Eng. 51(12), 123601 (2012) MCF, Laboratoire SOL (UBO) 3 Titre Contexte Objectifs identifiés Caractère novateur Applications biomédicales en imagerie optique linéaire et non-linéaire Le domaine de l’optique biomédicale et plus généralement de la biophotonique connaît un essor exceptionnel depuis une dizaine d’années. En témoigne par exemple le fait que le marché de la biophotonique dans le monde a dépassé celui des télécommunications optiques. C’est dans ce contexte à la fois dynamique et concurrentiel que le Laboratoire de Spectrométrie et Optique Laser développe des méthodes d’imagerie optique innovantes en vue d’applications biomédicales, en partenariat avec des entreprises (société EVOSENS à Plouzané et PERFOS à Lannion pour le développement d’un endo-microscope biphoton). Le LSOL est notamment reconnu dans le domaine de la polarimétrie linéaire (polarimétrie dite de Mueller) et nonlinéaire (par microscopie de génération de seconde harmonique). Le laboratoire a récemment développé des approches originales permettant de mieux exploiter les potentialités de ces techniques en termes de rapidité et de robustesse : M. Dubreuil et al, Optics Express, 15, 13660 (2007) ; C. Odin et al, Optics Express 16, 16151 (2008). L’objectif de la thèse est le développement couplé et la démonstration de complémentarité de méthodes d’imagerie linéaire et non-linéaire en vue d’applications au diagnostic biomédical ex situ (sur biopsies) en visant à terme l’imagerie endomicroscopique in situ (via une fibre optique). Une première étape du travail consistera à mettre au point un polarimètre de Mueller instantané prototype basé sur la technologie de tomographie de cohérence optique spectrale (spectral OCT), c’est-àdire utilisant une source de lumière modulée spectralement, une fibre optique et une analyse de Fourier rapide du signal. Une seconde étape sera d’implémenter le dispositif au sein d’un microscope multiphotonique développé au LSOL de manière à coupler les modes d’imagerie polarimétrique linéaire et non-linéaire au sein d’un même instrument. La dernière étape de la thèse sera consacrée à l’imagerie bimodale sur des échantillons biologiques (biopsies, peau, muscles, glandes salivaires…). Le projet est lancé au moment opportun, car tous les composants clés (optique, électronique, traitement du signal) sont disponibles commercialement depuis peu. En outre le site de l’UBO regroupe toutes les compétences en sciences physiques et en sciences du vivant pour créer la synergie nécessaire à l’aboutissement de ce travail de recherche. Le LSOL a récemment montré l’intérêt de coupler les approches d’optique linéaire et non-linéaire en vue du diagnostic de la fibrose du foie sur biopsie (M. Dubreuil et al “Mueller matrix polarimetry for improved liver fibrosis diagnosis”, Optics Letters 37, 1061-1063, 2012). L’étape nouvelle envisagée dans cette thèse est le développement de ces approches à la fois d’un point de vue technique pour satisfaire aux contraintes de rapidité et de souplesse imposées en imagerie biomédicale et d’un point de vue applicatif pour étendre les études à d’autres problématiques biomédicales pour lesquelles le LSOL a engagé des collaborations qui sont soutenues par l’UBO (notamment via la SFR ScInBioS) et le Région Bretagne (Projet de Recherche collaboratif régional avec la société EVOSENS – Plouzané et PERFOS – Lannion). Différents projets de recherche en cours ou en démarrage seront poursuivis dans le cadre de cette thèse : Collaborations nationales et internationales • quantification du collagène fibrillaire dans le diagnostic de la fibrose hépatique avec des équipes du CHU de Rennes et du Nottingham Digestive Diseases Centre (GB). Projet transversal soutenu par la Région Bretagne dans le cadre d’un programme de recherche collaboratif régional avec comme partenaires EVOSENS (Plouzané) et PERFOS (Plateforme d’Etudes et de Recherche sur les Fibres Optiques Spéciales – Lannion) et une équipe de biologistes (G. Baffet - INSERM) et de physiciens (C. Odin- IPR) de l’Université de Rennes 1. 4 • microstructure de la peau avec le Laboratoire de Neurobiologie cutanée (équipe du Professeur L. Projet transversal soutenu par l’appel à projet SFR ScInBioS en 2012. • imagerie des glandes salivaires dans le cadre du syndrome de Sjögren avec le Laboratoire d’Immunologie et Pathologie (équipe du Professeur J.-O. Pers - EA 2216-UBO). Projet transversal. • caractérisation de nouvelles sondes moléculaires membranaires pour la microscopie non-linéaire, avec le Laboratoire de Chimie (équipe du Professeur P.-A. Jaffrès - CEMCA – CNRS UMR 6521 - UBO. Projet exploratoire SOSIE soutenu par le BQR UBO 2012. Retombées Ce travail de recherche répondra à un besoin de la communauté médicale en termes d’imagerie in situ, c’est-à-dire d’imagerie non invasive en profondeur à l’échelle cellulaire tirant partie de différents contrastes sans nécessiter de marquage des tissus (réponse polarimétrique complète, fluorescence endogène et génération de second harmonique). Les champs d’application du dispositif imageur qui sera développé au cours de cette thèse couvriront potentiellement l’ensemble des domaines biomédicaux suivants: - le diagnostic précancéreux (essentiellement au niveau des épithéliums des organes creux), - les pathologies associées à la fibrose (foie, rein, poumon…), - la dermatologie et la cosmétologie (peau), - les pathologies du muscle à l’échelle du complexe actine-myosine (muscle septique et immobilisé, myopathies), - l’arthroscopie et la traumatologie (cartilages, tendons et ligaments riches en collagène), - l’odontologie et la stomatologie (carie dentaire, pathologies des glandes salivaires), - la neurologie et l’angiogenèse (sur modèle animal), - l’agroalimentaire (viande, poisson) et l’environnement (végétaux terrestres et marins). La formation apportée au futur doctorant lui offrira des perspectives professionnelles attractives, comme en témoigne les postes qu’occupent actuellement les anciens doctorants du directeur de thèse: • A. Thèse intitulée: "Microscopie multiphotonique de protéines fibrillaires : application à l'étude de la fibrose hépatique " soutenue en déc. 2010 à l’Université de Rennes 1 (ARED 100%). 5 Title Context Objectives Novelty of the project International collaboration Biomedical applications in linear and non-linear optical imaging Biomedical optics and biophotonics have known an exceptional growth for over ten years. For example the market of biophotonics in the world is bigger than that of optical telecommunications. In this both dynamic and competitive context, the laboratory SOL develops innovative optical imaging methods for biomedical applications in partnership with companies (EVOSENS society in Plouzané and PERFOS in Lannion for the development of a endo-biphoton microscope). The LSOL is particularly well-known in the area of linear polarimetry (called Mueller polarimetry) and nonlinear optics (through second harmonic generation microscopy). The laboratory has recently developed original approaches in order to make the most of these techniques in terms of speed and robustness: M. Dubreuil et al, Optics Express, 15, 13660 (2007); C. Odin et al, Optics Express 16, 16151 (2008). The aim of the thesis is the development and demonstration of coupling linear and non-linear optical images for biomedical diagnosis ex situ (biopsies) and so as to obtain in the medium term an endo-microscopic imaging in situ (via fiber optics). The first stage of the work is to develop a prototype snapshot Mueller polarimeter based on the technology of spectral optical coherence tomography (spectral OCT), based on a spectrally modulated light source, an optical fiber and a fast Fourier analysis of the signal. A second step will be to implement the device in a multiphoton microscope developed by LSOL to merge linear and non-linear polarimetric imaging in a single instrument. The final stage of the thesis is devoted to the bimodal imaging of biological samples (biopsies, skeletal muscle, salivary glands,...). The project is launched at the right time, because all the key components (optics, electronics, signal processing) are commercially available recently. Moreover UBO includes within its Brest site all the skills in physical science and life science to create the synergy needed to complete this research. The LSOL has recently shown the interest of combining linear and non-linear optics for the diagnosis of liver fibrosis on biopsy (Dubreuil et al "Mueller matrix Polarimetry for Improved liver fibrosis diagnosis," Optics Letters 37, 1061-1063, 2012). The innovation in this thesis is the development of these approaches for both technical progress (in order to take into account speed and flexibility constraints necessary for biomedical imaging) and applied studies (in order to extend the studies to other biomedical problems for which LSOL initiated collaborations supported by the UBO, ScInBioS SFR and the Brittany Region). Various research programs will be started or carried on in the context of this thesis: • scoring of fibrillar collagen in the diagnosis of liver fibrosis with teams coming from Rennes University and Nottingham Digestive Diseases Centre (GB). This cross-cutting project is supported by the Region of Brittany and implicates several partners: EVOSENS (company in Plouzané) and PERFOS (Platform Studies and Research on Fiber Optics Special in Lannion, a team of biologists (G. Baffet - INSERM) and a team of physicists (C. Odin-IPR) from University of Rennes 1. • microstructure of the skin with a skin Neurobiology Lab (Professor L. Misery - EA 4685- UBO). This cross-cutting project was supported by a PPF (2008-2012). D. Sevrain, Y. Le Grand, V. Buhé, C. Jeanmaire, G. Pauly, J.-L. Carre, L. This transversal project is supported by the call for proposals in 2012 ScInBioS SFR. • imaging of the salivary glands in the Sjögren's syndrome with the Laboratory of Immunology and Pathology (EA 2216 Professor J.-O. Pers). • characterization of new molecular probes for non-linear microscopy, with the Chemistry CEMCA– CNRS UMR 6521 – UBO (Professor P.-A. Jaffrès) . The pathfinder project SOSIE is supported by the BQR UBO 2012. 6 Expectations This research will fill a need in the medical community in terms of in situ imaging, that is to say, deep non-invasive imaging at the cellular level taking advantage of different contrasts without requiring staining tissues (full polarimetric response, endogenous fluorescence and second harmonic generation). The scope of the imaging device that will be developed during this thesis covers potentially the following biomedical fields: - Precancerous diagnosis (mainly in the epithelia of hollow organs - Diseases associated with fibrosis (liver, kidney, lung ...) - Cosmetology and dermatology (skin), - Diseases of muscle throughout the actin-myosin complex (tank and immobilized muscle, myopathies), - Arthroscopy and trauma (cartilage, tendons and ligaments rich in collagen), - Dentistry and stomatology (dental caries, diseases of the salivary glands), - Neurology and angiogenesis (animal model) - The food (meat, fish) and the environment (terrestrial and marine plants). Training given to future doctoral candidate should offer attractive career prospects to him, as evidenced by the positions currently occupied by the doctors supervised by the director of the thesis: • A. Leray: Research Fellow at CNRS (section 11) at the Institute of Biology of Lille since 2010. Thesis entitled: "multiphoton microscopy applied to biology" and sustained in October 2005 at the University of Rennes 1 (100% CDE) • T. Guilbert: Engineer INSERM holds the microscopy platform of the Cochin Hospital in Paris since 2012.
