Ces agrégats se propagent de cellules en cellules, mais les mécanismes impliqués sont mal connus. Cette propagation implique plusieurs étapes, parmi lesquelles la liaison d’agrégats présents dans le milieu extracellulaire à la membrane des neurones. L’objectif de cette thèse est dans un premier temps de déterminer les bases moléculaires et cellulaires de la liaison des agrégats à la membrane, afin de pouvoir ensuite développer des molécules capables d’empêcher cette étape cruciale dans le développement de la maladie. Pour cela on 1/ documentera les propriétés de surface des agrégats qui leur confèrent leur capacité de liaison (taille, morphologie, charge, hydrophobicité, etc) ; 2/ identifiera leurs partenaires de la membrane cellulaire ; 3/ utilisera l’ensemble des connaissances ainsi acquises pour développer de façon rationnelle des molécules capables d’empêcher la liaison des agrégats à la membrane. Ces études seront réalisées sur deux modèles complémentaires : les agrégats d’ -synucléine et de huntingtine, responsables respectivement des maladies de Parkinson et de Huntington. Ce projet multidisciplinaire fait appel à des techniques de biochimie des protéines, protéomique et biochimie cellulaire. Title of the project: BINDING OF AMYLOID FIBRILS TO THE CELLULAR MEMBRANE AND HOW TO PREVENT IT Summary of the project (15 lines maximum): The presence of fibrillar protein aggregates is the hallmark of more than 20 neurodegenerative diseases, including Alzheimer’s, Parkinson’s and Huntington’s diseases. It is established that these aggregates that are toxic to neuronal cells propagate from cell to cell. The molecular mechanisms underlying this phenomenon are subject of intense debate. The objectif of this PhD thesis is to understand one of the crucial step of this vicious cycle, namely the binding of these aggregates to the cellular membrane, in order to develop molecules that will specifically inhibit binding. We will 1/ document the surface properties of the aggregates responsible for their binding capacities (size, morphology, net charge, hydrophobicity, etc); 2/ identify their binding partners at the membrane; and finally 3/ use the accumulated knowledge to rationally design molecules that interfere with binding. We will use as models two complementary types of aggregates, -synuclein and huntingtin fibrils, respectively responsible for Parkinson’s and Huntington’s diseases. This is a highly multidisciplinary project, involving cellular biochemistry, protein chemistry and proteomic approaches. 3 Connaissances et compétences requises pour la thèse / Knowledge and skills required for the thesis L’étudiant devra impérativement maîtriser les techniques de biochimie cellulaire (culture de cellules eucaryote, immunoprécipitation, tri cellulaire, …) et/ou de biochimie des protéines (production, purification et caractérisation de protéines recombinantes, spectrométrie de masse). De part le mode de sélection de l’Ecole Doctorale, une moyenne de M2 d’au moins 14/20 est exigée. Le candidat bénéficiera du savoir-faire d’une équipe multidisciplinaire, ainsi que des différentes collaboration nationales et internationales de l’équipe. The applicant must be familiar with cellular biochemistry (cell culture, pull-down, cell sorting, microscopie, etc) and/or protein chemistry (production, purification and characterization of recombinant proteins, mass spectrometry) techniques. Due to the competitive scholarship examination modalities, a master’s degree average grade of at least 14/20 is mandatory. The applicant will benefit from the expertise of a mutlidisciplinary team as weel as from its many national and multinational collaborations.
