L’équipe s’intéresse notamment au contrôle génétique de la transition haploïdevers-diploïde et la détermination sexuelle chez Ectocarpus. Cette espèce présente un cycle digénétique haplo-diplophasique où les générations diploïde (sporophyte) et haploïde (gamétophyte) sont multicellulaires mais morphologiquement distinctes. Un crible génétique a identifié deux mutants, ouroboros (oro) et samsara (sam) affectés dans la progression du cycle de vie4. Ces mutants réitèrent le développement du gamétophyte lors de la phase diploïde. Les gènes ORO et SAM codent pour des régulateurs maîtres qui spécifient l’identité de l’individu diploïde entier. Contexte scientifique et enjeux (version anglaise facultative maximum 10 lignes) The Algal Genetics Group has been developing Ectocarpus as a model organism for the brown algae1 and lead a project that has included complete genome sequencing and the establishment of genetic tools for this species2,3. The team focused on the genetic control of the haploid-to-diploid transition and the sexual determination in Ectocarpus. This species exhibits a digenetic, haplodiplophasic life cycle where both diploid (sporophyte) and haploid (gametophyte) generations are multicellular but morphologically dissimilar. A recent genetic screen identified two mutants, ouroboros (oro) and samsara (sam), which are affected in life cycle progression4. These mutants continually reiterate the gametophyte generation even in diploid context. Objectif et plan de recherche (version française maximum 20 lignes) Les gènes ORO et SAM codent pour des facteurs de transcription qui contrôlent l’implémentation du programme génétique menant au développement du sporophyte. Des analyses transcriptomiques indiquent que ce programme implique une altération dans le patron d'expression de plusieurs centaines de genes. Le but de ce projet sera d'utilisé le système double hybride de levure afin d'identifier des protéines qui interagissent avec ORO et SAM. Une banque d'ADNc d'Ectocarpus sera construite dans un vecteur d'expression levure et la banque criblée avec des constructions "appât" ORO et SAM déjà existant au laboratoire. Une série de constructions de délétion, également disponibles dans le laboratoire hôte seront utilisées afin d'identifier des domaines des facteurs de transcription impliqués dans chaque interaction. Objectif et plan de recherche (version anglaise facultative maximum 20 lignes) The ORO and SAM genes encode two transcription factors that are required for the implementation of the sporophyte developmental program in Ectocarpus. Transcriptome analysis indicates that this program involves alterations in the expression patterns of several hundred genes. The aim of this project is to use the yeast two-hybrid system to search for proteins that interact with ORO and SAM during the implementation of the sporophyte program. An Ectocarpus cDNA library will be constructed in a yeast two-hybrid expression vector and the library will be screened with ORO and SAM bait constructs that are already available in the host laboratory. A set of deletion constructs is also available to identify which domains of the transcription factors are involved in each interaction. Techniques (version française maximum 6 lignes) Le projet emploiera des techniques de biologie moléculaire comme l’extraction d’ARN à partir de tissus d’Ectocarpus, la construction d’une banque d’ADNc et l’analyse des interactions protéine-protéine par la méthode de double-hybride de levure, ainsi que le séquençage Sanger afin de caractériser des clones correspondant aux protéines interagissant. Techniques (version anglaise facultative maximum 6 lignes) The project will employ molecular biology techniques including RNA extraction from Ectocarpus tissues, cDNA library construction and protein-protein interaction screening using the yeast two-hybrid system to identify interacting partners of ORO and SAM. Résultats attendus (version anglaise facultative maximum 6 lignes) 1) Construction and validation of an Ectocarpus yeast two-hybrid cDNA library. 2) Identification of proteins that interact with the transcription factors ORO and SAM. 3) Identification of the domains in the ORO and SAM proteins involved in the interactions with the identified protein. Following confirmation of the interactions by alternative methodologies, the data concerning these interactions will be combined with existing data describing the ORO and SAM genes for publication. Un crible génétique récent a identifié deux mutants, ouroboros (oro) et samsara (sam), qui réitèrent le programme de développement spécifique de la phase haploïde en contexte diploïde. Les gènes ORO et SAM sont deux régulateurs clés du développement qui déterminent quel programme génétique doit être exprimé par le zygote issu de la fécondation. Le but de ce projet sera d'identifié des protéines d'Ectocarpus qui interagissent avec ORO et SAM et d'identifier des domaines d'ORO et de SAM impliqués ces interactions. Descriptif du stage (version anglaise facultative 5 lignes maximum) Ectocarpus exhibits an alternation between two multicellular phases during its life-cycle: a diploid sporophyte and a haploid gametophyte. A recent genetic screen identified two mutants, ouroboros (oro) and samsara (sam), which reiterate the haploid-specific program during the diploid phase. The ORO and SAM genes encode the components of a key developmental regulator, determining identity of the organism during the life-cycle progression. The aim of this project will be to identify Ectocarpus proteins that interact with ORO and SAM and to determine which domains of the transcription factors are involved in these interactions.
