des rayonnements ionisants ont été utilisées pour générer des mutants de pois chiche résistants à Orobanche foetida. Les mutants de pois chiche les plus résistants à l’O. foetida ont été sélectionnés au champ infesté à partir de 3000 graines irradiées, réparties sur 3 stations différentes au nordouest de la Tunisie (Béja). Une étude des mécanismes physiologique de la résistance à l’Orobanche chez les mutants sélectionnés a été effectuée au laboratoire en utilisant les systèmes de co-culture et de récolte d’exsudats racinaire. Ces 2 systèmes nous ont permis de sélectionner 6 mutants de pois chiche résistants au parasite. Cette résistance est caractérisée par un faible taux de germination des graines d’orobanche (<50% en coculture et <25% avec les exsudats racinaires) et un nombre réduit d’accrochage par racine allant de 0 à 8 accrochages/ 2plantes. Il serait intéressant de compléter cette étude par une caractérisation moléculaire de la résistance chez les mutants sélectionnés. Using LC-PDA-MSn we have developed a specific protocol allowing the simultaneous and qualitative detection of 4-phenyl and 4-propylcoumarins in DCM bark, fruit and leaf extracts obtained from Mammea neurophylla. By comparison of their retention times, MS and UV data with that of authentic samples, nine, seven and five 4-phenylcoumarins could be directly identified in bark, leaf and fruit extracts respectively. On the other hand, interlocking UV spectra and ESI–MSn data analysis allowed us to deduce plausible structures of five, eight and four other coumarins in bark, leaf and fruit respectively by comparison with their reported spectral data. During this study new Mammea A/AA 9-hydroxy-cyclo F and Mammea A/AB 9-hydroxy-cyclo F were identified. In order to confirm our hypothesis, those new compounds were purified using flash chromatography and preparative HPLC. We also believe that this protocol will be useful in case of dereplicative studies of Mammea and related species. Ils sont une composante des pathologies neurologiques comme la maladie d’Alzheimer, et du vieillissement des tissus2,3. Ainsi des molécules capables d’inhiber leur formation ou de permettre leur élimination peuvent constituer des molécules d’intérêt thérapeutique. Ces AGEs sont issus d’une réaction de condensation entre un sucre et une amine primaire qui aboutit à la formation d’hétérocycles (Schéma 1). AGEs Schéma 1 : Formation des AGEs Il est possible d’empêcher l’accumulation dans l’organisme de protéines modifié par ces réactions de deux façons différentes. Soit par des molécules inhibant la formation des produits de Maillard, soit en détruisant certains intermédiaires. Cette deuxième catégorie cible plus particulièrement les intermédiaires dicarbonylés. Cependant, seule une famille de composée est décrite comme ayant cette capacité. Ce sont des analogues de l’alagébrium4,5,6,7. Afin d’identifier un plus grand nombre de breakers, un test a été mis en place permettant l’identification de ce type de molécule. Ce test est basé sur la capacité des rupteurs d’AGEs à rompre les liaisons dicarbonylés et la détection d’un composé fluorescent libéré au cours de la réaction8. Bien qu’elles ne représentent qu’environ 1% des protéines totales, PIN-a et PIN-b sont les principaux déterminants de la texture du grain et influenceraient le degré d’adhésion entre la matrice protéique et les grains d’amidon. Plus récemment, il a été observé que la présence de puroindolines pourrait jouer un rôle sur l’agrégation des protéines de réserve1. Mais leur rôle biologique demeure cependant inconnu. Nous avons construit des lignées surexprimant la PIN-a d’une part et la PIN-b d’autre part afin d’accroitre le stress oxydant et le stress du réticulum endoplasmique (RE) et d’en déduire les mécanismes liés à la synthèse massive des protéines de réserve dans les cellules de l’albumen de blé en développement. Contrairement au transgène Pin-a+, l’expression de Pin-b+ en T3 semble subir un contrôle plus strict et ne concerne que 2 lignées, suggérant des effets plus ou délétères du gène Pin-b à forte dose. De manière inattendue, les effets du stress oxydant au niveau des protéines et lipides semblent limités rendant la quantification des marqueurs difficile. Il se pourrait que les cellules des transformants mettent en place un système de détoxication efficace, ce qui limiterait les dommages causés par les espèces réactives de l’oxygène. La surexpression semble, tout de même, corrélée de manière significative à la texture du grain mature et à la teneur en protéines de réserve polymériques. JORISSEN LUNAM Université, Université d’Angers UMR CNRS 6112 LPGN-BIAF - Laboratoire des Bio-Indicateurs Actuels et Fossiles, 2 Boulevard Lavoisier, 49045 Angers Cedex, France The present study is a faunal analysis on the spatial and temporal variability of benthic foraminifera community in the Grevelingenmeer at three different seasons (in May, August and November 2011). Our faunal analyses indicate that abundant live foraminiferal faunas occur at stations 2 and 4 (water depth respectively 23 and 17 m) in August and November 2011 where oxygen conditions are not limiting. At station 1 (30-34 m), which is subject to seasonal anoxia, poor foraminiferal faunas were observed in May, August and November 2011. We suspect that these scarce foraminifera have been transported by currents into the deepest part of the channel. Both at stations 2 and 4, the faunas are almost exclusively composed of three species: Ammonia cf. A. batava, Elphidium ex. gr. E. excavatum and Trochammina cf. T. inflata. No major difference in species composition is observed between the two stations, although the faunas of station 4 are about two times richer than those of station 2. Most of the species living in the Grevelingenmeer are characterised by a morphology which differs from that of the typical species. Especially in Ammonia cf. A. batava, the pore density and pore size appears to be much higher than in typical A. batava. Additional analyses will be performed to find out whether this is a morphological adaptation to low oxygen concentrations. Keywords: benthic ecology, anoxia, Adriatique Sea, denitrification Session environnement : Oral 7 ANALYSE ET MODÉLISATION DES ÉVOLUTIONS A LONG TERME DE LA BIODIVERSITÉ PHYTOPLANCTONIQUE DANS LES ZONES CÔTIÈRES SOUS L’EFFET DES PRESSIONS ENVIRONNEMENTALES ET ANTHROPIQUES Tania HERNÀNDEZ FARIÑAS Laboratoire DYNECO/VIGIES Centre Ifremer de Nantes Le phytoplancton constitue un lien essentiel au sein des écosystèmes marins, de part son rôle de producteur primaire et de son importance dans le transfert de l’énergie à travers le réseau trophique. Du fait du contexte actuel de changements globaux et de l’influence du phytoplancton sur le fonctionnement des écosystèmes marins, il est essentiel d'étudier les facteurs intervenant et affectant la dynamique des communautés phytoplanctoniques. L’étude de la diversité phytoplanctonique est l’un des sujets les plus complexes du fait du nombre important d’espèces ainsi que de leur court cycle de vie. Les données analysées proviennent du Réseau de Surveillance du Phytoplancton et des Phycotoxines (REPHY), géré et mise en œuvre depuis 1983 par l'IFREMER (Institut français pour l'exploitation de la mer). Dans un premier temps, l'étude a consisté à déterminer la collection de données à analyser. Pour cela, les nombreuses séries temporelles disponibles ont été décrites sur la base de plusieurs critères dont la fréquence d’échantillonnage, le comptage d’espèces et la disponibilité des données environnementales. En parallèle, des niveaux d’agrégation de données doivent être définis en considérant les notions d’événements, d’espèces indicatrices, de groupes taxonomiques et fonctionnels. Des méthodes multi-variées ont été utilisées pour l'étude de la variabilité spatio-temporelle des peuplements phytoplanctoniques. Les résultats montrent une variabilité temporelle significative de la composition spécifique des communautés dans la Manche Orientale. Néanmoins, la difficulté de l’analyse est de déterminer les causes de cette variabilité, notamment l’influence potentielle des changements d’expert taxinomiste au cours du temps, l’une des problématiques de l’analyse des séries temporelles. TAILLEBOIS Laboratoire RCIM (Récepteurs et Canaux Ioniques Membranaires) UFR Sciences Angers, France Les pucerons sont responsables de centaines de millions d’euros de pertes annuelles dans les cultures. Le contrôle de ces ravageurs passe essentiellement par la lutte chimique avec une utilisation croissante des insecticides néonicotinoïdes. Cependant l’impact des insecticides sur les espèces non cible telles les abeilles, la pollution de l’environnement liée à leur rémanence ainsi que l’apparition de résistance chez les ravageurs nécessitent la mise en place de méthodes de lutte raisonnées. Il apparait donc crucial de mieux comprendre le mode d’action de ces molécules afin de réduire leur utilisation. L’objectif de cette étude est d’identifier les mécanismes de toxicité d’insecticides néonicotinoïdes chez le puceron du pois et de déterminer les caractéristiques moléculaires de leur cible : les récepteurs nicotiniques. Les tests d’intoxication orale et les tests de liaison sur les récepteurs nicotiniques montrent que le thiamethoxame est plus toxique (DL50=0,77µg/ml) que l’imidaclopride et la clothianidine (DL50=1,13 et 3,96µg/ml respectivement) tandis que seule l’imidaclopride présente une faible affinité pour les récepteurs nicotiniques. Ces données montrent que le thiamethoxame agit réellement comme un agoniste des récepteurs nicotiniques contrairement à ce qui est connu chez d’autres insectes2. Les récepteurs nicotiniques sont composés de cinq sous-unités, dont la nature influence les propriétés pharmacologiques. Bien que onze sous-unités aient été identifiées chez le puceron du pois3, l’analyse quantitative montre que les sous-unités α3, α7, α10 et β2 sont majoritairement exprimées et pourraient potentiellement être impliquées dans la formation des sites de liaison. En effet, la dynamique des océans est surtout animée par la circulation atmosphérique et par le réchauffement différentiel de notre planète. Les circulations atmosphériques et océaniques sont responsables de la répartition de la chaleur sur la surface de la terre et donc du climat. A certains endroits clés, les différences de température et/ou de salinité des masses d’eau entrainent un plongement des eaux. Ce mécanisme, régisseur de la circulation profonde, permet d’alimenter l’océan profond en oxygène et par conséquent le développement de la vie sur les fonds océaniques. En étudiant les faunes benthiques préservées dans les archives sédimentaires, on peut reconstituer la ventilation du fond marin dans le passé, et donc l’intensité des courants marins. Cette compréhension est essentielle pour notre connaissance des interactions entre le climat et la circulation océanique. Dans ce contexte, les faunes actuelles de foraminifères benthiques de la Mer d’Arabie sont étudiées le long d’un fort gradient d’oxygénation, mais également en relation avec la température, la salinité et la productivité primaire. Dix stations ont été prélevées le long de la ride de Murray, au Nord de la Mer d’Arabie. Ces stations constituent un transect d’oxygénation, de condition hypoxique à presque oxique (de 2µM à 78 µM). Les foraminifères sont majoritairement concentrés dans le premier centimètre du sédiment. La distribution des faunes actuelles benthiques semble répondre au changement qualitatif et quantitatif de la matière organique et de la concentration en oxygène. Cette étude servira de cadre de référence à l’interprétation d’archives sédimentaires Holocène et Quaternaire. A cause de difficultés méthodologiques et depuis les travaux d'Altenbach en 19851, aucune tentative de mesure de biomasse n'a été entreprise sur ces organismes. Dans cette étude, une méthode développée par Smith et al.2 basée sur la nano-spectrophotométrie a été adaptée afin de mesurer la biomasse des foraminifères. Cette méthode permet de mesurer le contenu protéique des foraminifères individuellement. La coquille calcitique de ces organismes rend inaccessible la majorité du cytoplasme de l'individu, et le rendement est donc très faible. Le moyen le plus simple d'appliquer cette méthode aux foraminifères est d'écraser le test, empêchant donc par la suite toute analyse morphométrique. Au cours de la thèse, une série d'expériences a été mise au point afin de tester différentes techniques afin de briser la membrane cytoplasmique et permettre un rendement maximum de la méthode, tout en préservant le test pour des mesures morphométriques et de masse de calcite. La nouvelle méthode est rapide, et facile à mettre en place, et les données sont reproductibles. Cette méthode a été appliquée sur des foraminifères planctoniques vivants prélevés au cours de trois missions en mer dans les Açores, le Golfe de Gascogne et les eaux Pacifiques Japonaises. Ces données ont ensuite permis d'estimer la biomasse des foraminifères en fonction de la taille, et, associées à des données faunistiques (abondances et répartition verticale), ont permis de faire une première estimation de la biomasse globale des foraminifères planctoniques actuels dans les eaux de surface océaniques. Les foraminifères planctoniques, à coquille en Carbonate de Calcium (CaCO3) bien fossilisable, sont responsables d’une partie importante du flux de carbonates dans l’océan ouvert1 et participent ainsi à la pompe biologique du carbone. L’objectif de cette thèse est d’évaluer la contribution des foraminifères planctoniques à la pompe biologique du Carbone (taux CCaCO3 coquille / Corg cytoplasmique) aujourd’hui et dans le passé, en ciblant une zone océanique réputée être un puits de carbone : l’Océan Austral2. La lagune du bassin d’Arcachon est recouverte en grande partie par l’herbier de Zostera noltii qui, de part sa dynamique saisonnière, accroît alors l’hétérogénéité latérale. Cette étude présente les résultats préliminaires de nouvelles techniques d’analyse utilisées dans le but d’améliorer nos connaissances sur les relations entre les systèmes racinaires des herbiers de Zostera noltii et la méiofaune tel que les foraminifères. Pour cela nous tentons i) de documenter à une échelle submillimétrique et en 2-dimensions des espèces redox majeures (e.g. H2S, Fe(II), PO4, NO3, etc.), dans les eaux interstitielles par l’utilisation combinée de DGT-DET et d’imagerie hyperspectrale, et dans le solide par microfluorescence X ; et ii) de décrire les espèces de foraminifères dans ces environnements en s’intéressant aux individus vivants par l’emploi d’un fluorochrone, le Cell Tracker Green. De plus, des mesures du métabolisme aérobie (taux de respiration) et anaérobie (taux de dénitrification) des espèces tolérantes à l’anoxie sont réalisées afin d’estimer la contribution de ces micro-organismes aux processus de la diagénèse précoce. A ce jour, les gels DET 2D montrent une large hétérogénéité des couches redox influencées par les activités de bioturbation des organismes et par le système racinaire. Les densités de foraminifères semblent également modifiées par la présence du système racinaire qui augmente la pénétration de l’oxygène et permet aux organismes aérobies de vivre plus en profondeur dans le sédiment. Les foraminifères sont capables de respirer l’oxygène et certains les nitrates tout comme les bactéries. Il semble donc que la présence de microenvironnements riches en nitrates, résultant du relargage d’oxygène par les racines, pourrait favoriser une distribution particulière des foraminifères. Elle est cultivée dans des fioles d’Erlenmeyer de 500 mL contenant un volume de 250 mL d'eau de mer artificielle. Ces fioles ne sont ni aérées ni agitées (culture en Batch). Elles sont soumises à un éclairement de 300 µmol de photons m-2 s-1, à une température de 24 ±1 °C et une photopériode de 14 h dans une salle de culture. Les éléments transposables (ET) sont de courtes séquences d’ADN ayant la capacité de se déplacer dans un chromosome ou même d’un chromosome à l’autre, au sein du génome qui les héberge. La première classification des ET repose sur le mécanisme de transposition. Les rétrotransposons sont des ET se déplaçant via un intermédiaire ARN (mode dit «copier-coller»), contrairement aux transposons qui se déplacent sans intermédiaire (mode dit «couper-coller»).L’élément mariner est un transposon qui a été découvert chez la drosophile Drosophila mauritiana. Des ET de type mariner (Mariner-Like-Element ou MLE) sont présents chez les animaux et les végétaux. Ils sont composés d’un gène de la transposase encadré par des séquences ITR (Inverted Terminal Repeat) et UTR (Untranslated Region). L'élément transposable de type MLE a bien été détecté chez A. acutiuscula. La présence d'un OFR (Open Reading Frame) complet pour le gène de la transposase a été mise en évidence dans son génome par la technique de PCR inverse. Dans la littérature, de nombreux exemples d'activation des ET en conditions de stress biotiques et abiotiques ont été décrits. Ainsi, nous avons recherché l'expression de MLE avant et après l'application de stress thermiques à 16 et 32 °C pendant 30 min, 2 h, 5 h et 8 h. L'expression des MLE a été détectée après avoir placé les microalgues pendant 5 h à 16 °C. Few studies are available on the evolution of bread during mastication1. The objective of this study was to develop a robust rheological method, using extrusion (capillary rheometry) to assess the breakdown of bread during chewing and to characterize the physical properties of bread bolus by its rheological behaviour, moisture content and particle size. Two breads of different texture were chewed for different durations by a healthy subject, until swallowing. An apparent viscosity of the bolus was determined by capillary rheometry, whereas its viscoelastic behaviour was determined by an oscillatory test on a rheometer equipped with a vane geometry. An imaging method was set up in order to determine the distribution of bread particle size after chewing. For both breads, the apparent viscosity of bread boluses followed a power law with a consistency index decreasing with chewing time; this consistency index reached similar values for the two breads before deglutition, about 10 times lower than bread initial consistency; same trend was obtained for viscoelastic moduli2. Meanwhile, the moisture content increased significantly, and mastication reduced the bread into an increasing number of small particles. The results suggested that plasticization of bread by water from saliva had more effect on the rheological behaviour of the bolus than its fragmentation. This approach could be used to study the oral destructuration of breads in relation to their sensory and nutritional properties, and to evaluate the influence of stimuli release and perception as a function of the texture of the product. The structure of pectin is based on a backbone of polygalacturonic acid, which is called homogalacturonan (HG) and segments consisting of alternating sequences of L-rhamnosyl and D-galacturonosyl residues ramified with side chains of arabinans, arabinogalactans and galactans. Pectin exhibit functional properties like gelling, thickening, and emulsifying that are widely used in food industry. Commercial pectin is almost exclusively derived from citrus peel. The project aims to find the variation of pectin components depending on plant sources and processing in relation with functional properties. In the current study orange, lemon, lime and grape fruits fresh and dry peels were used as experimental materials. The peels were subjected to gentle HNO3 extraction. Beside chemical composition assessment, functional SAG and YOG tests were applied in order to test gelling strength and stabilizing power of the samples. All pectic samples were rich in galacturonic acid (>80% w/w) and contained only small amounts of sugars (around 10% w/w). Arabinose, galactose and rhamnose were found to be the major neutral sugars. This fact confirms that HGs, the key pectic domains for gelling properties, are predominant in the citrus pectins studied. In pectins extracted from fresh peels, intrinsic viscosity and degree of esterification were slightly higher than in pectins extracted from dry peels. This might be related to the activity of some degradation enzymes during the drying process. Moreover, our findings support that pectins extracted from orange and grape fruit dry peels exhibit comparable gelling strengths. Pectins extracted from lemon and orange dry peels can be used as stabilizer in yoghurt drinks. Pectins extracted from grape fruit and orange fresh peels could be applicable in acidified yoghurt drinks. However, bakers practices - from wheat culture to bread - and their effects on bread quality are not enough documented. Moreover, SD yeast and lactic acid bacteria (LAB), constitute a “reservoir” for inter-specific and intra-specific diversity studies1,2. The aim of this project is i) to investigate the diversity of the traditional SD breadmaking practices and ii) to study the SD microbial dynamic and diversity during SD bread-making. This study is carried on with a network of French traditional organic bakers and farmers-bakers (RSP). Cultural diversity associated with bread-making was studied thanks to a semi-structured survey with 14 bakers. Microbial diversity was analyzed for five bakers with sampling and enumeration at different steps: “levain chef”, “levain tout point”, after kneading, before baking. Yeast and LAB isolates were identified by molecular methods (ARN 26S and ARN 16S gene sequencing, respectively). A Temporal Temperature Gel Electrophoresis (TTGE) approach had also been used to monitor the LAB dynamic. A high diversity in bread-making methods was found. The origin, conservation and age of the starter, the way starter and dough are fed, sourdough and dough consistencies differed. Yeast diversity was also important and species changed between bakers and during the bread-making process. In some cases, stable kinetics with a major species (Candida humilis or Kasachstania bulderi) and minor species (Candida carpophila or Hyphopichia pseudoburtonii) were found. In other cases, the dominant species changed over time. Concerning LAB diversity, no major variation was observed during SD breadmaking steps. Low interspecific diversity was detected between the different SD and Lactobacillus sanfranciscensis appeared as the major or dominant species in all SD. Future research prospects will be discussed concerning the metagenomic approach and the links between biodiversity versus bread-making practices. Paris, France. Starch is one of the main component of superior plants, as it can represent between 50% (corn, bean) and 90% (potato, manioc) of their dry-mass. This polysaccharide can be extracted as a flour and then processed into a massif material using techniques such as casting or extrusion. Previous studies have demonstrated that extruded starch has a shape-memory property, and can partially crystallize, which enhance the mechanical property1. Hence, it can be used for interesting applications in the industrial field2. Because of the fast degradation rate, extruded starch could be used as biodegradable medical devices with or without shape memory properties. This application needs knowledge of the material properties in a cell and tissue environments. The aim of this study is to fully characterize extruded starches from different botanical origins (potato, peas and corn) in vitro and in vivo. Scaffolds characterizations were performed (transformation analysis (DSC), water content (TGA), crystallization amount (DRX) and mechanical properties. Because of the final application, in vitro experiments will be also in progress using mimetic physiological medium conditions to study the effect on mechanical properties and the sacffolds’s cell toxicity effects. In vivo experiments will be then performed in order to evaluate the host tissue reactions after implantation. La consommation des français (9 g de sel par jour) est bien supérieure aux recommandations de l’Organisation Mondiale de la Santé (5 g maximum par jour). Le pain représente, à lui seul, 25% de cet apport quotidien. Or le sel est l’un de ses ingrédients jouant un rôle technologique et organoleptique fondamental1. Diminuer le taux de sel dans le pain français constitue donc un réel défi pour les technologues et les scientifiques. Des études sensorielles visant à tester deux stratégies de réduction en sel ont été réalisées sur des tranches entières ou uniquement des bouchons de mies préparés à partir de baguette française, par une quarantaine de juges. La première approche consistait à déterminer le seuil d’acceptabilité lié à une réduction progressive du taux de sel depuis 2% jusqu’à 1% par palier de 0,25%, par des tests hédoniques et triangulaires. La deuxième approche visait à évaluer l’effet de la granulométrie du sel sur la perception salée par des tests triangulaires. Les résultats obtenus ont tout d’abord permis de mettre en évidence que la présence de la croûte renforce l’intensité de la perception salée du pain. Aussi, les tests triangulaires ont montré la possibilité de diminuer la teneur en sel du pain de 20 à 30% (soit 1,50 voire 1,25 g de sel pour 100 g de farine) par rapport aux teneurs actuelles (1,8 g de sel en moyenne) même si, selon les tests hédoniques, les juges ont préféré les pains les plus salés (> 1,75 g de sel pour 100 g de farine). Enfin, comparée au sel conventionnel, cette étude a permis d’observer que l’utilisation de sel micronisé n’augmente pas de manière significative la perception salée du pain. Les modifications de la composition et de la texture de la matrice alimentaire affectant aussi la libération et la perception du sodium en bouche2, ces impacts sont actuellement étudiés pour permettre une réduction maximale de la teneur en sel dans la baguette française tout en conservant une intensité salée acceptable. The latter includes reserves deposition and, for most species, a sharp decrease in water content known as maturation drying. These seeds, defined as orthodox seeds, are desiccation tolerant, like the seeds of the Fabaceae model species Medicago truncatula. However, some seeds are defined as recalcitrant because they are sensitive to desiccation and shed at high water content. Survival in dry state requires cellular and molecular protection and reparation mechanisms set up concomitantly with others physiological process (reserve deposition, germination inhibition, dormancy initiation). Specific analysis of desiccation tolerance (DT) acquisition during orthodox seed development is difficult because of the superposition of these processes. To get around this impediment, we undertook the physiological and molecular analysis of Castanospermum australe recalcitrant seed maturation, a species that also belongs to the Fabaceae family. This enables us to perform a molecular comparison between the development of desiccation-sensitive and desiccation-tolerant seeds. This comparison is expected to provide putative mechanisms involved in DT acquisition. Here we present the first analysis of the transcriptome of C. australe sequenced with the Illumina technology. After annotation by similarity approach of the 18,000 sequenced obtained, we established a list of (putative) functions expressed in C. australe seeds. Similar lists were obtained from microarray data of different stages of M. truncatula seed development allowing the comparison of the functions expressed in the two species. This study reveals that the biological processes that are over-represented in the transcriptome of C. australe are related to metabolism, whereas biological process related to late development and regulation are underrepresented compared to the orthodox seed transcriptome. The molecular data suggests that the development of C. australe seeds is truncated compared to one of M. truncatula seeds, i.e. reserve deposition continues until abscission in these recalcitrant seeds which do not undergo late maturation. This hypothesis will be discussed. Knowledge about targeting-peptide cleavage sites is essential for the structural and functional characterization of the mature organellar proteins as well as for a deeper understanding of the import process. Because of the low consensus and high variability of presequences, bioinformatics of targeting-peptide cleavage fails to predict the length of the targeting peptide with high confidence. Therefore, we have developed a rapid and robust method to experimentally determine the cleavage site of the transit peptide for proteins imported into mitochondria or plastids. The protein precursor with GFP fused to its Cterminus is transiently expressed in cells (for animal proteins) or protoplasts (for plant proteins), allowing translocation into organelles, and removal of the transit peptide. After lysis, the matured protein is immunopurified using an anti-GFP antibody coupled to magnetic beads. The N-terminal amino sequence is then determined by Edman microsequencing or mass spectrometry. The method has been validated using proteins with known targeting peptide sequences and is suitable for animal and plant organelle-targeted proteins. Functional characterization in Xenopus oocytes using both electrophysiological and K15NO3 uptake approaches showed that MtNRT1.3 encodes a dual affinity nitrate transporter1. Switching between the two modes of action is regulated in part by phosphorylation of the threonine residue at 104. Indeed changing this threonine residue to aspartic acid by directed mutagenesis, mimicking a constitutively phosphorylated state, shows a defect in low affinity uptake activity. However, pharmacological treatment of oocyte, using activators or inhibitor of protein kinase A, shows that the dual affinity regulation would probably involve a second phosphorylation site. MtNRT1.3, as all NRT1(PTR) transporters in higher plants, contains 12 putative transmembrane domains2. MtNRT1.3-GFP or RFP fusions confirm the membrane localization, probably in the plasma membrane. MtNRT1.3 expression is down-regulated by high nitrate concentration while it is not regulated by a low nitrate concentration. In contrast, MtNRT1.3 expression is up-regulated by an exogenous addition of abscissic acid and also after water deficit or salt stress. These results suggest a role of MtNRT1.3 in response to abiotic stress; this hypothesis will be tested using RNAi mutants. Ces méthodes de modélisation ont notamment un intérêt en biologie de la conservation, puisqu’elles permettent d’estimer les zones les plus favorables à une espèce ou encore d’estimer une probabilité de présence de l’espèce dans des régions où celle-ci n’a pas encore été observée faute d’investigation. Ces modèles se basent sur la mise en relation de données d’observation de l’espèce et de variables environnementales. Une cause récurrente d’inexactitude de ces modèles est l’utilisation d’un jeu de données d’observations géographiquement biaisé. En effet, ces données sont couramment issues d’observations opportunistes et rarement d’un protocole de suivi rigoureux. Ceci peut conduire à la surestimation des paramètres environnementaux liés aux sites les plus régulièrement visités ou les plus densément peuplés, où la probabilité que quelqu’un ait observé l’espèce est la plus grande. A l’heure actuelle, il n’existe malheureusement pas de consensus sur la méthode à utiliser pour résoudre ce biais. Plusieurs méthodes décrites dans la littérature comme pouvant contribuer à résoudre un tel biais géographique sont ici analysées en prenant en exemple le râle des genêts (Crex crex). Les données de présence disponibles du râle des genêts présentent typiquement ce type de biais : plus de 80% des observations disponibles se trouvent en Europe de l’Ouest, tandis que seulement quelques-unes proviennent de Russie et d’Asie centrale, pourtant connus pour abriter la plus grande part de l’aire de répartition et des effectifs de l’espèce. Les performances relatives de différentes méthodes de résolution de ce biais sont évaluées, permettant ainsi de mieux estimer la distribution actuelle de cette espèce et d’améliorer la prise en compte d’un tel biais dans la réalisation de modèles de distribution. SECONDI Groupe Ecologie et Conservation des Vertébrés (GECCO), UFR Science, Université d’Angers, France Les prairies de fauches et le bocage sont des habitats qui ont fortement déclinés en Europe de l’Ouest. Des politiques de conservation encouragent leur protection cependant des conflits peuvent apparaitre quand des mesures concernant deux composants du paysage sont appliquées sur un même territoire. Nous avons étudié cette question dans la plaine inondable de la vallée de la Loire. L’occupation du sol de la zone est largement dominé par la prairie permanente, cependant la matrice paysagère varie grandement entre de larges prairies de fauche très ouvertes et un bocage très dense. Cette hétérogénéité de la matrice paysagère n’est pas prise en compte dans les politiques locales de conservation. Nous avons testé l’effet de la taille des parcelles ainsi que la densité de haies sur la richesse et l’occurrence des espèces de passereaux prairiaux. Les passereaux prairiaux évitent les petites parcelles et les zones de forte densité de haie. Ainsi, les oiseaux s’installent préférentiellement dans les grandes prairies sans haies. Ces résultats sont importants pour la gestion de ces habitats. Les politiques actuelles encouragent la plantation de haie dans les zones de prairie ouverte. Bien que cela puisse avoir des effets globalement favorables sur la communauté d’oiseaux, on peut s’attendre à des effets négatifs sur la communauté d’oiseaux prairiaux et ainsi une réduction de la diversité aviaire suite au remplacement des espèces spécialistes par des espèces généralistes. De plus, encourager financièrement la protection des oiseaux prairiaux dans des zones de bocage dense semble inapproprié. En revanche, une gestion évolutive de l’écosystème serait nécessaire. Cela peut impliquer un certain degré de planification du paysage dans le but de favoriser le maintien de matrices paysagères variées. LAHAYE Unité de recherche GRAPPE, UMT VINITERA, Ecole supérieure d’Agriculture, 55 rue Rabelais, B.P. 30748, F-49007 Angers cedex 01, France L’hypothèse d’un lien entre la variation de l’extractibilité des composés phénoliques et des propriétés mécaniques de la baie, appréhendées à travers le terme général de « texture » peut-être avancée (Le Moigne et al., 2008 ; Maury et al., 2009 ; Zouid et al. ; 2010). En effet, ces travaux ont notamment permis de démontrer l’intérêt d’appréhender la qualité texturale du raisin au travers de l’étude de la variation des propriétés mécaniques des baies issus du cépage Cabernet Franc au cours de la maturation. Cependant, peu de données ou de travaux sont référencés dans la littérature concernant l’analyse de ces propriétés et de leur évolution pour des cépages issus de variétés de raisins blancs. Dans le cadre de cette étude, pour le millésime 2010, l’évolution des propriétés mécaniques des baies de Chenin de quatre différentes parcelles du Val de Loire ont été étudiées sur deux dates de fin de maturation des raisins. Les propriétés mécaniques des baies ont été déterminées par des mesures de double compression non-destructive 20%, compression destructive 70%, traction sur pellicule, traction sur pédicelle et pénétrométrie. En parallèle, les mêmes mesures ont été appliquées sur des raisins de Cabernet Franc provenant de 2 parcelles, elles aussi issues de vignobles du Val de Loire, sur trois dates au cours de la maturation. La comparaison des résultats a montré des différences significatives en termes de propriétés mécaniques entre ces deux variétés Blanches (Chenin) et Rouge (Cabernet Franc) (Tableau 1). Ainsi il en ressort que les pellicules de Cabernet Franc sont plus fermes que celles des Chenin ex : Traction sur pellicule : Fmax(N) =2.04 et 1.20 respectivement pour Cabernet Franc et Chenin avec une significativité p<0.001. De plus, il est montré que la technique de pénétrométrie est capable pour toutes les parcelles, qu’elles soient de Cabernet Franc ou de Chenin, de montrer des différences significatives en fonction de la date. La compression destructive 70% permet quant à elle de discriminer significativement la date de maturation uniquement pour les parcelles issues de cépage Chenin. Les perspectives de ce travail seront par la suite de mieux comprendre l’origine de ces évolutions et de poursuivre par l’identification à une échelle microscopique (cellules et parois du raisin) des phénomènes responsables de la variation des propriétés mécaniques pendant sa maturation. Pomel (syn. Orobanche ramosa) dépend entièrement, pour son développement, des nutriments qu'elle prélève dans le phloème de la plante hôte, par le biais d'une structure anatomique spécialisée, l'haustorium. En France, la récente adaptation de ce parasite au colza d'hiver (Brassica napus L.) devient un réel problème agronomique dans les régions les plus touchées, où il cause d'importantes pertes de rendement. Étant donné le rôle majeur que joue la fertilisation dans la productivité du colza d'hiver, notre étude s'est intéressée principalement aux flux d'azote au sein de cette interaction. L'utilisation du 15N comme marqueur isotopique a permis de caractériser les flux d'azote ayant lieu entre un génotype sensible de colza (ES Alienor) et P. ramosa. Un flux précoce et important d'azote provenant du compartiment foliaire et dirigé quasi-uniquement vers le parasite, a été mis en évidence dès la sortie de vernalisation. Afin d'appréhender les composés azotés transférés, les profils d'aminoacides libres contenus dans les exsudats phloèmiens de feuilles de colza et dans les différents organes de l'orobanche, ont été déterminés (UPLC-PDA). Le Glu, Asp, Gln, S-Methylcysteine Sulfoxide "SMCSO", Ser et GABA sont principalement véhiculés par le phloème, alors que le SMCSO, Gln, Asp, Glu et Asp se trouvent majoritairement accumulés dans l'orobanche. L'étude de la métabolisation des aminoacides prélevés par l'orobanche au sein du phloème de l'hôte est donc en cours de réalisation, via le suivi du marquage issu de 15 N-aminoacides fournis à des tubercules isolés d'orobanche (UPLC/MS/MS et SMRI). La teneur en lignine, l’organisation de la lignocellulose et la variabilité tissulaire impactent négativement les rendements. Ainsi, les plantes herbacées et les graminées sont des matières premières plus intéressantes, car elles contiennent moins de lignine que les bois. La spectrométrie infrarouge à transformée de Fourier (FTIR) est connue comme une méthode puissante pour l’étude de la structure et la teneur des polysaccharides et de la lignine au sein d’un matériel pariétal. Une base de données de référence de lignines et de polysaccharides a été créée, et les signatures des unités guaiacyles, syringyles, et ester p-hydroxycinnamiques ont été établies. Tout cela pour différencier les matériels pariétaux de maïs en fonction de la teneur et de la structure des lignines Les spectres (2000–600 cm−1) ont été analysés par analyse en composantes principales. L’axe de la première composante principale (68% de la variance totale) sépare les lignées F2bm1 et F2. Les bandes mises en évidence (1730, 1666, 1242, 1053, 1035, 960 cm-1) peuvent être assignées aux polysaccharides et à l’acétylation, indiquant une différence entre F2 et F2bm1 en terme de quantité de lignine. La seconde composante principale (19% de l’inertie totale) discrimine F2bm1 par des signaux (1656, 1039 cm-1) associées aux cinnamaldehydes et aux polysaccharides, de F4 et F2bm3 par des bandes liées aux d’esters hydroxycinnamiques et guaiacyle (1513 et 1168 cm-1). Les bandes des polysaccharides dominent la zone 1470-1000 cm-1 et masquent les signaux des lignines, ce qui gêne la discrimination des échantillons en fonction de la lignine. La région 1000-750 cm-1 révèle des bandes d’absorption à 892, 856, 833 et 813 cm-1 assignées à la liaison β(1-4) glycosidique, et aux unités syringyles et guaiacyles. Un examen plus poussé a été réalisé par traitement en dérivée seconde. Les deux premières composantes principales expliquent 68 et 19% de l’inertie totale. However, additional information regarding the structural features of hemicelluloses is required to understand the fundamental relationships between the biological functions of hemicelluloses and their structures. Enzymes are actively utilized as selective tools in analytics and for targeted modifications of polysaccharide structures and function. In this study, apple var. Gala, and tomato var. Almiro hemicelluloses were isolated using a less degradative technique, and characterized as xyloglucans and glucomannans. These macromolecules and several other polysaccharides were used as substrates to screen for hemicelluloses degrading or modifying bacterial enzymes according to a medium-throughput screening procedure1. The lysates or the culture supernatants from different strains of Pseudoalteromonas were found active towards these hemicelluloses. The reaction products were characterized by chromatography and mass spectrometry to select the enzymes of interest. In future, the isolated enzymes will be purified and biochemically characterized, and used to elucidate the structural features of hemicelluloses to establish the relationship between their structures and functions. It is now well established that solid colloidal particles present the ability to adsorb at fluid interfaces to form socalled “Pickering emulsions”. The amazing properties of these systems in terms of stability and mechanical behaviour are due in part to the irreversible nature of such particle adsorption1. As a result, interest in Pickering emulsions has been renewed over the past ten years, especially for applications where the use of surfactants is undesirable. In a recent study, we showed that it was possible to obtain highly stable oil-in-water Pickering emulsions stabilized only by unmodified Cellulose Nanocrystals (CN)2. However, for the preparation of cotton nanocrystals, sulfuric acid hydrolysis, that is the most common procedure, introduces sulfate ester groups at the surface providing good colloidal stability thanks to charge repulsions while the absence of charge can lead to the production of mildly flocculated nanocrystals. If the charge density variation has already been studied for the formation of cholesteric liquid crystal oganisation3, none considered the impact on the wettability of these nanocrystals at an oil/water interface. We used this original ability of the nanocrystals to stabilise oil/water interface to study the influences of surface charge on their amphiphilic properties4. Therefore, various samples with controlled surface charge density were produced and evaluated for their ability to stabilize an oil/water interface. These observations with conductometric titration, electron microscopies (TEM and SEM), X-ray scattering or granulometry have leaded to a nanocrystal scale evaluation of the absorption mechanism at the interface. The pectic RGI is mainly composed of a repeating disaccharide unit [→2)-α-L-rhamnosep-(1→4)-α-Dgalacturonic acidp-(1→]n (RU)n] decorated primarily with arabinan and (arabino)-galactan side-chains. Monoclonal Antibodies (mAbs) are useful to probe pectin structural domains in situ. Several mAbs to the RG1 backbone1 and to arabinan and (arabino)-galactan side-chains2-4 have been developed but up to now, there is no mAb against rhamnose bearing arabinose or galactose units. Here, we report on the isolation of lowly branched RU oligosaccharides, for producing antibodies against RGI side chains connected with the backbone. RGI was extracted from sugar beet (RGI arabinan-rich) by NaOH (0.1M) and precipitated by 70% ethanol. The precipitate underwent an anion exchange chromatography where the neutral sugar rich RGI fraction was eluted first and well separated from residual homogalacturonan (HG). RGI fraction was then hydrolysed with 100 mM HCL to remove 80% of the arabinose. RGI backbone was split using a rhamnogalacturonase1 and the large fragments (mostly galacturonic acid rich RGI) were precipitated with ethanol 60%. The ethanol soluble fraction contained mainly rhamnose (19%) galacturonic acid (23%), arabinose (29%) and galactose (29%). The next step will be to check that the pool of oligosaccharides is not recognized by available mAbs against RGI, and to remove the recognised epitopes (if any) by affinity chromatography, using RGI mAbs as ligands. Pour augmenter les rendements de saccharification, la digestibilité des tiges doit être améliorée. Les lignines de la paroi végétale sont des biopolymères de monolignols liés à des acides hydroxycinnamiques comme l’acide férulique. Dans la paroi, les monomères et dimères d’acide férulique sont majoritairement présents, suivi de l’acide p-coumarique. Les acides féruliques sont liés aux hémicelluloses et servent de point d’ancrage à la polymérisation et à la réticulation des lignines. Un modèle incomplet de voie de biosynthèse des lignines existe mais la synthèse de l’acide férulique n’est pas clairement connue. Des expérimentations in vitro suggèrent que l’acide férulique est produit par l’acide caféique / acide 5-hydroxyferulique O-méthyltransférase (COMT) par O-méthylation de l’acide caféique. Afin de montrer le rôle de la COMT dans la production de lignine et d’acide férulique chez les graminées, la collection de graines mutagénéisées à l’azoture de sodium de l’INRA de Versailles a été criblée par TILLING, en collaboration avec l’URGV de Évry. Des lignées mutées dans un gène codant pour la COMT ont été identifiées. Des plantes homozygotes pour la mutation COMT ont été génotypées et analysées chimiquement. Parmi ces mutants, une lignée (5139) présente une diminution de 50% du ratio de lignine S/G. Des résidus 5-OHG caractéristiques d’une déficience en COMT apparaissent. Une diminution de 20% de la quantité d’acide p-coumarique est également constatée mais il n’y a pas de modification en quantité d’acide férulique. Plusieurs hypothèses sont à vérifier. La COMT mutée pourrait avoir une activité enzymatique diminuée mais elle serait encore fonctionnelle, ou alors, il existerait une autre enzyme qui interviendrait dans la synthèse de l’acide férulique et des monolignols de type S en complément de la COMT. Due to their chemical properties, they can potentially be converted into various different value added products including biofuels and industrially-relevant chemicals. Microorganisms are considered as an important source of biocatalysts for hydrolyzing xylans to improve the release of monosaccharides for subsequent fermentation or bioconversion to biofuels. However, the improvement of enzymatic biorefining requires a constant effort to discover an optimized enzymatic cocktail for xylans depolymerization. Several strategies are possible to explore new enzymatic tools such as genetic evolution of existing enzymes, systematic screening of various and complex biological media. Nevertheless, in order to optimize these strategies, the availability of easy to handle, reliable and rapid screening tests capable to explore large sample sets is of critical importance. Systems at nanometric scale can be used as a model which can be exploited to optimize enzymatic test at large scale. In this context, recent patented research has led to the development of thin films made from biopolymers deposited on a reflective substrate1. The nanometric dimensions of such films lead to the appearance of structural colours, as a result of interference phenomena. The action of a hydrolytic enzyme can modify the thickness of the layer and, consequently through the film colour change, it is possible to visually and rapidly detect enzyme activity1,2. In this work, elaboration of films composed of alternated layers of cellulose nanocrystals and xylans, using spin-coating method, are presented. Films construction is based on electrostatic interaction between negatively charged cellulose nanocrystals of cotton and cationic oat spells xylans3. Growth pattern of the multilayered films (from 1 to 8 bilayers), morphology analyses of cationic xylans adsorption onto cellulose nanocrystals surfaces and study of their sensitivity to hydrolyze by xylanase are described. 1. Washing and drying Nanometric film consisting of five cellulose nanocrystals/Xylan bilayers exhibits a uniformly blue structural color and has a thickness value which is around 100 nm. The film is then subjected to the action of xylanase solutions with different enzymatic activities (increasing activity from 1 to 7). After several minutes of incubation followed by washing and drying, the colour of the layer is modified. This chlorinated sesquiterpene derivative of fumagillin exhibited a specific antiproliferative activity against osteosarcoma cell lines and in vivo antitumor activity. In this work, natural structural analogs of ligerin were searched by investigation of the metabolic fingerprints of extracts of four morphologically different strains belonging to this new Penicillium species grown on 6 different culture media. For that purpose, we developed a LC-MS/MS-based strategy: a fragmentation modelization was established by thoroughly studying the fragmentation patterns of both ligerin and purified or synthetized derivatives such as fumagillin, fumagillol and TNP470. The use of this model allowed the detection and structural identification of several minor compounds related to ligerin. This study shows the interest of LC-HRMS/MS analyses of the metabolome of marine-derived fungi for the rapid identification and then the targeted purification of original bioactive secondary metabolites in a chemical series. Salt reduction in food is also becoming a major concern of public health authority. However, salt reduction can induce modifications of food preservation and organoleptic properties such as texture and taste. Hence, it is a challenge for food industry to reduce sodium content while maintaining food quality. As observed in literature, modification of salt spatial distribution by food process variations can increase saltiness perception1,2,3. The change of manufacturing process could also be an original way to reduce salt content without altering saltiness. The European project “PLEASURE” (7th framework 2012-1015) deals with this objective in order to reduce salt, sugar and fat content in complex matrices by changing the manufacturing process. The aim of the present work is to evaluate the impact of salt structure and incorporation ways on saltiness and texture properties of model pizza dough composed of flour, water and salt added at the level of 2g NaCl per 100g of flour. Three salt structuring and salt incorporation ways were studied: i) addition of total salt content preliminary dissolved in water at the beginning of dough mixing, ii) addition of total salt content under dry form after 3 min of water and flour mixing, iii) addition of 50% salt content preliminary dissolved in water at the beginning of dough mixing whereas 50% of salt under dry form were added after 3 min of mixing. Samples were then baked for sensory evaluations performed by 50 untrained panelists. A ranking test was performed to evaluate saltiness perception and a sorting test with descriptor generation was used to evaluate modifications of textural properties according to process variations of model pizza dough. Complementary triangular tests were also performed by 30 untrained panelists. Instrumental measurements were performed to evaluate total salt content and in vitro salt kinetics release in artificial saliva from model pizza dough. Instrumental and sensory results were compared. Sorting test revealed that textural perception of pizza model dough slightly varied according to process variations. However, ranking test demonstrated that saltiness perception was not affected by variation of initial salt structure and incorporation ways. This result was confirmed by triangular test analysis and in accordance with instrumental results. As expected, total content of salt of model pizza dough was the same whatever process condition. Whereas small differences of salt kinetics release, no significant differences were observed. Complementary experiments analyzing salt distribution in model cooked pizza dough allowed hypothesizing that initial heterogeneous repartition of salt, induced by variation of salt structure and incorporation way, was not conserved in cooked products. In fruit sector, quality managers need to be sure that very sensitive differences between close cultivars or even clones are detected. Understanding sensory results variability is a challenge, even more in fruit, evaluation as it is due to both individual differences between panelists and product heterogeneity. Indeed, fruit vary depending on different physiological and environmental factors. Even different parts of the same piece of fruit may vary. Both experiment design and data processing miss in integrating product variability and panel heterogeneity. This study shows how training process, performances measurement and profiling method can be adapted, in order to better master panel training for fruit (with apple as an example) and to get more insight in profile results. A trained panel can discriminate between very close apples, even within a single batch. For sensory profile, a methodology consisting in the sharing of each fruit by several assessors is proposed. Three apples cultivars were studied in three replicates. The standard analysis for such data is the analysis of variance (ANOVA). Having several assessors tasting the same fruit allows for testing of fruit effect. Product heterogeneity can therefore be tested with proper linear mixed modeling and results get more consistency. In the design where an apple is shared by several assessors, including fruit effect in the analysis of variance adds information. As a conclusion, training and performance measurement should be run with several assessors tasting the same piece of fruit. The panel leader will therefore correctly evaluate assessors’ capacity. Moreover profile should also be run in this way to take into account fruit variability in the batch and to get more reliable results about product differences. Preference mapping reaches this objective as it identifies sensory key drivers and gives a clear sensory target. Nevertheless, the impact of the product-space on preference key driver identification and hierarchy has not been studied although results consistency is a major point. The goal of this study was to investigate the importance of product-space choice in external preference studies by the use of CLV and CCSWA methods. The relative contribution of texture, taste and aroma on overall liking has been examined as well. 31 apple cultivars were precisely described with 15 descriptors using a conventional sensory profile. This characterisation led to the selection of three product-spaces of same size: a first one composed of cultivars, representative of the whole sensory variability of apples, a second one focusing on cultivars with a similar texture and a third one including apples with the same acidity level. All the products were tested by 224 consumers to get a hedonic score and in parallel, evaluated by the trained panel. The three product-spaces were analysed with the same methodology. Internal preference mapping, on one side, identified the different consumer segments; external preference mapping, on another side, linked sensory descriptive results and hedonic data using a CLV integrated approach1, for each segment. Afterward, results from the different product-spaces were compared with CCSWA method2. The results from the three product-spaces show different consumer segmentations and preference key driver identifications. When consumers test products, similar for one of the sensory dimensions, the impact of others on liking is highlighted. For example, acidity becomes a segmenting attribute when texture is similar for all the tested apples.
