Based upon theoretical and experimental data, the project is to test the hypothesis that commensal amoebias may help to restore this diversity and to improve the dysbiosisassociated disorders. It will be based on several animal models and the analysis of the gut microbiota. Despite several differences between diseases, a loss of diversity is a common feature of the dysbioses. We postulate that this loss of diversity may be related to a loss of bacterial predators (Mosca et al. Frontiers in Microbiology in press). Recent data reinforce this hypothesis showing that exposure to commensal amebias is associated with a dysbiosis. The aim of the project is to provide a proof of concept using animal models. C57B6 wild-type mice and mice characterized by a loss of bacterial diversity will be exposed to commensal amoebias and their microbiota composition will be analysed by sequencing the 16S mRNA. Conversely, the impact of amoeobicides treatment after exposure to amoebiases will also be tested. In parallel, a clinical impact of the exposure to amoebiases will be looked for on key animal models of inflammatory bowel diseases and metabolic disorders which are known to be associated with a dysbiosis and a loss of bacterial diversity.
