Dans quel contexte s’insère le sujet de stage (démarrage d’un projet, travail partiel d’un sujet de thèse …….) : Démarrage d’un projet Agropolis-Cariplo (MIC-CEREs) portant sur la caractérisation des organismes phytobénéfiques du blé, dont une partie concerne l’isolement et la caractérisation des propriétés phytobénéfiques des bactéries racinaires du blé. Description du stage : donner un résumé (contexte, problématique, matériels et méthodes). Context: The plant model chosen for this project is wheat (Triticum aestivum L. subsp. aestivum), one of the most important sources for food, animal feed, and industrial raw materials. Wheat associated microbial diversity is considered important for maintaining for the sustain ability of agriculture production systems. Epiphytic, endophytic and rhizospheric bacteria are associated with wheat crops and have been shown to promote plant growth. Plant growth promoting (PGP) bacteria are used as biological control agents for the suppression of soil borne pathogens, or may be use as bioinoculants so that it fits into a long-term sustainable agricultural system. Objectives: The principal aim of this project is to i) study the impact of soil origin and plant genotypes on the diversity of wheat endophytic bacteria, to better understand what shapes the endophytic communities of wheat, and ii) to isolate root-associated microbes and evaluate their Plant Growth-Promoting (PGP) properties on tender wheat Methods: - Plant Experiments: In vitro culture of wheat, plant inoculation with bacteria - Technical microbiology: culturable approach using specific media to isolate PGP bacteria, various phenotypic tests to evaluate Plant growth promoting potential of bacteria - Molecular biology: DNA/RNA extractions, quantification, PCR, electrophoresis, cloning, gfp tagging of bacteria - Bioinformatic: Sequence analysis, Phylogeny, statistics. Desired profile: The candidate will be student in master. He/She studied microbiology in plant-microbe interactions or microbial ecology; and will have knowledge of bacteriology, molecular biology, and analysis of diversity of microbe communities. This training requires a great motivation for laboratory work. Bioinformatics skills will be appreciated.
