ChemMate™ DAKO EnVision™ Detection Kit, Peroxidase/DAB, Rabbit/Mouse, is intended for use in immunocytochemistry together with the DakoCytomation TechMate™ and Autostainer Instruments. The kit is employed in a two-step procedure where the first step is incubation of the tissue with an optimally diluted primary rabbit or mouse antibody, and the second step is incubation with the ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV) reagent of the kit. This reagent is a peroxidase-conjugated polymer, which also carries antibodies to rabbit and mouse immunoglobulins. The reaction is visualized by the ChemMate™ DAB+ Chromogen also included in the kit. Introduction ChemMate™ DAKO EnVision™ Detection Kit, Peroxidase/DAB, Rabbit/Mouse, is for use on the DakoCytomation TechMate™ and Autostainer Instruments. The instructions for the two instruments differ and are, therefore, provided in separate sections. Reagents Materials provided Bottle A ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV) 100 mL, ready-to-use Dextran coupled with peroxidase molecules and goat secondary antibody molecules against rabbit and mouse immunoglobulins. In buffered solution containing stabilizing protein and preservative. Bottle B ChemMate™ Substrate Buffer 250 mL Buffered solution containing hydrogen peroxide and preservative. Bottle C ChemMate™ DAB+ Chromogen 5 mL, 50 x concentrated 3,3'-diaminobenzidine tetrahydrochloride in organic solvent. The colour of this reagent may vary from strong violet to colourless without having any influence on the performance of the kit. Precautions 1. 2. For professional users. Bottle A, ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV), contains material of animal origin and it cannot be excluded that trace amounts of human material could be present due to manufacturing procedures. Do not expose Bottle C, ChemMate™ DAB+ Chromogen or the Substrate Working Solution (CHROM) to strong light. The reagents loaded on the TechMate™ Instruments should not be reused. Bottle C, ChemMate™ DAB+ Chromogen contains 1.5% 3,3’-diaminobenzidine tetrahydrochloride, and is labelled: Harmful. R40 Limited evidence of a carcinogenic effect. R43 May cause sensitisation by skin contact. R68 Possible risk of irreversible effects. S35 This material and its container must be disposed of in a safe way. S36/37 Wear suitable protective clothing and gloves. As a main rule, persons under 18 years of age are not allowed to work with this product. Users must be carefully instructed in the proper working procedure, the dangerous properties of the product and the necessary safety instructions. Please refer to the Material Safety Data Sheet (MSDS) for additional information. Storage Store the kit at 2-8 °C. Do not use after expiration date stamped on the kit. Do not interchange kit components from different lots. If reagents are stored under any conditions other than those specified, the conditions must be verified by the user. The prepared Substrate Working Solution (CHROM) should be stored at 2-8 °C and used within 5 days. If unexpected staining is observed which cannot be explained by variations in laboratory procedures and a problem with the product is suspected, contact our Technical Services. Quality Control Each staining run should include a known positive control specimen to ascertain a proper performance of all the applied reagents. If the positive control specimen fails to demonstrate positive staining, labelling of test specimens should be considered invalid. A negative control reagent should be used with each specimen to identify any non-specific staining. If non-specific staining cannot be clearly differentiated from the specific staining, the labelling of the test specimen should be considered invalid. Interpretation of Results The diaminobenzidine-containing Substrate Working Solution gives a brown colour at the site of the target antigen recognized by the primary antibody. The brown colour should be present on the positive control specimen at the expected localization of the target antigen. If non-specific staining is present, this will be recognized as a rather diffuse, brown staining on the slides treated with the negative control reagent. Nuclei will be stained blue by the hematoxylin counterstain. (104958-002) K 5007/EFG/HGW/26.09.03 p. 6/47 INSTRUCTIONS FOR DakoCytomation TechMate™ INSTRUMENTS Summary and Explanation ChemMate™ DAKO EnVision Detection Kit, Peroxidase/DAB, Rabbit/Mouse, has been designed to give optimal staining on the TechMate™ Instruments when using the protocols outlined in the instruction below. The automatic immunostaining on the TechMate™ Instruments uses capillary action to draw up reagents from Reagent Trays, code No. S 2039, to cover specimens mounted on special Capillary Gap Microscope Slides. After a predetermined incubation time, reagents are removed from specimens by absorption into Absorbent Pads, code No. S 2027 for TechMate™ 500/500 Plus/1000, and code No. S 2043 for TechMate™ Horizon. Prior to staining on the TechMate™ Instruments, routinely fixed, paraffin-embedded tissue sections should be subjected to epitope retrieval in microwave oven using ChemMate™ Target Retrieval Solution, code No. S 2031, diluted 1:10. Please note that some primary antibodies require proteinase K pre-treatment of tissue for optimal staining. Endogenous peroxidase should be blocked with the Peroxidase-Blocking Solution, code No. S 2023. Owing to an effective washing procedure and the presence of carrier proteins in the DakoCytomation reagents, extra blocking steps to reduce non-specific background staining are unnecessary. The ChemMate™ Buffer Kit, code No. K 5006, contains Wash Buffers 1, 2, 3 and Water Wash. Wash Buffer 1 is used during the first part of the procedure. The formulation of this buffer contributes to the blocking of non-specific binding of the primary antibody and the ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV) detection reagent to the tissue sections. Wash Buffers 2 and 3 are identical and are used for the remaining part of the procedure. As Wash Buffers 2 and 3 do not contain sodium azide, they are used especially in conjunction with washing after incubation with the ENV detection reagent. Water Wash is used for washing steps at the end of the staining protocols. Primary antibodies are not provided with the kit. We recommend the use of concentrated DakoCytomation primary antibodies. Please see the guideline for optimal dilutions in Table 2. The ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV) detection reagent in the kit consists of a dextran backbone to which a large number of peroxidase (HRP) molecules and secondary antibody molecules have been coupled. A unique chemistry is used for the coupling reaction, which permits the binding of up to 100 HRP molecules and up to 20 antibody molecules per backbone. The secondary antibody coupled to the dextran backbone has been raised in goats. It reacts equally well with rabbit and mouse immunoglobulins, thus only one detection reagent is required for rabbit and mouse primary antibodies. The ENV detection reagent is provided ready-to-use in a dropper bottle. The substrate system consists of two components, ChemMate™ DAB+ Chromogen, a concentrated diaminobenzidine (DAB) solution, and the ChemMate™ Substrate Buffer containing hydrogen peroxide. Before use, the DAB+ Chromogen must be diluted in the Substrate Buffer. The substrate system produces a brown colour at the site of the target antigen. ChemMate Hematoxylin, code No. S 2020, is recommended for counterstaining as it provides a clear blue, nuclear staining. The stained tissue sections may be mounted with either aqueous or organic-solvent-based mounting medium. (104958-002) K 5007/EFG/HGW/26.09.03 p. 7/47 Reagents A. Reagent preparation Substrate Working Solution (CHROM) The DAB-containing Substrate Working Solution (CHROM) is prepared by mixing thoroughly 50 parts of ChemMate™ Substrate Buffer (Bottle B) with 1 part of ChemMate™ DAB+ Chromogen (Bottle C). Prepare only the volume required for the number of slides that shall be stained. Please see Table 1 below. One well corresponds to two slides that have been paired. Use CHROM within 5 days (store in darkness at 2-8 °C). (104958-002) K 5007/EFG/HGW/26.09.03 p. 8/47 Specimen Collection and Preparation The specimens may be routinely fixed, paraffin-embedded tissue sections and frozen tissue sections. A variety of fixatives may be used for specimen preservation. Specimens should not be fixed in neutral buffered formalin for more than 24 hours and preferably for a shorter time. The optimal thickness of paraffin-embedded sections is approximately 4 µm. The optimal thickness of frozen sections is 4-6 µm. All specimens should be mounted on ChemMate™ Capillary Gap Microscope Slides and placed in the slide holder "face to face" in pairs. As the slides have already been coated with adhesive, the user is advised not to treat the slides further, as this may have a detrimental effect. Three types of Capillary Gap Microscope Slides are available: 1. Slides with grey, 75 µm thick painted areas. These are for paraffin-embedded tissue sections and should be paired with similar slides. 2. Slides with blue, 100 µm thick painted areas. These are for frozen sections and smears and should be paired with similar slides. 3. Slides with yellow, 155 µm thick painted areas. These slides should not be mounted with tissue sections. They are for use with tissue sections which have been mounted on ordinary, non-capillary gap slides. Simply pair the ordinary slide with the yellow slide to create a proper capillary gap. NOTE: Ordinary slides must have a width of no more than 25 mm to be able to fit into the slide holder. Specimens should be mounted on that part of the slide which is farthest from the painted label end of the slide. It is important that specimens be placed on the same side of the slide that holds the painted label area. As the tissue sections are placed "face to face" during the staining procedure, the sections should be mounted on the slides as flat and wrinkle-free as possible. Too many wrinkles or clumps will inhibit the capillary action and may cause inconsistent staining results. Paraffin sections should be mounted from a pre-heated water bath containing distilled or deionized water. The water bath should contain no additives (such as gelatin, poly-L-lysine etc.). Sections should be dried by heating, generally at a temperature not above 60 °C for a minimum of 60 minutes (e.g. overnight). To ensure proper adherence of sections to slides it is important to drain the water from beneath the sections prior to the oven drying process. Procedure Before running protocols on the DakoCytomation TechMate™ Instruments, please read carefully the Operator's Manual for the dedicated TechMate™ Instrument. Prior to starting the TechMate™ Instrument, slides must be organized for the staining run: Routinely fixed, paraffin-embedded tissue sections: After deparaffinization and hydration to buffer (water), tissue sections should be subjected to epitope retrieval in microwave oven (see section A below). A few epitopes do not tolerate the treatment in microwave oven. Please refer to the instructions for the individual DakoCytomation primary antibody. In addition to being heated in a microwave oven, a few epitopes require a short enzyme digestion on the TechMate™ Instrument. Please refer to Table 2 and to the instructions for ChemMate™ Proteinase K, code No. S 2019. Those specimens that require enzyme digestion should be grouped together. If no specimens require enzyme digestion, use protocol ENVP for all slides (this protocol is performed in approximately 2 ¼ hours). (104958-002) K 5007/EFG/HGW/26.09.03 p. 9/47 If some specimens require enzyme digestion, use protocol ENVPE for all slides (this protocol is performed in approximately 2 ¼ hours). For those specimens that do not require enzyme digestion, simply replace the proteolytic enzyme in the well with Proteinase K Diluent, code No. S 2032, or Washing Buffer 2 or 3 from ChemMate™ Buffer Kit, code No. K 5006. A. Epitope retrieval in microwave oven It is recommended to follow carefully the instructions below in order to ascertain reproducible epitope retrieval. To standardize the treatment, it is recommended to load the slide holder fully with slides, even if only one slide may hold tissue; this ensures an identical heating of sections in every run. At the termination of the epitope retrieval step, slides must be left in the buffer for at least 20 minutes at room temperature. During this cooling, the TechMate™ Instrument can be prepared for the staining run. Microwave oven procedure 1. Place the deparaffinized tissue sections in a ChemMate™ Slide Holder, code No. S 2029. Transfer the ChemMate™ Slide Holder to a ChemMate™ Incubation Container, code No. S 2030, filled with 200 mL of ChemMate™ Target Retrieval Solution, code No. S 2031, diluted 1:10. 2. Place the Container in the microwave oven with the lid at an angle (e.g. diagonally). Use an 800 watt microwave oven on high setting. Microwave for 5 minutes after the buffer has come to a boil. Add 50 mL of deionized water to replace the evaporated water. Microwave for another 5 minutes. Remove the Container from the oven and let it cool at room temperature for at least 20 minutes. 3. Exchange the buffer with deionized water and leave for a short time. Now the slides are ready to be loaded into the TechMate™ Slide Holder. NOTE: Boiling of the buffer is normal and desirable. Not all 800 watt microwave ovens are similar in heat transfer. Therefore, to secure a local standardization, the microwave oven should be tested with slides without tissue the first time to find the total microwaving time necessary for obtaining 5 minutes of actual boiling. B. Guideline for optimal dilution of primary antibodies and proteinase K pretreatment Table 2 provides a dilution guideline for DakoCytomation concentrated primary antibodies used with the ChemMate™ DAKO EnVision™ Detection Kit, Peroxidase/DAB, Rabbit/Mouse, and applied on formalin-fixed, paraffin-embedded sections. The primary antibodies should be diluted in ChemMate™ Antibody Diluent, code No. S 2022. (104958-002) K 5007/EFG/HGW/26.09.03 p. 10/47 Table 2. Staining protocol The protocols ENVP and ENVPE, and templates for reagent positions on the instrument can be viewed on the TechMate™ screen before and during the run. The protocols list number of steps, step name and incubation times for the individual steps. The templates give the positions of the reagents and absorbent pads on the TechMate™ Instrument. It is recommended to fill in an Antibody Positioning Sheet for slide pairs which should be treated individually with reagents, e.g. primary antibodies, from the 10-well trays. Please refer to the Operator's Manual. (104958-002) K 5007/EFG/HGW/26.09.03 p. 11/47 1. Load reagents into the 10-well trays on the TechMate™ Instrument according to Reagent Template and Antibody Positioning Sheet. Take care to load proper reagents and proper volumes. Pay particular attention to the loading of primary antibodies. 2. Prior to loading slides into the slide holder it is recommended to pre-wet the slides with Wash Buffer 2 from the ChemMate™ Buffer Kit, code No. K 5006. This can be done conveniently by leaving the slides in pairs, and organized according to the Antibody Positioning Sheet, for a few minutes in a 10-well tray containing Wash Buffer 2. This prewetting will ensure adequate fluid flow in the capillary gap. 3. After pre-wetting, place the slide pairs in the TechMate™ Slide Holder according to the Antibody Positioning Sheet. Please make sure that the slides are in alignment. 4. Just before the run, place the TechMate™ Slide Holder on Pad2 in the TechMate™ Instrument to drain Wash Buffer 2 from the capillary gap. Observe carefully the draining of slides in order to control that all slides have been correctly paired and that the alignment of slide pairs has been done properly. 5. Place the TechMate™ Slide Holder in the Wash Buffer (e.g. Wash Buffer 1 or Wash Buffer 2) used in the first step of the protocol to be run, in order to fill the capillary gaps with Wash Buffer. 6. Place the TechMate™ Slide Holder in HOME position before starting the run. Please refer to Chapter 5 "RUNNING A SINGLE PROTOCOL" in the Operator's Manual. NOTE: The final steps in the protocols are washing in Water Wash (from ChemMate™ Buffer Kit, code No. K 5006), designated H2O on the template and in the protocol. If the user prefers to prepare the specimens for mounting in aqueous mounting medium, the HOME position can be loaded with water after initiation of the run. On TechMate™ Horizon, the H2O position is equal to the HOME position. The volumes to be loaded into the trays for the individual reagents are given below. Prior to staining on the Autostainer Instruments, some routinely fixed, paraffin-embedded tissue sections should be subjected to epitope retrieval in microwave oven using the Target Retrieval Solution specified in Table 3 or the instructions for the primary antibody. Please note that some primary antibodies require additional proteinase K pre-treatment of tissue for optimal staining. Endogenous peroxidase should be blocked with Peroxidase-Blocking Solution, code No. S 2023. Owing to an effective washing procedure and the presence of carrier proteins in the DakoCytomation reagents, extra blocking steps to reduce non-specific background staining is unnecessary. The DakoCytomation Wash Buffer, Concentrated x 10, for Immunocytochemistry, code No. S 3006, is recommended. Primary antibodies are not provided with the kit. We recommend the use of concentrated DakoCytomation Antibodies. Please see the guideline for optimal dilution in Table 3. The ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV) detection reagent in the kit consists of a dextran backbone to which a large number of peroxidase (HRP) molecules and secondary antibody molecules have been coupled. A unique chemistry is used for the coupling reaction, which permits the binding of up to 100 HRP molecules and up to 20 antibody molecules per backbone. The secondary antibody coupled to the dextran backbone has been raised in goats. It reacts equally well with rabbit and mouse immunoglobulins, thus only one detection reagent is required for rabbit and mouse primary antibodies. The ENV detection reagent is provided in a dropper bottle and has to be poured into an Autostainer Reagent Vial, code No. S 3425, before use. The substrate system consists of two components, ChemMate™ DAB+ Chromogen, a concentrated diaminobenzidine (DAB) solution, and the ChemMate™ Substrate Buffer containing hydrogen peroxide. Before use, the DAB+ Chromogen must be diluted in the Substrate Buffer. The substrate system produces a brown colour at the site of the target antigen. The mixed chromogen solution should be poured into an Autostainer Reagent Vial, code No. S 3425, before use. DakoCytomation Hematoxylin, code No. S 3301, is recommended for counterstaining on the Autostainer Instruments as it provides a clear blue, nuclear staining. Reagent preparation Substrate Working Solution (CHROM) The DAB-containing Substrate Working Solution (CHROM) is prepared by mixing thoroughly 20 µL ChemMate™ DAB+ Chromogen (Bottle C) and 1 mL ChemMate™ Substrate Buffer (Bottle B). Prepare only the volume required for the number of slides that shall be stained. Use CHROM within 5 days (store in darkness at 2-8 °C). For preparation of reagents not provided with the kit, please see the instructions included with the individual reagent. Specimen Collection and Preparation The specimens may be routinely fixed, paraffin embedded tissue sections and frozen tissue sections. A variety of fixatives may be used for specimen preservation. Specimens should not be fixed in neutral buffered formalin for more than 24 hours and preferably for a shorter time. The optimal thickness of paraffin-embedded sections is approximately 4 µm. The optimal thickness of frozen sections is 4-6 µm. The specimens should be mounted on microscope slides. The sections should be mounted on the slides as flat and wrinkle-free as possible. Too many wrinkles will have an impact on the staining results. NOTE: The microscope slides must have a width of no more than 26 mm to be able to fit into the slide holder. Paraffin sections should be mounted from a pre-heated water bath containing distilled or deionized water. The water bath should contain no additives (such as gelatin, poly-L-lysine etc.). Sections should be dried by heating, generally at a temperature not above 60 °C for a minimum of 60 minutes (e.g. overnight). To ensure proper adherence of sections to slides it is important to drain the water from beneath the sections prior to the oven drying process. Procedure Before staining according to the template on the DakoCytomation Autostainer instruments, please read carefully the Operators Manual for the dedicated Autostainer Instrument. Prior to starting the Autostainer Instrument, slides and reagents must be placed according to the Slide Layout Map and the Reagent Layout Map. Routinely fixed, paraffin-embedded tissue sections After deparaffinization and hydration to buffer (water), tissue sections should be subjected to epitope retrieval in microwave oven (see section A below). A few epitopes do not tolerate the treatment in microwave oven. Please refer to the instructions for the individual DakoCytomation primary antibody. Si une coloration imprévue est observée qui ne peut pas s'expliquer par des variations dans les procédures du laboratoire et si le réactif paraît défectueux, contacter nos services techniques. Contrôle qualité Chaque technique d'immunomarquage doit inclure un spécimen de contrôle positif connu pour vérifier les performances correctes de tous les réactifs utilisés. Si le spécimen de contrôle positif ne montre pas une coloration positive, le marquage des spécimens de l'essai doit être considéré comme invalide. Un réactif de contrôle négatif doit être utilisé avec chaque spécimen pour identifier toute coloration non spécifique. S'il est impossible de différencier clairement une coloration non spécifique de la coloration spécifique, le marquage des spécimens analysés doit être considéré comme invalide. Interprétation des résultats La solution de substrat contenant la diaminobenzidine donne une couleur brune sur le site de l'antigène cible reconnu par l'anticorps primaire. Cette couleur brune doit être présente sur le spécimen de contrôle positif sur le site prévu de l'antigène cible. CONSIGNES D'UTILISATION des instruments TechMate™ DakoCytomation Résumé et explications Le ChemMate™ DAKO EnVision Detection Kit, Peroxidase/DAB, Rabbit/Mouse est conçu pour assurer une coloration optimale sur les instruments TechMate™ lors des protocoles décrits dans les consignes ci-dessous. L'immunomarquage automatique sur les instruments TechMate™ se fait par en attirant le réactif par capillarité depuis les Reagent Trays, code S 2039, pour recouvrir les spécimens montés sur des lames de microscope spéciales à capillaire creux. Après un temps d'incubation prédéfini, les réactifs sont éliminés des spécimens par absorption par les Absorbent Pads, code S 2027 pour le TechMate™ 500/500 Plus/1000, et code S 2043 pour le TechMate™ Horizon. Avant leur coloration sur les instruments TechMate™, les coupes incluses en paraffine normalement fixées doivent être soumises à une restauration de l'épitope dans un four à microondes avec la ChemMate™ Target Retrieval Solution, code S 2031, diluée à 1:10. Note: pour un marquage optimal, certains anticorps primaires nécessitent un pré-traitement des tissus à la protéinase K. La peroxydase endogène doit être bloquée avec la Peroxidase-Blocking Solution, code S 2023. Grâce à une procédure de lavage efficace et à la présence de protéines porteuses dans les réactifs DakoCytomation, il n'est pas nécessaire d'effectuer d'autres étapes de blocage pour réduire le bruit de fond non spécifique. Le ChemMate™ Buffer Kit, code K 5006, contient des Tampons de lavage 1, 2, 3 et un lavage à l'eau (Water Wash). Le tampon de lavage 1 est utilisé pendant la première partie de la procédure. La formulation de ce tampon contribue à bloquer la liaison non spécifique de l'anticorps primaire et du réactif ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV) sur les coupes de tissus. Les tampons 2 et 3 sont identiques et sont utilisés pour le reste de la procédure. Les tampons 2 et 3 ne contenant pas d'azide de sodium, ils sont utilisés plus spécialement conjointement au lavage après incubation avec le réactif de détection ENV. L'eau de lavage (Water Wash) est utilisée pour les étapes de lavage à la fin des protocoles d'immunomarquage. Les anticorps primaires ne sont pas fournis avec le kit. Nous recommandons d'utiliser les anticorps primaires concentrés DakoCytomation. Veuillez consulter les recommandations de dilution optimales dans le tableau 2. Le réactif ChemMate™ DAKO EnVision™,EnVision™/HRP, Rabbit/Mouse (ENV) inclus dans le kit est constitué d'un squelette de dextran sur lequel ont été couplées un grand nombre de molécules de peroxydase (HRP) et d'anticorps secondaires. Une réaction chimique unique est utilisée pour la réaction de couplage qui permet la liaison de jusqu'à 100 molécules d'HRP et 20 molécules d'anticorps par squelette. L'anticorps secondaire couplé au squelette de dextran est d'origine caprine. Il réagit aussi bien avec les immunoglobulines de lapin et de souris, et donc, un seul réactif de détection est nécessaire pour les anticorps primaires de lapin et de souris. Le réactif de détection ENV est fourni prêt à l'emploi dans un flacon compte-gouttes. Le système de substrat est composé de deux éléments, la solution ChemMate™ DAB+ Chromogen, une solution de diaminobenzidine (DAB) concentrée, et le ChemMate™ Substrate Buffer qui contient du peroxyde d'hydrogène. Différents fixateurs peuvent être utilisés pour les conserver. Les spécimens ne doivent pas être fixés dans du formol tamponné neutre pendant plus de 24 heures et de préférence pour une durée encore plus réduite. L'épaisseur optimale des coupes incluses en paraffine est d'environ 4 µm L'épaisseur optimale des coupes congelées est de 4 à 6 µm Tous les spécimens doivent être montés sur des ChemMate™ Capillary Gap Microscope Slides et placées dans le porte-lames "face à face" par paires. Les lames étant déjà recouvertes d'adhésif, un traitement supplémentaire n'est pas conseillé plus car cela pourrait avoir un effet préjudiciable. Trois types de lames de microscope à capillaire creux sont disponibles : 1. Lames avec zones peintes en gris de 75 µm d'épaisseur. Ces lames sont destinées aux coupes de tissus incluses en paraffine et doivent être appariées avec des lames similaires. 2. Lames avec zones peintes en bleu de 100 µm d'épaisseur. Ces lames sont destinées aux coupes de tissus congelées et aux frottis et doivent être appariées avec des lames similaires. 3. Lames avec zones peintes en jaune de 155 µm d'épaisseur. Ces lames ne doivent pas être montées avec des coupes de tissus. Elles sont destinées aux coupes de tissus déjà montées sur des lames ordinaires sans capillaires creux. Il suffit de coupler la lame ordinaire et la lame jaune pour obtenir un capillaire creux correct. REMARQUE: La largeur des lames ordinaires ne doit pas être supérieure à 25 mm pour pouvoir les placer dans le porte-lames. Les spécimens doivent être montés sur la partie de la lame la plus éloignée de l'extrémité peinte portant l'étiquette. Il est important de placer les spécimens sur le côté de la lame où se trouve l'étiquette peinte. Les coupes de tissus étant placées "face à face" pendant la procédure d'immunomarquage, elles doivent être montées sur les lames le plus à plat et avec le moins de plissements possible. Des plissements ou des amas excessifs inhibent la capillarité et peuvent provoquer un marquage irrégulier. Les coupes incluses en paraffine doivent être montées après un séjour dans un bain d'eau distillée ou déionisée préalablement chauffée. Le bain d'eau ne doit contenir aucun additif (tel que la gélatine, la poly-L-lysine etc...). Les coupes doivent être séchées à la chaleur, généralement à une température ne dépassant pas 60 °C pendant un minimum de 60 minutes (par exemple pendant la nuit). Pour assurer une bonne adhérence des coupes sur les lames, il est important d'évacuer l'eau qui se trouve dessous avant le processus de séchage au four. Procédure Avant d'exécuter un protocole sur les instruments TechMate™ de DakoCytomation, lire attentivement le Manuel d'utilisation de l'instrument TechMate™ concerné. Avant de mettre en marche l'instrument TechMate™, les lames doivent être organisées pour la procédure d'immunomarquage : Coupes de tissus normalement fixées incluses en paraffine : Après déparaffinisation et hydratation avec le tampon (eau), les coupes de tissus doivent être soumises à une procédure de restauration de l'épitope au four à micro-ondes (voir la section A ci-dessous). Quelques épitopes ne supportent pas le traitement au four à micro-ondes. Se reporter aux consignes pour l'anticorps primaire DakoCytomation concerné. (104958-002) K 5007/EFG/HGW/26.09.03 p. 23/47 En plus du réchauffement au four à micro-ondes, certains épitopes nécessitent une courte digestion enzymatique sur l'instrument TechMate™. Se reporter au Tableau 2 et aux consignes pour la ChemMate™ Proteinase K code S 2019. Les spécimens qui nécessitent une digestion enzymatique doivent être regroupés. Si aucun spécimen ne nécessite une digestion enzymatique, le protocole ENVP peut être utilisé pour toutes les lames (ce protocole dure environ 2 ¼ heures). Si certains spécimens demandent une digestion enzymatique, exécutez le protocole ENVPE pour toutes les lames (ce protocole prend environ 2 ¼ ). Pour les spécimens qui ne nécessitent pas de digestion enzymatique, il suffit de remplacer l'enzyme protéolytique dans le puits par du ChemMate™ Proteinase K Diluent, code S 2032, ou le tampon de lavage 2 ou 3 du ChemMate™ Buffer Kit, code K 5006. A. Restauration de l'épitope au four à micro-ondes Il est recommandé de suivre soigneusement les consignes ci-dessous pour garantir une restauration de l'épitope reproductible. Pour standardiser le traitement, il est recommandé de charger entièrement le porte-lames avec les lames, même si une seule comporte une coupe; cela assure un réchauffement identique des coupes à chaque analyse. A la fin de l'étape de restauration de l'épitope, les lames doivent rester dans le tampon au moins 20 minutes à température ambiante. Pendant cette étape de refroidissement, l'instrument TechMate™, peut être préparé pour l'immunomarquage. Procédure au four à micro-ondes 1. Placer les coupes de tissu déparaffinées dans un ChemMate™ Slide Holder, code S 2029. Transférer le ChemMate™ Slide Holder dans un ChemMate™ Incubation Container, code S 2030, rempli de 200 ml de ChemMate™ Target Retrieval Solution code S 2031, diluée à 1:10. 2. Placer le bac dans le four à micro-ondes avec le couvercle en diagonale. Utiliser un four de 800 W sur réglage fort. Chauffer au micro-ondes pendant 5 minutes après avoir porté le tampon à ébullition. Ajouter 50 ml d'eau déionisée pour remplacer l'eau évaporée. Chauffer au micro-ondes pendant 5 minutes supplémentaires. Retirer le bac du four et laisser refroidir à température ambiante pendant au moins 20 minutes. 3. Remplacer le tampon par de l'eau déionisée et laisser pendant un moment. Les lames sont maintenant prêtes à être chargées dans le TechMate™ Slide Holder. REMARQUE: Il est normal et souhaitable que le tampon bout. Tous les fours à micro-ondes de 800 W ne transfèrent pas la chaleur de la même manière. Donc, pour garantir une standardisation locale, le four doit être testé avec des lames sans coupes de tissus la première fois, pour déterminer le temps nécessaire pour obtenir 5 minutes d'ébullition effective. B. Les matrices donnent les positions des réactifs et des tampons absorbants sur l'instrument TechMate™. Il est recommandé de remplir une Feuille de positionnement de l'anticorps pour (104958-002) K 5007/EFG/HGW/26.09.03 p. 25/47 les paires de lames devant être traitées individuellement avec les réactifs, par exemple les anticorps primaires des plateaux à 10 puits. Veuillez consulter le Manuel d'utilisation. 1. Charger les réactifs dans les plateaux à 10 puits sur l'instrument TechMate™ selon la matrice du réactif et la feuille de positionnement de l'anticorps. Veiller à charger les réactifs et les volumes corrects. Faire particulièrement attention au chargement des anticorps primaires. 2. Avant de charger les lames dans le porte-lames, il est recommandé de les pré-humidifier avec du tampon de lavage 2 du ChemMate™ Buffer Kit, code K 5006. Cela peut se faire facilement en laissant les lames en paires disposées selon la Feuille de positionnement de l'anticorps, pendant quelques minutes dans un plateau à 10 puits contenant du tampon de lavage 2. Ce mouillage assure un flux adéquat de liquide dans le capillaire en creux. 3. Après le mouillage, placer les paires de lames dans le porte-lames TechMate ™selon la Feuille de positionnement de l'anticorps. S'assurer que les lames sont alignées. 4. Juste avant l'analyse, placer le TechMate™ Slide Holder sur le tampon absorbant 2 dans l'instrument TechMate™ pour égoutter le tampon de lavage 2 du capillaire en creux. Surveiller soigneusement l'égouttage des lames pour vérifier que toutes les lames sont appariées correctement et si l'alignement est correct. 5. Placer le TechMate™ Slide Holder dans le tampon de lavage (par exemple tampon de lavage 1 ou 2) utilisé pour la première étape du protocole pour remplir les capillaires en creux de la solution tampon. 6. Placer le TechMate™ Slide Holder sur la position HOME avant de commencer le traitement. Consulter le Chapitre 5 "EXECUTION D'UN SEUL PROTOCOLE " dans le Manuel d'utilisation. Préparation des réactifs Solution de substrat (CHROM) La solution de substrat (CHROM) se prépare en mélangeant intimement 20 µL de ChemMate DAB+ Chromogen (flacon C) et 1 mL de ChemMate™Substrate Buffer (flacon B). Préparer uniquement le volume requis pour le nombre de lames à marquer. Le CHROM doit être utilisé dans les 5 jours (conserver dans l'obscurité entre 2 et 8 °C) Pour la préparation des réactifs non fournis avec le kit, consulter le mode d'emploi de chaque réactif. Collecte et préparation des échantillons Les spécimens peuvent être des coupes de tissus normalement fixées incluses en paraffine et des coupes de tissus congelées. Différents fixateurs peuvent être utilisés pour les conserver. Les spécimens ne doivent pas être fixés dans du formol tamponné neutre pendant plus de 24 heures et de préférence pour une durée encore plus réduite. L'épaisseur optimale des coupes incluses en paraffine est d'environ 4 µm. L'épaisseur optimale des coupes congelées est de 4 à 6 µm. Les coupes de tissus doivent être montées sur des lames de microscope. Les coupes doivent être montées sur les lames le plus à plat et avec le moins de plissements possible. Des plissements excessifs affectent le marquage. REMARQUE: La largeur des lames de microscope ne doit pas être supérieure à 26 mm pour pouvoir les placer dans le porte-lames. Les coupes incluses en paraffine doivent être montées après un séjour dans un bain d'eau distillée ou déionisée préalablement chauffée. Le bain d'eau ne doit contenir aucun additif (tel que la gélatine, la poly-L-lysine etc...). Les coupes doivent être séchées à la chaleur, généralement à une température ne dépassant pas 60 °°C pendant un minimum de 60 minutes (par exemple pendant la nuit). Pour assurer une bonne adhérence des coupes sur les lames, il est important d'évacuer l'eau qui se trouve dessous avant le processus de séchage au four. Avant de mettre en marche l'instrument Autostainer, les lames et les réactifs doivent être placés selon la matrice de positionnement des lames et la matrice de positionnement des réactifs. ChemMate™ DAKO EnVision™ Detection Kit, Peroxidase/DAB, Rabbit/Mouse, ist zusammen mit den DakoCytomation TechMate™- und Autostainer-Geräten zur Verwendung in der Immunzytochemie bestimmt. Der Kit wird in einem zweistufigen Verfahren eingesetzt, bei dem der erste Schritt in der Inkubation des Gewebes mit einem optimal verdünnten primären Kaninchen- oder Mausantikörper besteht und der zweite Schritt in der Inkubation mit dem ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV)-Reagenz des Kits. Bei diesem Reagenz handelt es sich um ein Peroxidase-konjugiertes Polymer, das auch Antikörper gegen Kaninchen- und Mausimmunglobuline trägt. Die Reaktion wird mithilfe von ChemMate™ DAB+ Chromogen visualisiert, das ebenfalls zum Lieferumfang des Kits gehört. Einleitung ChemMate™ DAKO EnVision™ Detection Kit, Peroxidase/DAB, Rabbit/Mouse, ist für die Verwendung zusammen mit den DakoCytomation TechMate™- und Autostainer-Geräten bestimmt. Die Anleitungen für die beiden Geräte weichen voneinander ab und werden daher getrennt aufgeführt. Wie bei allen aus (104958-002) K 5007/EFG/HGW/26.09.03 p. 33/47 biologischen Materialien gewonnenen Produkten müssen die ordnungsgemäßen Handhabungsverfahren eingehalten werden. 3. S36/37 Bei der Arbeit geeignete Schutzhandschuhe und Schutzkleidung tragen. Personen unter 18 Jahren ist es grundsätzlich nicht gestattet, mit diesem Produkt zu arbeiten. Benutzer müssen sorgfältig in die richtigen Arbeitsverfahren eingewiesen und über die gefährdenden Eigenschaften des Produkts und die notwendigen Sicherheitsvorschriften informiert werden. Weitere Angaben sind dem Sicherheitsdatenblatt zu entnehmen. Lagerung Kit bei 2 – 8 °C lagern. Nicht nach dem auf dem Kit angegebenen Verfallsdatum verwenden. Komponenten von Kits unterschiedlicher Chargen dürfen nicht gegeneinander ausgetauscht werden. Falls die Reagenzien unter anderen Bedingungen als den beschriebenen aufbewahrt werden, so müssen diese vom Anwender verifiziert werden. Qualitätskontrolle Jeder Färbungsdurchgang muss eine bekanntermaßen positive Kontrollprobe enthalten, um eine korrekte Leistung aller angewendeten Reagenzien zu gewährleisten. Wenn die positive Kontrollprobe keine positive Anfärbung erbringt, muss die Markierung der Testproben als ungültig angesehen werden. Es sollte bei jeder Probe ein Negativkontrollreagenz verwendet werden, um unspezifische Anfärbung zu identifizieren. Wenn eine unspezifische Färbung nicht von der spezifischen Färbung unterschieden werden kann, muss die Markierung der Testprobe als ungültig angesehen werden. Interpretation der Ergebnisse Die Diaminobenzidin enthaltende Substrate Working Solution ergibt an der Stelle des Zielantigens, das von dem primären Antikörper erkannt wurde, eine braune Farbe. Die braune Färbung muss bei der positiven Kontrollprobe an der erwarteten Position des Zielantigens vorliegen. Wenn eine unspezifische Anfärbung vorliegt, wird dies auf den mit dem Negativkontrollreagenz behandelten Objektträgern als eine eher diffus erscheinden braune Anfärbung wahrgenommen. Zellkerne werden von der Hämatoxylin-Gegenfärbung blau eingefärbt. (104958-002) K 5007/EFG/HGW/26.09.03 p. 34/47 ANLEITUNGEN FÜR DakoCytomation TechMate™-GERÄTE Zusammenfassung und Erläuterung ChemMate™ DAKO EnVision Detection Kit, Peroxidase/DAB, Rabbit/Mouse, wurde darauf ausgelegt, dass bei Befolgung der in der nachstehenden Anleitung beschriebenen Verfahrensprotokolle mit den TechMate™-Geräten eine optimale Anfärbung erreicht wird. Die automatisierte Immunanfärbung mit den TechMate™-Geräten verwendet Kapillarwirkung, um Reagenzien aus den Reagent Trays, Code-Nr. S 2039, aufzuziehen und Proben einzudecken, die auf speziellen Kapillarspalt-Mikroskopobjektträgern fixiert wurden. Nach einer vorher festgelegten Inkubationszeit werden die Reagenzien von den Proben durch Absorption in saugfähige Absorbent Pads, Code-Nr. S 2027 für TechMate™ 500/500 Plus/1000, und Code-Nr. S 2043 für TechMate™ Horizon, entfernt. Vor dem Anfärben mit den TechMate™-Geräten müssen nach Routineverfahren fixierte, paraffineingebettete Gewebeschnitte mit Hilfe von ChemMate™ Target Retrieval Solution, Code-Nr. S 2031, im Verhältnis 1:10 verdünnt, im Mikrowellengerät einer Epitopdemaskierung unterzogen werden. Es ist zu beachten, dass einige primäre Antikörper für eine optimale Färbung eine Vorbehandlung des Gewebes mit Proteinase K benötigen. Endogene Peroxidase muss mit der Peroxidase-Blocking Solution, Code-Nr. S 2023, blockiert werden. Aufgrund eines effektiven Waschverfahrens und des Vorhandenseins von Trägerproteinen in den DakoCytomation-Reagenzien sind zusätzliche Blockierungsschritte zur Reduzierung nicht spezifischer Hintergrundfärbung nicht erforderlich. Der ChemMate™ Buffer Kit, Code-Nr. K 5006, enthält Wash Buffers 1, 2 und 3 und Water Wash. Wash Buffer 1 wird während des ersten Teils der Färbeprozedur eingesetzt. Die Formulierung dieses Puffers trägt zur Blockierung einer unspezifischen Bindung des primären Antikörpers und des ChemMate™ DAKO EnVision™/HRP, Kaninchen/Maus (ENV)Nachweisreagenz an die Gewebeschnitte bei. Waschpuffer 2 und 3 sind identisch und werden für den restlichen Teil des Verfahrens verwendet. Da Waschpuffer 2 und 3 kein Natriumazid enthalten, werden sie insbesondere in Verbindung mit dem Waschen nach der Inkubation mit dem ENV-Nachweisreagenz angewendet. Water Wash wird für die Waschschritte am Ende der Färbungsprotokolle eingesetzt. Primäre Antikörper gehören nicht zum Lieferumfang des Kits. Es wird die Verwendung konzentrierter primärer Antikörper von DakoCytomation empfohlen. Siehe Richtwerte für optimale Verdünnungen in Tabelle 2. Das zum Kit gehörende ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV)Nachweisreagenz besteht aus einem Dextran-Rückgrat, an das eine große Zahl Peroxidase (HRP)-Moleküle und sekundäre Antikörpermoleküle gekoppelt wurden. Der Kopplungsreaktion liegt ein einzigartiger Chemismus zugrunde, der die Bindung von bis zu 100 HRP-Molekülen und bis zu 20 Antikörpermolekülen pro Rückgrat ermöglicht. Der sekundäre Antikörper, der an das Dextran-Rückgrat gekoppelt ist, wurde in Ziegen herangezogen. Er reagiert gleich gut mit Kaninchen- und Mausimmunglobulinen, so dass nur ein Nachweisreagenz für primäre Kaninchen- und Mausantikörper benötigt wird. Das ENV-Nachweisreagenz wird gebrauchsfertig in einer Tropfenzählerflasche geliefert. Das Substratsystem besteht aus zwei Komponenten, ChemMate™ DAB+ Chromogen, einer konzentrierten Diaminobenzidin (DAB)-Lösung und ChemMate™ Substrate Buffer mit Wasserstoffperoxid. Vor dem Gebrauch muss das DAB+-Chromogen in dem Substratpuffer verdünnt werden. Das Substratsystem erzeugt an der Stelle des Zielantigens eine braune Färbung. Reagenzienvorbereitung Substrate Working Solution (CHROM) Die DAB enthaltende Substrate Working Solution (CHROM) wird durch gründliches Mischen von 50 Teilen ChemMate™ Substrate Buffer (Flasche B) mit 1 Teil ChemMate™ DAB+ Chromogen (Flasche C) angesetzt. Nur das für die anzufärbende Anzahl Objektträger benötigte Volumen ansetzen. Siehe Tabelle 1 unten. Eine Kavität der Mikrotiterplatte entspricht zwei einander paarig zugeordneten Objektträgern. (104958-002) K 5007/EFG/HGW/26.09.03 p. 36/47 Probengewinnung und -vorbereitung Bei den Proben kann es sich um routinemäßig fixierte, paraffineingebettete Gewebeschnitte und Gefrierschnitte handeln. Es kann eine Vielfalt von Fixativen für die Probenkonservierung verwendet werden. Proben dürfen nicht länger als 24 Stunden und vorzugsweise über einen kürzeren Zeitraum in neutralem gepuffertem Formalin fixiert werden. Die optimale Dicke paraffineingebetteter Gewebeschnitte beträgt circa 4 µm. Die optimale Dicke von Gefrierschnitten beträgt circa 4-6 µm. Der Einschluss aller Proben muss auf ChemMate™ Capillary Gap Microscope Slides erfolgen und die Proben sind im Objektträgerhalter in Paaren mit zueinander weisenden Vorderseiten zu platzieren. Da die Objektträger bereits mit Haftmittel überzogen worden sind, wird von einer weiteren Behandlung der Objektträger abgeraten, weil sich dies nachteilig auswirken könnte. Die folgenden drei verschiedenen Typen von Kapillarspalt-Objektträgern sind erhältlich: 1. Objektträger mit grauen Bereichen, Auftragsdicke 75 µm. Sie sind für paraffineingebettete Gewebeschnitte bestimmt und müssen mit ähnlichen Objektträgern in Paaren verwendet werden. 2. Objektträger mit blauem Bereich, Auftragsdicke 100 µm. Sie sind für Gefrierschnitte und Abstriche bestimmt und müssen mit ähnlichen Objektträgern in Paaren verwendet werden. 3. Objektträger mit gelbem Bereich, Auftragsdicke 155 µm. Auf diese Objektträger dürfen keine Gewebeschnitte aufgebracht werden. Sie sind zur Verwendung für Gewebeschnitte bestimmt, die auf normalen Objektträgern ohne Kapillarspalt fixiert worden sind. Normalen Objektträger einfach mit dem gelben Objektträger zu einem Paar anordnen, um einen geeigneten Kapillarspalt zu erzeugen. HINWEIS: Normale Objektträger dürfen nicht breiter als 25 mm sein, damit sie in den Objektträgerhalter passen. Die Proben müssen auf dem Teil des Objektträgers fixiert werden, der sich am weitesten vom etikettierten Objektträgerende entfernt befindet. Es ist wichtig, dass Proben auf der gleichen Seite des Objektträgers platziert werden, auf der sich die mit Auftrag versehene Region befindet. Da die Gewebeschnitte während der Färbeprozedur mit aufeinander zuweisenden Vorderseiten angeordnet werden, müssen die Schnitte auf den Objektträgern so flach und faltenfrei wie möglich fixiert werden. Zu vielen Falten oder Klumpen hemmen die Kapillarwirkung und können zu widersprüchlichen Färberesultaten führen. Paraffinschnitte müssen aus einem vorgewärmten Wasserbad heraus fixiert werden, das destilliertes oder entionisiertes Wasser enthält. Das Wasserbad darf keine Zusätze (wie Gelatine, Poly-L-Lysin usw.) enthalten. Die Gewebeschnitte müssen durch Erwärmen getrocknet werden, normalerweise bei einer Temperatur von nicht mehr als 60°C über eine Mindestdauer von 60 Minuten (z.B. über Nacht). Um gutes Haften der Schnitte an den Objektträgern zu gewährleisten, ist es wichtig, vor der Trocknung im Mikrowellengerät das Wasser unter den Gewebeschnitten ablaufen zu lassen. Prozedur Vor Ausführen der Protokolle auf den DakoCytomation TechMate™-Geräten ist das Bedienerhandbuch für das spezifische TechMate™-Gerät sorgfältig durchzuarbeiten. Vor dem Einschalten des TechMate™-Geräts müssen die Objektträger für den Anfärbungsvorgang vorbereitet werden: Routinemäßig fixierte, paraffineingebettete Gewebeschnitte: Nach der Entparaffinierung und Rehydrierung in Puffer (Wasser) müssen die Gewebeschnitte einer Epitopdemaskierung im Mikrowellengerät unterzogen werden (siehe Abschnitt A unten). Zusätzlich zur Erwärmung in einem Mikrowellengerät erfordern einige Epitope eine kurze Enzymverdauung im TechMate™-Gerät. Siehe Tabelle 2 und die Anleitungen für ChemMate™ Proteinase K, Code-Nr. S 2019. Proben, die eine Enzymverdauung benötigen, müssen als Gruppe zusammengefasst werden. Wenn keine der Proben eine Enzymverdauung benötigt, wird Protokoll ENVP für alle Objektträger angewendet (dieses Protokoll erfordert eine Durchführungszeit von ungefähr 2 ¼ Stunden). Wenn einige Proben eine Enzymverdauung benötigt, wird Protokoll ENVPE für alle Objektträger angewendet (dieses Protokoll erfordert eine Durchführungszeit von ungefähr 2 ¼ Stunden). Bei den Proben, die keine Enzymverdauung benötigen, wird lediglich das proteolytische Enzym in der Kavität durch Proteinase K Diluent, Code-Nr. S 2032, oder Waschpuffer 2 oder 3 aus dem ChemMate™ Buffer Kit, Code-Nr. K 5006, ersetzt. A. Epitopdemaskierung im Mikrowellengerät Die im Folgenden gegebenen Anleitungen sollten sorgfältig befolgt werden, um eine reproduzierbare Epitopdemaskierung zu gewährleisten. Zum Standardisieren des Verfahrens wird empfohlen, den Objektträgerhalter auch dann vollständig mit Objektträgern zu beladen, wenn nur ein Objektträger eine Gewebeprobe enthält. Dies sorgt für eine identische Erwärmung der Gewebeschnitte bei jedem Durchgang. Bei Beendigung des Schritts der Epitopdemaskierung müssen die Objektträger mindestens 20 Minuten bei Raumtemperatur im Puffer belassen werden. Während dieses Abkühlens kann das TechMate™-Gerät für den Färbevorgang vorbereitet werden. Verfahren im Mikrowellengerät 1. Die entparaffinierten Gewebeschnitte in einem ChemMate™ Slide Holder, Code-Nr. S 2029 platzieren. Den ChemMate™ Slide Holder in einen ChemMate™ Incubation Container, CodeNr. S 2030 transferieren, der mit 200 ml, im Verhältnis von 1:10 verdünnter ChemMate™ Target Retrieval Solution, Code-Nr. S 2031, befüllt wurde. 2. Den Behälter mit dem Deckel angewinkelt (z.B. diagonal) in das Mikrowellengerät stellen. Ein 800-Watt-Mikrowellengerät auf höchster Stufe verwenden. Nachdem der Puffer zum Kochen gebracht wurde, weitere 5 Minuten lang im Mikrowellengerät erhitzen. 50 mL entionisiertes Wasser hinzugeben, um verdampftes Wasser zu ersetzen. Weitere 5 Minuten im Mikrowellengerät erhitzen. Den Behälter aus dem Mikrowellengerät entfernen und bei Raumtemperatur mindesten 20 Minuten abkühlen lassen. 3. Puffer gegen entionisiertes Wasser austauschen und kurze Zeit stehen lassen. Die Objektträger können jetzt in den TechMate™ Slide Holder eingesetzt werden. HINWEIS: Aufkochen des Puffers ist normal und erwünscht. Nicht alle 800-WattMikrowellengeräte haben die gleiche Wärmeübertragungsleistung. Daher muss das Mikrowellengerät zur Sicherstellung einer lokalen Standardisierung beim ersten Mal mit Objektträgern ohne Gewebeproben getestet werden, um die für ein 5 Minuten dauerndes tatsächliches Kochen erforderliche Gesamtverweilzeit im Mikrowellengerät festzustellen. Für Objektträgerpaare, die individuell mit (104958-002) K 5007/EFG/HGW/26.09.03 p. 40/47 Reagenzien behandelt werden sollten, z.B. primäre Antikörpern, wird empfohlen, anhand der Platten mit 10 Kavitäten ein Antibody Positioning Sheet (Lageplan der Antikörperanordnung) auszufüllen. Siehe Bedienerhandbuch. 1. Reagenzien gemäß Schema und Antibody Positioning Sheet in die Mikrotiterplatten mit 10 Kavitäten des TechMate™ Geräts einbringen. Darauf achten, die richtigen Reagenzien und Volumina einzufüllen. Besondere Sorgfalt ist beim Einfüllen der primären Antikörper geboten. 2. Vor dem Einsetzen der Objektträger in den Objektträgerhalter wird empfohlen, die Objektträger mit Waschpuffer 2 aus dem ChemMate™ Buffer Kit, Code-Nr. K 5006, vorzubefeuchten. Hierzu werden die in Paaren und nach dem Antibody Positioning Sheet angeordneten Objektträger einige Minuten lang in einer Mikrotiterplatte mit 10 Kavitäten belassen, die Waschpuffer 2 enthält. Dieses Vorbefeuchten gewährleistet einen angemessenen Flüssigkeitsstrom im Kapillarspalt. 3. Nach dem Vorbefeuchten die Objektträgerpaare gemäß dem Antibody Positioning Sheet in den TechMate™ Slide Holder setzen. Darauf achten, dass die Objektträger korrekt ausgerichtet sind. 4. Unmittelbar vor dem Testdurchgang den TechMate™ Slide Holder auf „Pad2“ in das TechMate™-Gerät setzen, um Waschpuffer 2 aus dem Kapillarspalt abzulassen. Das Ablaufenlassen der Objektträger sorgfältig beobachten, um zu prüfen, dass alle Objektträger richtig in Paaren angeordnet worden sind und die Ausrichtung der Objektträgerpaare korrekt vorgenommen wurde. 5. TechMate™ Slide Holder in den Waschpuffer (z.B. Waschpuffer 1 oder Waschpuffer 2) setzen, der im ersten Schritt des anzuwendenden Protokolls benutzt wurde, um die Kapillarspalten mit Waschpuffer zu füllen. 6. TechMate™ Slide Holder in die Position „HOME“ stellen, bevor der Testdurchgang gestartet wird. Siehe Kapitel 5 des Bedienerhandbuchs. Vor dem Anfärben unter Verwendung der Autostainer-Geräte müssen einige routinemäßig fixierte, paraffineingebettete Gewebeschnitte mithilfe der in Tabelle 3 angegebenen Target Retrieval Solution oder der Anleitung für den primären Antikörper im Mikrowellengerät einer Epitopdemaskierung unterzogen werden. Es sollte beachtet werden, dass einige primäre Antikörper für eine optimale Anfärbung die zusätzliche Proteinase K-Vorbehandlung des Gewebes benötigen. Endogene Peroxidase muss mit Peroxidase-Blocking Solution, Code-Nr. S 2023, blockiert werden. Aufgrund eines effektiven Waschverfahrens und dem Vorhandensein von Trägerproteinen in den DakoCytomation-Reagenzien sind zusätzliche Blockierungsschritte zur Reduzierung nicht spezifischer Hintergrundfärbung nicht erforderlich. Es wird DakoCytomation Wash Buffer, Concentrated x 10, for Immunocytochemistry, Code-Nr. S 3006, empfohlen. Primäre Antikörper gehören nicht zum Lieferumfang des Kits. Es wird die Verwendung konzentrierter Antikörper von DakoCytomation empfohlen. Siehe Richtwerte für optimale Verdünnungen in Tabelle 3. Das zum Kit gehörende ChemMate™ DAKO EnVision™/HRP, Rabbit/Mouse (ENV)Nachweisreagenz besteht aus einem Dextran-Rückgrat, an das eine große Zahl Peroxidase (HRP)-Moleküle und sekundäre Antikörpermoleküle gekoppelt wurden. Der Kopplungsreaktion liegt ein einzigartiger Chemismus zugrunde, der die Bindung von bis zu 100 HRP-Molekülen und bis zu 20 Antikörpermolekülen pro Rückgrat ermöglicht. Der sekundäre Antikörper, der an das Dextran-Rückgrat gekoppelt ist, wurde in Ziegen herangezogen. Er reagiert gleich gut mit Kaninchen- und Mausimmunglobulinen, so dass nur ein Nachweisreagenz für primäre Kaninchen- und Mausantikörper benötigt wird. Das ENV-Nachweisreagenz wird in einer Tropfenzählerflasche geliefert und muss vor dem Gebrauch in ein Autostainer Reagent Vial, Code-Nr. S 3425, gefüllt werden. Das Substratsystem besteht aus zwei Komponenten, ChemMate™ DAB+ Chromogen, einer konzentrierten Diaminobenzidin (DAB)-Lösung, und ChemMate™ Substrate Buffer mit Wasserstoffperoxid. Vor dem Gebrauch muss das DAB+-Chromogen in dem Substratpuffer verdünnt werden. Das Substratsystem erzeugt an der Stelle des Zielantigens eine braune Färbung. Die angesetzte Chromogenlösung muss vor dem Gebrauch in ein Autostainer Reagent Vial, Code-Nr. S 3425, gefüllt werden. Zur Gegenfärbung wird DakoCytomation Hematoxylin, Code-Nr. S 3301, empfohlen, da es eine eindeutige blaue Anfärbung des Zellkerns erbringt. Die Schnitte müssen so flach und faltenfrei wie möglich auf den Objektträgern aufgebracht werden. Zu viele Falten beeinträchtigen die Anfärbungsergebnisse. (104958-002) K 5007/EFG/HGW/26.09.03 p. 43/47 HINWEIS: Objektträger dürfen nicht breiter als 26 mm sein, damit sie in den Objektträgerhalter passen. Paraffinschnitte müssen aus einem vorgewärmten Wasserbad heraus fixiert werden, das destilliertes oder entionisiertes Wasser enthält. Das Wasserbad darf keine Zusätze (wie Gelatine, Poly-L-Lysin usw.) enthalten. Die Gewebeschnitte müssen durch Erwärmen getrocknet werden, normalerweise bei einer Temperatur von nicht mehr als 60 °C über eine Mindestdauer von 60 Minuten (z.B. über Nacht). Um gutes Haften der Schnitte an den Objektträgern zu gewährleisten, ist es wichtig, vor der Trocknung im Mikrowellengerät das Wasser unter den Gewebeschnitten ablaufen zu lassen. Prozedur Bevor Färbeverfahren nach der Template der DakoCytomation Autostainer-Geräte durchgeführt werden, ist das Bedienerhandbuch für das jeweilige Autostainer-Gerät sorgfältig durchzuarbeiten. Vor dem Einschalten des Autostainer-Geräts müssen Objektträger und Reagenzien gemäß des Slide Layout Map (Lageplan der Objektträgeranordnung) und des Reagent Layout Map (Lageplan der Reagenzienanordnung) angeordnet werden. Routinemäßig fixierte, paraffineingebettete Gewebeschnitte Nach der Entparaffinierung und Hydratrion in Puffer (Wasser) müssen die Gewebeschnitte einer Epitopdemaskierung im Mikrowellengerät unterzogen werden (siehe Abschnitt A unten). Einige Epitope tolerieren die Behandlung im Mikrowellengerät nicht. Siehe Anleitungen für die jeweiligen primären Antikörper von DakoCytomation. Zusätzlich zur Erwärmung in einem Mikrowellengerät erfordern einige Epitope eine kurze Enzymverdauung im Autostainer-Gerät. Siehe Tabelle 3 und die Anleitungen für ChemMate™ Proteinase K, Code-Nr. Siehe auch Bedienerhandbuch. Vor dem Einsetzen der Objektträger gemäß der Slides Layout Map wird empfohlen, die Objektträger 5 Minuten lang in DakoCytomation Wash Buffer, Code-Nr. S 3006, zu benetzen. Die Reagenzien in die Autostainer Reagent Vials, Code-Nr. S 3425, einfüllen und die Fläschchen gemäß der Reagent Layout Map im Reagenziengestell anordnen. Darauf achten, die richtigen Reagenzien und Volumina einzufüllen. Besondere Sorgfalt ist beim Befüllen der Fläschchen mit primärem Antikörper geboten. Wasser- und Pufferpumpen anlaufen lassen.
