When metastases are present, prognosis is highly reduced because current chemo, radio and targeted therapies are not efficient. Nonetheless, novel immune checkpoint blockers appear as promising tools for restoring anticancer surveillance, although side effects are severe. Most melanomas are sporadic, up to 8% are familial. Melanoma affects not only humans but also animals like horses, dogs, or minipigs. While solar and sunbed UV radiation exposure is the best known causative agent in human melanomas, animals models of melanoma most frequently carry a genetic predisposition. Among spontaneous mammalian models, MeLiM minipigs are particularly interesting because their skin and melanomas are structurally similar to human’s and because melanomas spontaneously regress. Melanomas develop during the perinatal period and can metastasise. Eventually they regress from the third month of age onwards. The immune system seems to be successfully involved in this phenomenon. Pathways involved in melanoma progression are not well characterized (Egidy et al 2008). Preliminary results point at pathways also found activated in human melanomas, although in the absence of the most frequent human’s mutations. These mutations mainly affect BRAF and NRAS oncogenes, or NF1 tumor suppressor leading to the constitutive activation of MAPK or MAPK and mTOR pathways, respectively (TCGA, 2015). MeLiM melanocytic lesions are quite big in size. MeLiM melanomas display abundant proliferation marks together with cell cycle arrest marks. These data may suggest a spatial cell heterogeneity in proliferative capacity or alternatively, a cell autonomous arrest that could add to the belated immune effect. We also work with the mouse melanoma model Tyr::NRasQ61K, Cdkn2a-/-, Pax3GFP/+ which develops cutaneous melanoma later in life (Campagne et al 2016). In this model, melanocytes carry the constitutively active NRas protein which drives skin hyperpigmentation due to the proliferative effect of the mutation, together with the knock out of the tumor suppressor Cdkn2a locus, and the fluorescent melanocytic reporter allele. Yet additional changes are needed to drive melanocytic transformation. Melanomas in this model often produce metastasis to other organs, they show abundant proliferation marks but few cell cycle arrest marks. Cancer regression is not obvious in this model. Aims: We wish 1) to determine whether proliferation and cell cycle arrest marks are colocalised in melanoma cells in MeLiM melanomas sampled at different ages, 2) to establish the proportion of single or Master Biologie Santé Paris Saclay Proposition de stage 2016-2017 double labeled cells at different ages, and 3) to analyse the signaling pathways associated with the groups. Methods: Fresh melanoma cells obtained from slices of excised lesions will be isolated and characterized by flow cytometry for cell type and cell cycle markers. Alternative slices will be saved for immunohistological examination. Pharmacological treatments of primary cultures will help establish the active signaling pathways. Additional signs of proliferation, quiescence, and arrest / senescence will be analysed. Mouse Tyr::NRasQ61K, Cdkn2a-/-, Pax3GFP/+ melanoma cells will be used as controls. Applicants will work with fresh clinical samples from the 2 animal models to be used for in situ and in vitro characterisation. They will practise tissue culture techniques, antibody powered strategies, from cytometry to structured illumination microscopy, image processing.
