Il y a fait ses études et a obtenu en 1974 un doctorat d’Etat à l’Université d’Aix-Marseille III sous la direction du Professeur Marcel Bertrand. Sa carrière académique débute à Lyon en 1974 en qualité d’Assistant dans le laboratoire du Professeur Jacques Goré où il est rapidement promu Maître Assistant (1978). Après avoir passé deux ans comme post-doctorant boursier OTAN à l’université de Californie à Berkeley, sous la direction du Professeur K. Peter C. Vollhardt, il rentre à l’université de Lyon I comme Maître de Conférences en 1983. En 1985, il est nommé Professeur de Chimie Organique à l’université Pierre et Marie Curie (UPMC-Paris). De juillet à septembre 1986, il retourne à l’université de Californie Berkeley comme boursier Fulbright. Il est nommé membre junior de l’Institut Universitaire de France en 1991 puis membre senior en 2002 et renouvelé en 2007. Il a été, entre autres, directeur du Laboratoire de Chimie Organique (UMR 7611) pendant dix ans, directeur de l’Ecole Doctorale de Chimie Moléculaire de Paris Centre (ED 406), membre élu du CNU (32ème section), délégué de la CPU pour la chimie au CNRS et membre du conseil scientifique de l’ANR. Actuellement, il est directeur de l’Institut Parisien de Chimie Moléculaire (UMR 7201) – lequel rassemble les forces vives en chimie moléculaire de l’UPMC– et de la Fédération de Chimie Moléculaire de Paris-Centre, membre du conseil scientifique de ERA-Net Chemistry, président du comité de rédaction de l’EJOC (Wiley) et président du comité organisateur du prochain congrès de l’ESOC 17. Depuis 2007, il est délégué scientifique à l’Agence d’Évaluation de la Recherche et de l’Enseignement Supérieur (AERES). Il a structuré ses activités de recherche autour de diverses thématiques comprenant le développement de nouvelles approches efficaces et sélectives pour la synthèse de systèmes polycycliques complexes par des processus en cascade faisant intervenir des réactions catalysées par des métaux de transition (Co, Au, Pt, Pd) ou des processus radicalaires, et d’autre part la construction de nouveaux matériaux hybrides en vue d’applications en biochimie et en physique. D’autre part, il a su créer des liens avec le monde industriel à travers diverses collobarations impliquant des compagnies chimiques et pharmaceutiques. Il est l’auteur de 297 publications et chapitres de livres et de 9 brevets et il a dirigé ou codirigé 73 thèses. Depuis 1987, il a donné des conférences dans 217 manifestations nationales et internationales. Il a participé à l’organisation de 11 conférences nationales et internationales. De plus, il a été Professeur Invité de plusieurs universités en Europe (Suisse, Italie, Espagne, Belgique) et dans le reste du monde (Japon, Israël, Taïwan, Chine, Australie). Les choix scientifiques réalisés découlent en quasi-totalité d'un questionnement simple : si nombre d'avancées en chimie sont rendues possibles grâce à la conception d'outils moléculaires ciblés pour des applications (en biologie et médecine par exemple, mais aussi aux frontières de la physique), toute réalisation des concepts est-elle possible? En d'autres termes, comment trouver des méthodes permettant un accès viable à n'importe quel objet que l'esprit puisse imaginer? Pour notre équipe, c'est l'invention ou la mise au point de nouvelles réactions qui ouvre des accès à des structures nouvelles pouvant être ajoutées à la panoplie de celles possibles. Découvrir de nouvelles conditions réactionnelles a également un grand intérêt puisque cela permet soit de clore des problèmes rencontrés pour des substrats spécifiques (extension de la gamme d'accès à des structures connues), soit de préparer des objets fonctionnels de manière acceptable pour la communauté, tout ceci en associant efficacité et respect de l’environnement. C'est ainsi que les efforts ont porté sur l'élaboration de processus économes en étapes comme les processus en cascade ou domino, lors desquels plusieurs processus élémentaires ont lieu consécutivement lors d'une seule manipulation. De cette manière, des molécules cycliques dont la grande complexité moléculaire est installée en une seule étape à partir de substrats commerciaux ou simples peuvent être obtenues. Elles incluent des réactions auparavant considérées comme interdites, si possible en conditions catalytiques. Dans ces deux cas, le chimiste de synthèse dispose de toute la classification périodique pour moduler les réactivités à souhait : le tableau périodique est notre abécédaire. La myriade de structures accessibles au chimiste moléculaire a naturellement débouché ces dernières années vers un intérêt plus marqué pour les interfaces. Mais la philosophie reste la même : la compréhension et la maîtrise de la grammaire moléculaire permet la conception et la synthèse d'objets moléculaires ciblés ainsi que des méthodes permettant d'y avoir accès. Cela explique que ces projets interfaciaux se fassent en collaboration avec d'autres groupes. 7 • Radicaux et hétéroéléments : Les radicaux libres, espèces à nombre impair d'électrons, ont longtemps été considérés comme trop réactifs pour être exploités. Une génération de pionniers, en particulier Marc Julia, s'est attelée à montrer qu'on pouvait les domestiquer et en ont déterminé les grandes lignes de réactivité. Ces intermédiaires étaient alors mûrs pour une utilisation en synthèse. Ainsi, ils ont été impliqués dans des processus en cascade de plus en plus sophistiqués. De ce point de vue, en diminuant le nombre d'étapes de synthèse, les cascades participent également d'une chimie plus verte ; d’ailleurs à ce jour, notre laboratoire détient le record mondial d'étapes élémentaires en une seule transformation. Des processus auparavant considérés comme interdits — 5-endo-trig et 4-exo-dig radicalaires — ont été mis en évidence. O N N I N hν pyridine, air N N N via : N Luotonine A 8 O O N N O O N N I N N N N3 N R O O N H N N R R = H, Me, i-Pr, CF3 Toujours dans le cadre du développement d’une chimie soucieuse de l’environnement mais aussi dans un souci d’ouvrir cette discipline à la synthèse pharmaceutique, notre laboratoire a tenté de trouver un palliatif à l’utilisation systématique d’hydrures d’organo-étain, des médiateurs radicalaires neurotoxiques. En collaboration avec le laboratoire du Pr. Dennis Curran (Université de Pittsburgh, USA), une nouvelle catégorie de donneurs d’atome hydrogène, moins toxiques et capables d’engendrer des espèces radicalaires, dans laquelle des carbènes hétérocycliques azotés (NHCs) activent la liaison B−H du borane BH3 ont été développés et récemment utilisés en polymérisation radicalaire. H H2B N N Ph BH2 R N N N Ph + R H N L'étape ultime du contrôle concernait la stéréosélectivité de réactions radicalaires : cela a été réalisé en utilisant des hétéroatomes comme le soufre pour moduler à souhait les réactivités. Ces derniers sont donc des outils puissants pour le chimiste de synthèse, ce d'autant plus que beaucoup des processus peuvent être rendus catalytiques en métaux, ce qui assure une viabilité économique et écologique importante en minimisant les déchets produits. L'activité de notre équipe s'est portée sur l'étude de transformations synthétiques catalysées par les complexes du cobalt (cyclotrimérisation [2+2+2] — dont la première occurrence remonte à Berthelot ! —, première catalyse organométallique de la Conia-ène réaction), puis, plus récemment, par ceux du platine et de l'or (cycloisomérisation de systèmes carbonés polyinsaturés, modulation par des fonctionnalités installées à des endroits précis). Une étude mixte structure/réactivité/modélisation a permis d'unifier les différents résultats expérimentaux en chimie de l'or et de contribuer à trancher un débat ancien concernant le mécanisme de la cyclotrimérisation catalysée par le cobalt. Dans le cadre d’une collaboration avec l’équipe du Dr. Claude Cochet, biologiste au CEA de Grenoble, il a été montré que certaines enzymes comme les kinases pouvaient être inhibées par des polyoxométallates suivant un mode d’action inédit. Ce résultat permet d’espérer le développement d’une nouvelle génération de médicaments destinés à la lutte contre le cancer. Pour un rapide survol…..
