Dieser Krieg spielt sich meist hinter den Kulissen ab – bis die Körperabwehr einmal unterliegt und eine Infektion ausbricht. Aber auch ein übereifriges Immunsystem kann gefährlich werden: Dann lösen harmlose Substanzen eine allergische Reaktion aus oder körpereigene Strukturen werden durch Autoimmunerkrankungen wie Multiple Sklerose, Lupus Erythematodus oder Rheumathoide Arthritis attackiert. Marc Schmidt-Supprian untersucht mit seiner Forschungsgruppe „Molekulare Immunologie und Signaltransduktion“, wie die Entwicklung von Immunzellen über zelluläre Signalwege gesteuert wird. Oder auf fatale Weise fehlschlägt: Denn bei Fehlern in der Übertragung kann Krebs entstehen. Die Forscher manipulieren gezielt die Aktivität bestimmter Gene in der Maus, um deren Funktion im Immunsystem zu entschlüsseln. Drei Zelltypen stehen dabei im Vordergrund: „Natürliche Killer-T-Zellen“, kurz NKT-Zellen, erkennen Fette und gehören zur ersten Vorhut der Immunabwehr. Together with his research group “Molecular Immunology and Signal Transduction” Marc Schmidt-Supprian is studying how intracellular signaling guides the development of immune cells. Or how it goes awry with fatal consequences: Cancer can develop when there are errors in the signal transduction. Schmidt-Supprian and his research team are specifically manipulating the activity of certain genes in the mouse in order to elucidate their function in the immune system. Three cell types are in the foreground: natural killer T cells (NKT) recognize lipids and belong to the first vanguard of the immune defense. Mast cells, by contrast, are activated by allergens and then release – just like the NKT cells – messenger substances in great quantities. Das besondere Augenmerk SchmidtSupprians gilt jedoch seit Jahren den Antikörperproduzierenden B-Zellen. Jedes der kleinen Antikörper-Moleküle erkennt und neutralisiert hochspezifisch eine einzige aus Milliarden verschiedener Strukturen. Die Produktion von Antikörpern ist die Stärke der B-Zellen und zugleich ihre Schwäche, denn die Synthese der hochvariablen Proteine beruht auf molekularen Veränderungen des DNA-Moleküls. Werden dabei neben der erwünschten Variabilität in den Antikörper-Genfragmenten auch zelluläre Gene verändert, resultieren daraus gefährliche Defekte, die zur Entstehung von Lymphomen (Krebs des lymphatischen Systems) beitragen können. Fraglich ist noch, auf welche Weise die pathologischen Genveränderungen Krebs verursachen können. Im Fadenkreuz der Forscher stehen hier Gene der zellulären Signalübertragung. Denn für viele Lymphomarten ist die dauerhafte Aktivierung des sogenannten NF-ĸB-Signalwegs charakteristisch, der die Aktivität von Hunderten von Genen beeinflusst. Schmidt-Supprian konnte zeigen, dass die Lebensdauer der B-Zellen dramatisch ansteigt, wenn dieser Signalweg etwa wegen einer chronischen Entzündung oder eines Gendefekts dauerhaft aktiv ist. The production of antibodies is a strength of the B cells and at the same time their weakness because the synthesis of the highly variable proteins is based on molecular alterations of the DNA molecule. If, in addition to introducing the desired variability in the antibody gene fragments, also cellular genes are altered, dangerous defects are the result that can contribute to the development of lymphomas (cancer of the lymphatic system). It is still unclear just how these pathological gene mutations can cause cancer. Here the researchers are setting their sights on genes responsible for cellular signal transduction. Characteristic for many kinds of lymphomas is the permanent activation of the NF-ĸB signaling pathway, which influences the activity of hundreds of genes. Schmidt-Supprian was able to show that the life span of B cells increases dramatically when this signaling pathway is permanently active due to for example a chronic inflammation or a gene defect. If the immune cells then begin to divide, cancer can develop. In the animal model Schmidt-Supprian and his team want to elucidate the underlying cellular and molecular mechanisms of these processes.
