Technical Information Buffer preparation Coating buffer: 1 or 2 tablets provided in the complete kits. Available also separately in bulk quantities (Art. No. 110110). One tablet dissolved in 100 ml of double distilled water results in 50 mM carbonate-bicarbonate buffer (pH 9.6) containing 0.02 % NaN3. Washing buffer: 1 or 2 pouches at 50 g (Art. No. 110152) provided in the complete kits. Available also separately in bulk quantities (Art. No. 110153/110154). Dissolve the content of one pouch in 5000 ml of double distilled water. The resulting buffer will be a 10 mM phosphate buffer (pH 7.4) containing 140 mM NaCl, 3 mM KCl, and 0.05 % Tween 20 (PBST). Contains no NaN3! Use within 2 days or add a preservative (e.g. sodium azide) to prevent microbial growth. The content of the pouch can also be dissolved in 500 ml of water to obtain a 10x stock solution, which is stable at room temperature. Dilute this stock solution 10x before use. Extraction buffer «General»: 250 ml or 500 ml concentrate (10x) provided in the complete kits. Available also separately (Art. No. 110120). 100 ml of 10x concentrate made up with double distilled water to 1000 ml results in 20 mM Tris buffer (pH 7.4 at 25°C) containing 137 mM NaCl, 3 mM KCl, 2 % PVP 24kD, 0.05 % Tween 20 and 0.02 % NaN3. Colored with E102 and E131. Extraction buffer «Bulbs & Tubers»: 25 or 50 g powdered egg albumin provided in the complete kit. Available also separately as set (Art. No. 110122). To prepare one liter of extraction buffer «Bulbs & Tubers», first dissolve 10 g of egg albumin in 20 ml with a spatula in a small cup, then make up to 1000 ml with extraction buffer «General». Prepare freshly before use! Extraction buffer «Grapevine»: 500 ml or 1000 ml concentrate (5x) provided in the complete kits. Available also separately (Art. No. 110123). 200 ml of 5x concentrate made up with double distilled water to 1000 ml results in 200 mM Tris buffer (pH 8.2 at 25°C) containing 137 mM NaCl, 2 % PVP 24kD, 1 % PEG 6kD, 0.05 % Tween 20 and 0.02 % NaN3. Colored with E102 and E127. Note: During storage of the concentrate, but also of the diluted buffer, the formation of crystals can be observed causing turbidity or even a slight precipitation. However, the turbidity or precipitation will not affect the quality and performance of the buffer. Anyway, in most cases the crystals will go in solution when the buffer has warmed up. Extraction buffer «Caulimo»: 250 ml or 500 ml concentrate (10x) packed with powder of egg albumin and urea as package provided with the complete kits. Available also separately (Art. No. 110124). Details see on the Product Information «Cauliflower mosaic virus». Conjugate buffer: 10 ml or 20 ml concentrate (10x) provided in the complete kits. Available also separately (Art. No. 110140/110142). 10 ml of 10x concentrate made up with double distilled water to 100 ml results in 20 mM Tris buffer (pH 7.4 at 25°C) containing 137 mM NaCl, 3 mM KCl, 1 mM MgCl2, 2 % PVP 24kD, 0.05 % Tween 20, 0.2 % BSA, and 0.02 % NaN3. Colored with E127 and E131. Substrate buffer: 20 ml or 2 x 20 ml substrate buffer (5x) provided in the complete kits. Available also separately (Art.No. 110130/110132). 20 ml of 5 x concentrate made up with double distilled water to 100 ml results in 1 M diethanolamine pH 9.8, containing 0.02 % NaN3. Add one pNPP tablet (20 mg) per 20 ml buffer 15 min. before use. Substrate (pNPP tablets): 5 or 10 single packed tablets containing each 20 mg of pNPP (p-nitro-phenyl-phosphate) provided in the complete kits. Available also separately (Art. No. 110080/110180). For long-term storage, keep the substrate at -20°C! Open the blister pack without touching the content and add one tablet per 20 ml of substrate buffer to obtain a solution of 1 mg/ml. Prepare freshly before use! Health hazard To prevent microbial growth, the buffers contain 0.02 - 0.2 % sodium azide which is toxic when swallowed and irritant if it comes in contact with skin, eyes and mucosae; nevertheless, these products are not considered to present a hazard to health when used in accordance to the principles of Good Laboratory Practice and Good Standards of Occupational Hygiene. Disponible aussi individuellement (Réf. 110110). Dissoudre une tablette dans 100 ml d’eau distillée pour obtenir un tampon bicarbonate (50mM carbonate, pH 9.6) contenant 0.02 % de NaN3. Tampon de lavage: Les kits contiennent des sachets de 50 g en poudre (Réf. 110152). Dissoudre un sachet dans 5000 ml pour obtenir un tampon phosphate (10 mM carbonate, pH 7.4) contenant 140 mM NaCl, 3 mM KCl et 0.05 % de Tween 20 (PBST). Ne contiens pas de NaN3. A utiliser dans les deux jours après reconstitution (ajouter un conservateur – azide de sodium- pour éviter les contaminations microbiennes au delà de 2 jours). Le contenu d’un sachet peut aussi être dissous dans 500 ml pour obtenir une solution concentrée 10 fois et stable à température ambiante 15-25°C. Cette solution doit être diluée x10 juste avant utilisation. Tampon d’extraction «general»: 250 ml ou 500 ml concentré 10 fois (x10) fournis dans les kits complets. Disponible aussi individuellement (Réf. 110120). 100 ml de solution concentrée x10 sont ajoutés à 900 ml d’eau distillée pour former 1000 ml de tampon Tris (20 mM, pH 7.4) contenant 137 mM NaCl, 3 mM KCl, 2 % PVP 24kD, 0.05 % Tween 20 et 0.02 % NaN3. Colorée en vert avec E102 et E131. Tampon d’extraction spécial «bulbes et tubercules»: 25 g ou 50 g d’albumine d’œuf en poudre fournie dans les kits complets. Disponible aussi individuellement (Réf. 110122). Pour préparer 1 litre de tampon, dissoudre 10 g d’albumine d’œuf en poudre dans 20 ml de tampon. Compléter ensuite jusqu’à 1000 ml avec du tampon d’extraction «général». A utiliser juste après la préparation! Tampon d’extraction spécial «Caulimo»: Voir Information produit «Cauliflower mosaic virus». Tampon d’extraction spécial «vigne»: 500 ml ou 1000 ml concentré 5 fois (x5) fournis dans les kits complets. Disponible aussi individuellement (Réf. 110123). 200 ml de solution x5 sont ajoutés à 800 ml d’eau distillée pour former 1000 ml de tampon Tris (200 mM, pH 8.2 à 25°C) contenant 137 mM NaCl, 2 % PVP 24 kD, 1 % PEG 6kD, 0.05 % Tween 20 et 0.02 % NaN3. Colorée en orangé avec E102 et E127. A noter: La formation d’un léger précipité peut apparaître au cours de la conservation. Il n’affecte pas la qualité et les performances du tampon, et disparaît en général quand celui-ci se réchauffe. Tampon de conjugué: 10 ml ou 20 ml concentré 10 fois (x10) fournis dans les kits complets. Disponible aussi individuellement (Réf. 110140/110142). 10 ml de solution concentrée x10 sont ajoutés à 90 ml d’eau distillée pour former 100 ml de tampon Tris (20 mM, pH 7.4) contenant 137 mM NaCl, 3 mM KCl, 1 mM MgCl2, 2 % PVP 24kD, 0.05 % Tween 20, 0.2 % BSA et 0.02 % de NaN3. Colorée en violet rosé avec E127 et E131. Tampon de substrat: 20 ml ou 2 x 20 ml concentré 5 fois (x5) fournis dans les kits complets. Disponible aussi individuellement (Réf. 110130/110132). 20 ml de solution concentrée x5 sont ajoutés à 80 ml d’eau distillée pour former 100 ml de tampon diethanolamine (1M, pH 9.8) contenant 0.02 % de NaN3. Ajouter 1 tablette de pNPP (20 mg) dans 20 ml de tampon de substrat 15 minutes avant utilisation. Tablettes de substrat (pNPP): 5 ou 10 tablettes conditionnées individuellement contenant chacune 20 mg de pNPP (para Nitro-Phhenyl-Phosphate) fournis dans les kits complets mais aussi disponible séparément (110080/110180). Pour une meilleure conservation stocker à –20° C. Ouvrir l’emballage individuel sans toucher la pastille et la faire dissoudre dans 20 ml de tampon substrat reconstitué pour obtenir une solution concentrée à 1 mg/ml. A utiliser juste après la préparation. Risques pour la santé L’azide de sodium NaN3 est un conservateur utilisé dans certains de nos tampons. C’est un produit qui peut être toxique s’il est ingéré ou mis au contact de la peau, des yeux et des muqueuses. Cependant, ce produit ne présente pas de danger pour la santé ou l’environnement s’il est utilisé conformément aux règles de bonnes pratiques de laboratoire. Dieses Produkt ist auch separat als Bulkware (Art. Nr. 110110) erhältlich. Eine Tablette aufgelöst in 100 ml doppelt destilliertem Wasser ergibt einen 50 mM Carbonat-Bicarbonat Puffer pH 9.6 mit 0.02 % NaN3. Waschpuffer: Im «Kompletten Kit 480 bzw. 960» sind 1 bzw. 2 Puffer-Beutel à 50 g enthalten. Der Artikel ist auch separat erhältlich (Art. Nr. 110152) sowie als Bulkware zu 250 g (Art. Nr. 110153) und zu 1250 g (Art. Nr. 110154). Das Pulver in 5000 ml doppelt destilliertem Wasser aufgelöst, ergibt einen Phosphatpuffer mit 140 mM NaCl, 3 mM KCl und 0.05 % Tween 20 (PBST) pH 7.4. Bitte beachten, dass der Puffer kein NaN3 enthält! Der Waschpuffer sollte deshalb innerhalb von 2 Tagen aufgebraucht werden. Zu Lösungen welche länger als zwei Tage in Gebrauch sind, sollte ein Konservierungsmittel (z.B. NaN3) zugefügt werden, um Keimwachstum zu verhindern. Um eine 10-fach konzentrierte Pufferlösung (10x) zu erhalten, wird der Inhalt des Beutels in 500 ml Wasser aufgelöst. Das Konzentrat sollte bei Raumtemperatur aufbewahrt und vor Gebrauch 10-fach verdünnt werden. Extraktionspuffer «General»: Im «Kompletten Kit 480 bzw. 960» sind 250 ml bzw. 500 ml 10-fach konzentrierte (10x) Pufferlösung enthalten. Das Produkt ist auch separat erhältlich (Art. Nr. 110120). 100 ml des 10-fach Konzentrates werden mit 900 ml doppelt destilliertem Wasser verdünnt. Die verdünnte Lösung ist ein 20 mM Tris–Puffer mit 137 mM NaCl, 3 mM KCl, 2 % PVP 24 kD, 0.05 % Tween 20 und 0.02 % NaN3, pH 7.4 (bei 25°C). Die Pufferlösung ist mit E102 and E131 grün gefärbt. Extraktionspuffer «Zwiebeln und Knollen»: Im «Kompletten Kit 480 bzw. 960» sind 25 bzw. 50 g Ovalbumin enthalten. Das Produkt ist auch separat erhältlich (Art. Nr. 110122). 10 g Ovalbumin werden mit 20 ml Wasser in einem kleinen Gefäss verrührt und danach mit einem Liter Extraktionspuffer «General» versetzt. Der Puffer sollte immer kurz vor Gebrauch frisch zubereitet werden. Extraktionspuffer «Weinreben»: Im «Kompletten Kit 480 bzw. 960» sind 500 ml bzw. 1000 ml 5-fach konzentrierte (5x) Pufferlösung enthalten. Das Produkt ist auch separat erhältlich (Art. Nr. 110123). 200 ml des 5-fachen Konzentrates mit 800 ml doppelt destilliertem Wasser verdünnt ergibt einen 200 mM Tris Puffer mit 137 mM NaCl, 2 % PVP 24kD, 1 % PEG 6 kD, 0.05 % Tween 20 und 0.02 % NaN3, pH 8.2 (bei 25°C). Die Pufferlösung ist mit E102 and E127 orange gefärbt. Achtung: Während der Lagerung des konzentrierten Puffers können sich Kristalle bilden, d.h. der Puffer erscheint dann trübe. Diese Bildung von Kristallen hat keinen Einfluss auf die Qualität oder Funktion des Puffers. In den meisten Fällen lösen sich die Kristalle bei Erwärmen des Puffers auf. Extraktionspuffer «Caulimo»: Für die Bestimmung von CaMV. Details siehe unter der Produktinformation «Cauliflower mosaic virus». Konjugatpuffer: Im «Kompletten Kit 480 bzw. 960» sind 10 ml bzw. 20 ml 10-fach konzentrierte (10x) Pufferlösung enthalten. Das Produkt ist auch separat erhältlich (Art. Nr. 110140/110142). 10 ml des 10-fach Konzentrates mit 90 ml doppelt destilliertem Wasser verdünnt ergibt einen 20 mM Tris Puffer mit 137 mM NaCl, 3 mM KCl, 1 mM MgCl2, 2 % PVP 24 kD, 0.05 % Tween 20, 0.2 % BSA und 0.02 % NaN3, pH 7.4 (bei 25°C). Die Pufferlösung ist mit E127 und E131 violett gefärbt. Substratpuffer: Im «Kompletten Kit 480 bzw. 960» sind 20 ml bzw. 2 x 20 ml 5-fach konzentrierte (5x) Pufferlösung enthalten. Das Produkt ist auch separat erhältlich (Art.Nr. 110130/110132). 20 ml des 5-fachen Konzentrates mit 80 ml destilliertem Wasser verdünnt ergibt einen 1 M DiethanolaminePuffer mit 0.02 % NaN3, pH 9.8. Die Substratlösung wird hergestellt, indem eine pNPP Tablette à 20 mg 15 Min. vor Gebrauch in 20 ml Substratpuffer aufgelöst wird. Substrat-Tabletten (pNPP-Tabletten): Im «Kompletten Kit 480 bzw. 960» sind 5 bzw. 10 einzeln verpackte pNPP (paraNitrophenylphosphat) Tabletten à 20 mg enthalten. Die Tabletten sind auch separat erhältlich (Art. Nr. 110080/110180 ). Die Tabletten sollten für eine längerfristige Lagerung bei -20° C aufbewahrt werden! Herstellung der gepufferten Substratlösung: Verpackung öffnen, ohne den Inhalt (Tablette) zu berühren und in 20 ml Substrat-Puffer lösen. Die so erhaltene Substratlösung hat eine pNPP-Endkonzentration von 1 mg/ml. Die Lösung sollte immer kurz vor Gebrauch frisch zubereitet werden. Gesundheitsgefährdung Die meisten Puffer enthalten 0.02 - 0.2 % Natriumazid (NaN3) als Konservierungsmittel, welches beim Verschlucken zu Vergiftungen oder bei Haut-, Augen- und Schleimhautkontakt zu Irritationen führen kann. Die Produkte sind jedoch nicht gesundheitsgefährdend, sofern die Prinzipien der «Guten Laborpraxis» und der «Vorschrift der Arbeitsplatzhygiene» beachtet und eingehalten werden.
