Here we will take advantage of reporter and deleter mice generated in the lab to determine the function of specific stromal subsets in the inflammatory and fibrotic process, identify the molecular crosstalk with immune cells and determine their net impact on immune responses and disease pathogenesis (lung/skin diseases). As fibrosis often correlates with high inflammation, broad range anti-inflammatory drugs are used for the treatment of these diseases. However these drugs have limited success, serious side effects and most importantly they do not restore the appropriate structure of the organ, a prerequisite for function. The underlying biological process regulating fibrosis remains poorly understood. Our lab has previously shown that stromal mesenchymal cells wrapped around vessels, resembling pericytes, are progenitors to a majority of pro-inflammatory collagen overproducing cells accumulating in the scar tissue after acute injury (Dulauroy et al., Nature Medicine 2012). Now we aim at understanding mechanisms of activation and regulation of such cells in chronic inflammatory diseases with a fibrotic component (such as pulmonary fibrosis and systemic sclerosis, an autoimmune disease leading to fibrosis of the skin and internal organs), decipher the molecular crosstalk with specific subsets of immune cells, and determine their net impact on immune responses and disease pathogenesis.
