Résumé du projet (15 lignes maximum) : Les NADPH oxydases (NOXs) constituent une famille d’enzymes dont l’unique fonction est de générer les formes réactives de l’oxygène (FRO/ROS). Ces molécules, lorsqu’elles sont produites de façon contrôlée, régulent de nombreuses fonctions biologiques parmi lesquelles la défense anti-infectieuse de l’hôte. Cependant, du fait de leur haute cytotoxicité, une production excessive ou inappropriée de ROS peut entraîner des lésions tissulaires sévères conduisant au développement de nombreuses pathologies inflammatoires. NOX1, abondamment exprimée dans les cellules épithéliales du colon, serait impliquée dans la défense immunitaire innée de la muqueuse intestinale. Cependant, son dysfonctionnement, pourrait contribuer à la genèse des maladies inflammatoires chroniques de l’intestin (MICI). En effet au cours des MICI, une absence de la résolution de la réponse inflammatoire est associée à une production élevée de ROS et à une synthèse accrue de cytokines, parmi lesquelles les cytokines de type Th17 (IL17, IL22) et Treg (TGF , IL10). Nous avons récemment montré que NOX1 était modulée positivement par l’IL-17 et négativement par le TGF dans les cellules épithéliales du colon. Par conséquent, NOX1 pourrait être un effecteur central en aval de ces cytokines et jouer un rôle clé dans le développement et la progression des MICI. Dans ce contexte, les objectifs du projet seront de : 1) décrypter les mécanismes par lesquels ces cytokines régulent NOX1 2) définir les processus par lesquels NOX1 contribue à la genèse de la pathologie 3) compléter cette étude chez l’homme par l’analyse des biopsies issues de patients atteints de MICI et de patients sains. Title of the project: Role of NOX1 from epithelial cells in the development of inflammatory disorders Summary of the project (15 lines maximum): NADPH oxidases (or NOXs) are a unique family of enzymes dedicated to the production of reactive oxygen species (ROS). When produced under controlled conditions, ROS are important mediators of various biological functions, including host defence against pathogens. However, as ROS are highly cytotoxic molecules, their inappropriate or excessive production may induce the damage of surrounding tissues, thereby contributing to inflammatory disorders. NOX1 is abundantly expressed in colon epithelial cells, where they could be involved in mucosal innate immunity in physiological conditions. However, dysfunction of NOX1 could be linked to pathological situations in the colon such as inflammatory Bowel Diseases (IBD). Indeed, during IBD, the lack of resolution of the inflammatory response is associated with high production of ROS and high synthesis of cytokines, including Th17 (IL17, IL22) and Treg (TGF , IL10) cytokines. We recently showed that NOX1 was positively regulated by IL17 and negatively by TGF in intestinal epithelial cells. Therefore, NOX1 might be a central effector downstream of these cytokines, and might play a key role in the development and progression of MICI. In this context, the objectives of the thesis will be: 1) to decipher the mechanisms through which Th17 and Treg regulate NOX1. 2) To define how NOX1 could contribute to the genesis of MICI. 3) To complete this study in human by analyzing biopsies from patients with MICI and healthy patients.
