SOUGAKOFF Wladimir, PhD MCU-PH / Associate Professor of Molecular Biology Chef d'Equipe / Team Leader Emergence et propagation des multi-résistances aux antibiotiques / Emergence and diffusion of Multiple Resistance against Antibiotics Wladimir Sougakoff a obtenu un doctorat en biochimie, option microbiologie, de l’Université Pierre et Marie Curie (UPMC, Paris, France). Il est diplômé de l'Institut Pasteur de Paris en microbiologie, option bactériologie. Il a complété sa formation par un post doc dans le département « Antibiotiques » chez Hoffmann-La Roche à Bâle (Suisse). Il a obtenu son habilitation à diriger des recherches (HDR) à l'UPMC en 2002. Il est responsable d'un laboratoire hospitalier de routine qui met en oeuvre des techniques de biologie moléculaire pour l'identification et la détection de la résistance aux antibiotiques à partir d’isolats cliniques. Ses centres d’intérêt en recherche fondamentale portent sur l’étude des mécanismes moléculaires de résistance des bactéries aux antibiotiques, et sur l’épidémiologie moléculaire des bactéries multi-résistantes. Wladimir Sougakoff obtained his Ph.D. degree in biochemistry, option microbiology from UPMC, Paris, France. He received his degree in microbiology, option bacteriology, from Institut Pasteur, Paris, France. He was a research fellow at the Antibiotics department at Hoffmann-La Roche in Basel, Switzerland, from 1990 to 1992. He was habilitated as Research Director (HDR) at UPMC in 2002. He is in charge of a hospital laboratory which implements routine molecular biology techniques for the identification and detection of resistance to antibiotics in clinical isolates. His research interests is in the molecular mechanisms involved in resistance to antibacterial drugs and molecular epidemiology of multidrug-resistant bacteria. Multi-résistance chromosomique chez Mycobacterium tuberculosis (MDR TB) Les objectifs de cette recherche sont (i) le décryptage des mécanismes de résistance aux antibiotiques en étudiant au niveau du génome les mutations impliquées dans la résistance aux médicaments anti-tuberculeux; (ii) l'étude des liens entre résistance aux antibiotiques et évolution phylogénétique des souches MDR; (iii) la caractérisation biochimique et structurelle de complexes enzymatiques d'intérêt pour le développement de nouveaux antituberculeux (ATP synthase). (iv) l'évaluation de l'impact de l'état immunitaire de l'hôte sur le développement et la diversité de mutants résistants sélectionnés in vivo sous traitement antiTB. Ces activités de recherche viennent en support des activités cliniques et de diagnostic du Centre National de Référence des Mycobactéries et de la Résistance des Mycobactéries aux Antituberculeux (CNR-MyRMA) auquel je participe dans le cadre hospitalier. Mécanismes moléculaires liés à la la propagation des résistances plasmidiques Cette recherche vise à mieux comprendre les mécanismes moléculaires contribuant à la propagation de plasmides portant plusieurs gènes de résistance chez les entérobactéries, en ciblant deux agents pathogènes humains majeurs: Escherichia coli et Klebsiella pneumoniae. Les objectifs spécifiques sont: (i) analyser des séquences de plasmides de résistance dans ces deux espèces, notamment les fonctions impliquées dans la dissémination et le maintien de ces plasmides multi-résistants dans l'hôte bactérien. (ii) évaluer l'apport des systèmes de partition et des systèmes toxine-antitoxine sur la capacité de diffusion de plasmides MDR présentant une gamme exceptionnellement étendue de bactéries hôtes. As a whole, my research program is intended to increase our understanding of the molecular mechanisms of development and diffusion of multiple drug resistance in Mycobacterium tuberculosis (chromosome model) and in Enterobacteriaceae (plasmid model). Chromosome-born multidrug resistance (MDR) in M.tuberculosis (Mtb) Our main objectives are: (i) To decipher the drug-resistance machinery in Mtb by studying at the genome level the variety of mutations involved in resistance to anti-tuberculous drugs. (ii) to study the links between drug-resistance spreading and phylogenic lineages of Mtb. (iii) to investigate at biochemical and structural levels M. tuberculosis macromolecular complexes of interest for the development of new drugs (ATP synthase). (iv) to evaluate the impact of the immune status of the host on the development and diversity of resistant mutants selected in vivo under antiTB therapy. My research interests parallel my clinical focus on anti-mycobacterial resistance performed in the frame of the National Reference Centre for Mycobacteria and Anti-mycobacterial Resistance (CNRMyRMA). Molecular mechanisms contributing to the spread of plasmids carrying multiple resistance genes The second part of our project aims at investigating the molecular mechanisms contributing to the spread of plasmids carrying multiple resistance genes in Enterobacteriaceae, with a specific focus on the two major human pathogens, Escherichia coli and Klebsiella pneumonia: (i) By studying plasmid sequences in these two species to increase our understanding of the functions involved in the spreading and the maintenance of multi-resistant plasmids in the bacterial host. (ii) By evaluating the contribution of partition systems and toxin-antitoxin systems on the diffusion capacity of MDR plasmids showing an unusually extended bacterial host range.
